BACKGROUND: Spontaneous hyperadrenocorticism (HAC) is rare in cats. Clinical findings, diagnostic test results, and response to various treatment options must be better characterized.
OBJECTIVES: To report the clinical presentation, clinicopathologic findings, diagnostic imaging results, and response to treatment of cats with HAC.

Trilostane is a competitive inhibitor of the 3 beta-hydroxysteroid dehydrogenase enzyme system localized in the adrenal cortex and in the gonads. This inhibitor reduces the production of cortisol, aldosterone and androstendione. Castrate-resistant prostate cancer (CRPC) progression is driven by intracrine androgen synthesis despite low circulating androgens. This study investigates the effects of active vitamin D (1,25-(OH)₂-D₃) on intracrine steroidogenesis in LNCaP xenograft tumors. This study aimed to retrospectively describe the clinical progression following diagnosis of iatrogenic hypocortisolemia (iHC) in 48 dogs receiving trilostane for pituitary-dependent hyperadrenocorticism. Cortisol concentrations were ≥1.5 mg/dL within 6 mo following diagnosis of iHC in 76.3% of dogs (95% confidence interval [CI] 59.8-88.6%). The efficacy of trilostane in the treatment of canine pituitary-dependent hyperadrenocorticism (PDH) was evaluated in 78 dogs with the condition which were treated for up to three years. The metabolism of DHT in the cytosol of the quail uropygial gland was found to be so high that the steroid was almost completely inactivated within 2 hours of incubation at 0 C. In these conditions, DHT cannot be used for the characterization of androgen receptors. By contrast, R 1881 and mibolerone, which are not metabolized, can be used as alternative ligands. Moreover, the extremely high metabolism of DHT questions the physiologic role of this steroid in the quail uropygial gland.

The etiology and pathogenesis of nonbacterial prostatitis remains unclear. This study provides evidence that genetic background, advancing age, and hormonal imbalance are important etiologic factors for prostatitis in rats. Three patients with idiopathic hyperaldosteronism were continuously treated with trilostane, a competitive inhibitor of adrenal 3 beta-hydroxysteroid dehydrogenase (3 beta-HSDH) (3 to 4 2/3 years).
