Effects of gas composition and pH on kinetics of lung angiotensin-converting enzyme.

Given the pH dependence of enzymes in general and the potential importance of a blood and alveolar gas composition dependency on the interpretation of changes in the hydrolysis of angiotensin-converting enzyme (ACE) substrates by pulmonary endothelial ACE, we examined the influence of Pco2 and Po2 on the hydrolysis of a synthetic ACE substrate (benzoyl-phenylalanyl-alanyl-proline, BPAP) on passage through isolated rabbit lungs. Perfusate pH values of about 7.1, 7.4, and 7.9 were obtained by ventilating the lungs with gas containing different CO2 concentrations and Po2 values of approximately 1

[Induction of blood plasma carboxycathepsin (angiotensin I-converting enzyme) in normo- and hypertensive rats in response to a single administration of captopril].

The experiments were carried out to elucidate the effect of carboxycathepsin (CC) activity inhibition by a specific inhibitor--captopril--on plasma enzyme concentration in normotensive rats and rats with renovascular hypertension. A single oral administration of captopril (10 mg/kg body weight) produced an increase in CC concentration in both hypertensive and sodium-depleted normotensive rats with a parallel decrease in arterial pressure, but had no effect on sodium-repleted normotensive rats. It is suggested that the increase in plasma CC concentration is a compensatory response to the inhibi

Imipenem: first of a new class of beta-lactam antibiotics.

Imipenem, the first of a new class of carbapenem antibiotics, has potent activity against most clinically important species of bacteria, including isolates resistant to other antibiotics. The drug is well distributed to most tissues and fluids after intravenous administration; however, levels in cerebrospinal fluid are modest. Most of the drug is eliminated in the urine, where it is metabolized by an enzyme on the brush border of the renal tubular cells; cilastatin is given simultaneously to inhibit this inactivation. Adverse effects include a syndrome of nausea and hypotension, especially aft

Clinical prediction of the adult respiratory distress syndrome.

The use of clinical, physiologic, and laboratory parameters in the prediction or early detection of ARDS has been reviewed. From both a clinical and research standpoint, the ability to identify patients at risk is extremely important. The selection of patients according to predisposing clinical events has been the most successful thus far. The use of physiologic variables and gauges of injury severity have been of limited value, particularly for assessing ARDS risk in the individual patient. Only a handful of the proposed mediators or markers of acute lung injury have been studied prospectivel

Serum angiotensin converting enzyme in sarcoidosis: sensitivity and specificity in diagnosis: correlations with disease activity, duration, extra-thoracic involvement, radiographic type and therapy.

Serum angiotensin converting enzyme (SACE), despite certain limitations, has been found to be a useful adjunct to the diagnosis, assessment of disease activity, and management of sarcoidosis. The spectrofluorimetric assay was used to measure SACE in 50 normal controls, 76 patients with tuberculosis (42 pulmonary, 16 lymphatic and 18 military cases), 20 patients with cryptogenic fibrosing alveolitis, 50 patients with silicosis, three patients with extrinsic allergic alveolitis, 10 patients with Crohn's disease, two patients with Gaucher's disease, and 128 patients with sarcoidosis on 303 occasi

67Ga scintigraphy, serum lysozyme and angiotensin-converting enzyme in pulmonary sarcoidosis.

67Ga scintigraphy, using a 67Ga accumulation score, was compared with serum lysozyme (LZM) and angiotensin-converting enzyme (ACE) levels in 34 patients with biopsy-proven, pulmonary sarcoidosis. Serum LZM and ACE values varied between 1.0 to 21.3 mg/l (mean 4.3 +/- 3.4 mg/l) and 33 to 146 U/l (mean 69 +/- 26 U/l), respectively. Normal values were found in 26 and 35% of the patients. All patients, however, had abnormal 67Ga uptake in the pulmonary hilar lymph nodes and/or parenchyma. The follow-up of untreated and treated patients supports the suggestion that 67Ga scintigraphy is more useful f

The level of blood kinins, their creation and inactivation after one exposure to intense ultraviolet radiation in rabbits.

Experimental rabbits were exposed to ultraviolet radiation once for 45 minutes, and blood samples were obtained from the carotid artery in these animals 45 min and 3, 6 and 24 hours after the end of this exposure. In the group of control rabbits blood samples were obtained in the same way without previous exposure to radiation. The hairs on the back were cut closely at the skin and this skin area was exposed to ultraviolet rays from a Hanau Q 400 burner at 405--289 nm wavelengths and at an intensity of 134 000 erg/sec/cm2, using an UG 2 T Schott filtre and an absorber of long-wave radiation. Muscle breakdown and lysosomal activation (biochemistry).

Muscle tissue levels of lysosomal catheptic enzymes, such as cathepsins D, A, B1, C, and dipeptidyl peptidase II, were measured in control subjects and patients with muscular dystrophies, polymyositis, and certain denervating diseases. The results show that, in general, the activities of these enzymes are increased in muscles of patients with muscular dystrophies and other diseases. The increases in cathepsin D and autolytic activities are not significant until the late stage of the disease process. Cathepsins A, B1, and C are, however, significantly elevated in mildly affected dystrophic and

Serum angiotensin-converting enzyme in leprosy and coccidioidomycosis.

Serum angiotensin-converting enzyme levels were found to be elevated in 71.4% of 42 leprosy patients, both treated and untreated, but in only one of 13 patients with disseminated coccidioidomycosis. The elevations with leprosy were present in association with each of the three major categories: lepromatous, borderline, or tuberculoid. Sulfone therapy had no immediate effect on the elevated serum levels, although long-term sulfone therapy appeared to result in lowering of the level. Corticosteroid therapy had a more immediate and dramatic effect on reducing the elevated angiotensin-converting e

Effects of 12-week aquatic HIIT on blood pressure lipid profile and BaPWV in postmenopausal women with different ACE genotypes.

The angiotensin-converting enzyme (ACE) insertion/deletion (I/D) polymorphism influences renin-angiotensin-aldosterone system activity and may modulate cardiovascular adaptations to exercise. Yet, evidence regarding genotype-dependent responses to aquatic high-intensity interval training (HIIT) in postmenopausal women is limited. We aimed to compare the effects of 12-week aquatic HIIT on blood pressure, lipid profile, and arterial stiffness between postmenopausal women with the ACE II genotype and those carrying at least one D allele (ID/DD genotype). Sixty postmenopausal women aged 45-75 year

Genetic Association of the Ins/Del Variant of ACE and Risk of Cardiomyopathy: A Case-Control Study and Updated Meta-Analysis.

INTRODUCTION: Cardiomyopathy is a complex condition influenced by multiple genes and environmental factors. It has been suspected that cardiomyopathy is affected by the ACE gene's I/D polymorphism. Our study aimed to evaluate the association between this polymorphism and cardiomyopathy risk in the Jammu population of North India, alongside a meta-analysis to determine the specific risks associated with different types of cardiomyopathy. METHOD: In the case-control study, we opted for a convenient sampling technique to gather patients from hospitals. Meanwhile, for the meta-analysis registered

Screening and molecular dynamics simulation of ACE inhibitory tripeptides derived from milk fermented with Lactobacillus delbrueckii QS306.

Peptides in milk fermented with Lactobacillus delbrueckii QS306 before and after ultrahigh pressure treatment were identified using proteomics. Subsequently, 16 stable tripeptides were screened out based on activity score prediction, PeptideCutter analysis, and hydrophobicity calculations. Among them, WRP, WSR, and YRP showed the best angiotensin-converting enzyme (ACE) inhibitory activity, and their semi-inhibitory concentrations were 46.707, 300.121, and 89.555 μM, respectively. WRP and WSR were competitive inhibitors, whereas YRP was non-competitive. Gastrointestinal simulation revealed tha

The potential role of renin-angiotensin system in mild traumatic brain injury.

Traumatic brain injury (TBI) is a serious public health problem, affecting 69 million people worldwide annually. Mild TBI (mTBI) comprises the majority of the cases and remains the most neglected TBI severity. Its intricate pathophysiology involves complex cellular and molecular processes that remain uncomprehended. Although the renin-angiotensin system (RAS) has its well-known roles in blood pressure regulation and fluid balance, accumulating evidence demonstrates its active expression and signaling in the central nervous system. Over the past years, pre-clinical studies have been supporting

Dysregulated ACE/Ang II/Ang1-7 signaling provokes cardiovascular and inflammatory sequelae of endotoxemia in weaning preeclamptic rats.

Weaning preeclamptic (PE) rats exhibit exaggerated endotoxic signs of hypotension and cardiac autonomic neuropathy. Considering the role of renin-angiotensin system (RAS) in maternal programming during PE, we investigated the hypothesis that gestational modulation of offensive (Angiotensin II, Ang II) and defensive (Ang 1-7) components of RAS alleviates cardiovascular hyperresponsiveness of weaning PE mothers to postpartum endotoxemia. PE was induced by treating pregnant rats with the nitric oxide synthase inhibitor L-NAME (50 mg/kg/day) for 7 consecutive days starting from gestational day 14.
