Changes in the immunospecificity of the nuclear antigens were demonstrated in the colon chromatin of rats treated with 1,2-dimethylhydrazine. Tumor-specific nuclear antigen appeared in the early stages of chemically induced colon carcinogenesis. Sodium barbiturate, in conjunction with the carcinogen, induced a higher level of nuclear antigen activity than that obtained with carcinogen alone. The rise of immunoactivity in carcinogen-treated colon chromatin can be abolished by simultaneous treatment with disulfiram or butylated hydroxytoluene.

In this review, authors considered characteristics of depression and alcohol addiction. Depressive disorders comorbid to alcoholism are heterogeneous in their nature and structure and can be observed during alcohol withdrawal syndrome and in remission. Antidepressants, and first of all selective serotonin reuptake inhibitors (SSRIs), along with the specific anti-relapse drugs (disulfiram, acamprosate and naltrexone), are the drugs of choice in treatment of alcohol addiction. Efficient combinations of pharmacotherapy and psychotherapy can improve outcomes of both comorbid disorders.

Quinazoline oxide--an intermediate in chlordiazepoxide synthesis--is the most potent contact allergen in the pharmaceutical industry. Penicillins proved to be potent allergens. Conditions of the technological process favor development of hypersensitivity to tetracyclines. No single case of allergy to erythromycin was noted. In case of employees hypersensitive to disulfiram and aminophylline cross reactions with compounds of similar structure were observed. The authors discuss also some problems concerning contact allergy in all persons occupationally dealing with various medicines.

Nickel is the most common cause of allergic contact dermatitis in females. The dermatitis can be maintained both by direct contact and by ingestion of nickel. In 9 out of 17 patients suffering from dermatitis, a diet with a low nickel content has improved their condition. Eleven patients with chronic nickel hand dermatitis were given a daily dosage of 200 to 400 mg of tetraethylthiuramdisulfide (Antabuse). Burkholderia pseudomallei infection causes melioidosis, which is often fatal if untreated. There is a need to develop new and more effective treatments for melioidosis. This study reports apo and cofactor-bound crystal structures of the potential drug target betaine aldehyde dehydrogenase (BADH) from B. pseudomallei. A structural comparison identified similarities to BADH from Pseudomonas aeruginosa which is inhibited by the drug disulfiram. This preliminary analysis could facilitate drug-repurposing studies for B. pseudomallei.

1H NMR methods are described which allow direct studies of the Cys34 binding site of albumin in intact human blood plasma in vitro. Antiarthritic gold drugs and the alcohol-aversive drug disulfiram induce a structural transition detectable via H epsilon 1 and H delta 2 resonances of His3 of albumin, and reactions of cystine, glutathione and captopril in plasma have also been investigated. Contrary to most assumptions, little of the albumin in normal plasma appears to be blocked at Cys34 as a cystine disulfide.
