We report on some aspects of progression of chronic disease and its association with acute manifestations with special reference to grades of lymphoedema in bancroftian filariasis. These analyses were based on the clinical history and clinical findings of 1300 individuals at the time of their first visit to the filariasis clinic at a centre in south India. The mean number of adenolymphangitis (ADL) attacks in one year was 4.9 +/- 1.7, 5.5 +/- 0.9 and 10.4 +/- 3.2 in patients with grade I, grade II and grade III lymphoedema respectively. The mean duration of oedema was 47.4 +/- 5.9 days, 6.2 +/

BACKGROUND: Ivermectin, a modified avermectin, is widely known to be an ectoparasiticidal agent in animals but its effect on human ectoparasites is not known.
METHODS: As a part of a chemotherapy trial with ivermectin against Wuchereria bancrofti microfilaraemia, two males with clinical scabies were studied. Three skin scrapings were taken from the lesions of each of the cases and examined with potassium hydroxide solution for the presence of eggs, larvae or adult mites.
RESULTS: Following a single oral dose of ivermectin at either 100 micrograms or 20 micrograms/kg body weight both the itch

One hundred Brugia pahangi infective larvae (L3) caused microfilaremic (mf + ve) infection in 56% of inbred PVG rats. Adult worms were recovered consistently from infected rats but worm recovery was very low, only 1-3% of L3 inoculated survived to adulthood and the worms were dispersed in a wide range of anatomical sites. This suggested that lack of microfilaremia may be due to the low probability of male and female worms meeting in the same site and thus may be numerically and topographically based. When the number of infective larvae inoculated was increased to 500, the percentage of mf + ve

Calotropis procera (Aiton) W.T. Aiton, a perennial shrub of the Apocynaceae family, is widely valued in Ayurveda, Unani, Siddha, and homeopathy for its broad therapeutic potential, attributed to a diverse phytochemical repertoire that includes alkaloids, triterpenoids, flavonoids, saponins, steroids, fatty acids, glycosides, and proteases. Traditionally, it is used to treat bronchitis, asthma, leprosy, eczema, elephantiasis, joint disorders, various skin ailments, and is also employed as a laxative, anthelmintic, and antiulcer agent. Phytochemical investigations of its latex, leaves, flowers, …

Filariases are diseases caused by infection with filarial nematodes and transmitted by insect vectors. The filarial roundworm Dirofilaria immitis causes heartworm disease in dogs and other carnivores. D. immitis is closely related to Onchocerca volvulus, Wuchereria bancrofti and Brugia malayi, which cause onchocerciasis (river blindness) and lymphatic filariasis (elephantiasis) in humans and are neglected tropical diseases. Serum N-glycosylation is very sensitive to both pathological infections and changes in mammalian biology due to normal aging or lifestyle choices. Here, we report significa…

Lymphatic filariasis, commonly called elephantiasis, poses a burden of estimated level of 5.09 million disability adjusted life year. Limitations of its sole drug, diethylcarbamazine (DEC) drive exploration of effective filarial target. A few plant extracts having polyphenolic ingredients and some synthetic compounds possess potential dihydrofolate reductase (DHFR) inhibitory effect. Here, we postulated a plausible link between folates and polyphenolics based on their common precursor in shikimate metabolism. Considering its implication in structural resemblance based antagonism, we have attem…

A pilot study was conducted to determine if host genetic factors influence susceptibility and outcomes in human filariasis. Using the candidate gene approach, a well-characterized population in South India was studied using common polymorphisms in six genes (CHIT1, MPO, NRAMP, CYBA, NCF2, and MBL2). A total of 216 individuals from South India were genotyped; 67 normal (N), 63 asymptomatic microfilaria positive (MF+), 50 with chronic lymphatic dysfunction/elephantiasis (CP), and 36 tropical pulmonary eosinophilia (TPE). An association was observed between the HH variant CHIT1 genotype, which co…

Brugia malayi is a human filarial nematode parasite that causes lymphatic filariasis or 'elephantiasis' a disfiguring neglected tropical disease. This parasite is a more tractable nematode parasite for the experimental study of anthelmintic drugs and has been studied with patch-clamp and RNAi techniques. Unlike in C. elegans however, calcium signaling in B. malayi or other nematode parasites has not been achieved, limiting the studies of the mode of action of anthelmintic drugs. We describe here the development of calcium imaging methods that allow us to characterize changes in cellular calciu…

Lymphatic filariasis, a mosquito-borne parasitic disease, can lead to lymphoedema and elephantiasis. This study describes the results of a baseline survey of a lymphoedema morbidity management programme in Togo. A convenience sample of 188 people with lymphoedema was asked about symptoms, treatment preferences and quality of life. Those with higher stage lymphoedema were more likely to have experienced an acute attack (odds ratio=1.9; P=0.002). Although only 28.2% of those surveyed reported currently using any lymphoedema treatment, 80.3% had used treatments in the past, primarily traditional …

We conducted a pilot study to test rapid assessment procedures for the community diagnosis of lymphatic filariasis in some rural communities … is in particular. The use of self-administered questionnaires provided data comparable with data on elephantiasis in the community from a case search. Examination of a random 30-40 adult males for hydroceles provided a good correlation with the community microfilaria prevalence, with a correlation coefficient of linear regression r = 0.79. These individual rapi …

Lymphatic filariasis affects predominantly the poorer sector of a community, who can least afford to have the disease. According to an estim … ter. In brugian filariasis, adenolymphangitis is most commonly observed at the inguinal region, and elephantiasis predominantly involves the leg below the knee. In bancroftian filariasis, the lymphatics of the male genitalia are frequently affected, leading to epididymo-orchitis and hydrocele. Lymphatic filariasis runs an accelerated clinical course in previous …

Onchocerciasis--infection by Onchocerca volvulus--has four cardinal manifestations: dermatitis, subcutaneous nodules, sclerosing lymphadenit … y fibrosis and obstruction of lymph flow, a condition causing adenolymphocele ("hanging groin") and elephantiasis of the genitalia. Some patients appear to have immune tolerance to degenerating microfilariae, perhaps as a result of exposure in utero to microfilarial antigens in the maternal circulation. In contrast, other patients (Yemenites, for example) have …

BACKGROUND AND AIM: Lymphatic filariasis, also known as elephantiasis, still remains a problem in Indonesia. The primary causative species of this disease are the filarial worms Wuchereria bancrofti and Brugia spp. This study was conducted to identify the diversity of species and behavior of mosquitoes and to determine the mosquitoes that could be potential vectors of filariasis.
MATERIALS AND METHODS: Mosquito samples derived from Hulu Sungai Utara (HSU) district in the 2017 multicenter study conducted in Indonesia were used in this cross-sectional study. The diversity of mosquito species wa…
