Gluten sensitivity and 'normal' histology: is the intestinal mucosa really normal?

BACKGROUND: Early pathogenetic events of gluten intolerance may be overlooked in patients with serologic markers of celiac disease and normal intestinal mucosa by both conventional histology and immunohistochemistry. AIMS: To investigate if a submicroscopical damage of the absorptive cell surface was associated with developing gluten sensitivity. PATIENTS AND METHODS: Duodenal biopsies of seven subjects with positive anti-endomysial antibodies and normal histology underwent ultrastructural evaluation of the epithelial surface by means of both scanning and transmission electron microscopy. Gastrointestinal function, divalent metal transporter-1 expression and intestinal iron absorption.

Iron absorption involves two carriers, one involved in the uptake of iron across the microvillus membrane of the enterocyte and the other in its transfer to the plasma at the basolateral surface. The uptake phase is thought to involve divalent metal transporter-1 (DMT1) which may move from the cytoplasm to the microvillus membrane under conditions of iron deficiency. To examine this possibility we used fasted animals previously fed an iron-deficient diet and then gavaged with iron. We measured the processes of iron absorption using in vivo gut sacs and correlated the changes observed with the

Autoradiographic localization of (3H)-thymidine incorporation in the small intestinal epithelium of fetal sheep.

Aspects of the cell kinetics of the developing intestinal epithelium of fetal sheep were investigated by autoradiography. The sites of (3H)- … , before rising to 0.25 (+/- 0.04) at 136 days. These results demonstrate that the proliferation of enterocytes in utero is qualitatively similar to that in the adult, being confined to a distinct progenitive zone. Such a finding is an important first step in understanding changes in enterocyte structure and function, since this may, as seen in the adult, be …

Constitutive Interferon Epsilon Expression Shapes Antiviral Epithelial States in the Female Reproductive Tract and Intestine.

Antiviral defenses at mucosal barriers are essential for preventing viral entry and systemic infection. Interferon epsilon (IFNε) is a uniqu … s. Extending these analyses to the intestine, we found that IFNε is highly expressed in villous-tip enterocytes of the small intestine in vivo , where it sustains inflammatory enterocyte subsets and maintains type III IFN expression. Loss of Ifnε depleted these subsets and rendered mice more susceptible to enteric viral infection. Together, these findings est …

Intestinal hepcidin overexpression promotes iron deficiency anemia and counteracts iron overload via DMT1 downregulation.

Hepcidin is the key hyposideremic hormone produced primarily by the liver. However, recent reports reveal extrahepatic functional sources of … testinal hepcidin as a regulator of systemic iron homeostasis, acting on DMT1 on the apical side of enterocytes, with potential therapeutic relevance for hematologic or iron disorders.

Infectious diseases affecting the small bowel - what not to miss.

PURPOSE OF REVIEW: This review summarizes infectious diseases involving the small bowel (SB) with a focus on recent literature related to diagnosis and pathophysiology. RECENT FINDINGS: Typical symptom for SB infections is diarrhea, mostly self-limiting. Pathogens include bacteria, viruses, fungi, protozoan parasites, and helminths. Host-pathogen interaction is of special interest in infections with potentially severe or prolonged course. Research uses increasingly enterocyte cell culture systems. SARS-CoV2 can also infect enterocytes via angiotensin converting enzyme 2 (ACE2) receptor and ca…

Abnormal regulation of intestinal calbindin (CaBP9k) and calmodulin in the spontaneously hypertensive rat.

This review focuses particularly on abnormalities of Ca-binding proteins in transporting epithelia, which have been observed in various models of experimental hypertension. The enterocyte content of integral membrane Ca(2+)-binding protein (IMCAL) and calbindin-9k and the renal tubular calbindin-28k content have been shown to be decreased in the spontaneously hypertensive rat (SHR), compared with normotensive control rats (WKY). Similarly, calmodulin content was decreased in several tissues including the intestine; however, calmodulin activity was increased. In recent studies, the authors exam

Single-cell transcriptomic landscape of the southern green stink bug (Nezara viridula) midgut.

BACKGROUND: The southern green stink bug (SGSB), Nezara viridula, is a globally distributed hemipteran pest that damages many economically important crops. Its midgut supports digestion, defense, symbiosis, and interactions with orally delivered control agents, yet the cellular composition of this tissue remains poorly characterized. We therefore developed a single-cell transcriptomic atlas of the N. viridula midgut. RESULTS: Single-cell RNA sequencing of two biological replicates yielded a quality-filtered data set of 13,763 cells. Unsupervised clustering identified 12 transcriptionally dist

Methionine deficiency and its hydroxy analogue influence chicken intestinal 3-dimensional organoid development.

Methionine and its hydroxy analogue (MHA) have been shown to benefit mouse intestinal regeneration. The intestinal organoid is a good model that directly reflects the impact of certain nutrients or chemicals on intestinal development. Here, we aimed to establish a chicken intestinal organoid culture method first and then use the model to explore the influence of methionine deficiency and MHA on intestinal organoid development. The results showed that 125-μm cell strainer exhibited the highest efficiency for chicken embryo crypt harvesting. We found that transforming growth factor-β inhibitor (

Fenofibrate reduces intestinal damage and improves intestinal recovery following intestinal ischemia-reperfusion injury in a rat.

PURPOSE: Fenofibrate (FEN) is known as a nuclear receptor activator which regulates many pathophysiological processes, such as oxidative stress, inflammation, and leukocyte endothelium interactions. Recent studies have demonstrated an anti-oxidant, anti-inflammatory, and anti-ischemic role of FEN in the attenuation of ischemia-reperfusion (IR) injury in the kidney, liver, brain, and heart. The purpose of the present study was to examine the effect of FEN on intestinal recovery and enterocyte turnover after intestinal IR injury in rats. METHODS: Male Sprague-Dawley rats were divided into four

Cell differentiation and proliferation in the gastrointestinal tract with respect to the local immune system.

Human intestinal lamina propria T cells have a low expression of the CD45RA antigen and a high expression of the CD45RO antigen. This phenotype is characteristic for memory T cells. In addition, T cells in the effector compartment of the mucosa bear surface antigens that are very rarely found in other sites of the immune system. Intestinal T cells also express functional IL-2 receptors, and IL-2 receptor alpha-chain mRNA, and are able to synthesize high amounts of IL-2. However, other markers of memory T cells, as CD29, are not expressed in high density in the lamina propria, indicating that l

Faecal free fatty acids in tropical sprue and their possible role in the production of diarrhoea by inhibition of ATPases.

Faecal excretion of fatty acids is increased in patients with tropical sprue because of unabsorbed dietary fatty acids. The excretion of fatty acids correlates well with faecal wet weight. In vitro unsaturated fatty acids inhibited Na K-ATPase and Mg-ATPase isolated from basolateral membranes of enterocytes and colonocytes. These findings are a possible explanation for the observed abnormalities in water and electrolyte absorption by the colon in patients with tropical sprue and steatorrhoea.

The influence of selected gastrointestinal parasites on apoptosis in intestinal epithelial cells.

Studies on the parasite-host interaction may provide valuable information concerning the modulation of molecular mechanisms as well as of the host immune system during infection. To date, it has been demonstrated that intestinal parasites may affect, among others, the processes of digestion in the gastrointestinal system of the host, thus limiting the elimination of the parasite, the immune response as well as inflammation. However, the most recent studies suggest that intestinal parasites may also affect modulation of the apoptosis pathway of the host. The present paper presents the latest sc

[Genetic hemochromatosis and the HFE gene: from molecular genetics to clinical diagnosis].

More than 90% of patients with genetic hemochromatosis carry a characteristic mutation in the HFE-gene (C282Y). HFE modulates the iron uptake by the transferrin receptor. Duodenal crypt cells of HFE-knockout mice show low intracellular iron concentrations which lead to an upregulation of the divalent metal transporter and enhanced iron uptake by duodenal enterocytes. Heterozygosity for the C282Y mutation appears to alter the course of other liver diseases like porphyria cutanea tarda and nonalcoholic steatohepatitis.

The life cycle of Cryptosporidium baileyi n. sp. (Apicomplexa, Cryptosporidiidae) infecting chickens.

The life cycle and morphology of a previously undescribed species of Cryptosporidium isolated from commercial broiler chickens is described. The prepatent period for Cryptosporidium baileyi n. sp. was three days post oral inoculation (PI) of oocysts, and the patent period was days 4-24 PI for chickens inoculated at two days of age and days 4-14 for chickens inoculated at one and six months of age. During the first three days PI, most developmental stages of C. baileyi were found in the microvillous region of enterocytes of the ileum and large intestine. By day 4 PI, most parasites occurred in …
