BACKGROUND: Metabolic dysfunction-associated steatotic liver disease (MASLD) is a prevalent chronic liver disorder driven by obesity and metabolic dysfunction. MASLD progresses to metabolic dysfunction-associated steatohepatitis, which is characterized by inflammation, hepatocyte injury, and fibrosis, increasing the risk of cirrhosis and liver failure. Recent studies suggest that neutrophil extracellular traps (NETs) and extracellular DNA (ecDNA) contribute to liver inflammation and fibrogenesis. However, their role in MASLD pathogenesis remains incompletely understood.
AIM: To investigate th…

NADPH oxidase 5 (NOX5) is a calcium-dependent enzyme that generates reactive oxygen species (ROS) and plays critical roles in immune responses, the functions of which have been shown to be regulated by the pro-inflammatory mediator leukotriene C4 (LTC4). In the present study, a novel NOX5 homology was identified from Ruditapes philippinarum (designated as RpNOX5). Phylogenetic analysis and conserved motif characterization supported RpNOX5 as a new member of the invertebrate NOX5 family. RpNOX5 contains typical NOX5 structural domains, including EF-hand motifs, ferric reductase domain, FNR-like

Leukotriene B4 (LTB4) is a pro-inflammatory lipid mediator that cooperates with CXC chemokine receptor type 4 (CXCR4) in regulating immune responses in vertebrates, but their function in bivalve innate immunity remains largely unknown. This study investigated the involvement of LTB4 and a CXCR4-like protein in extracellular traps (ET) formation in Ruditapes philippinarum. Metabolomics revealed that LTB4 was a key differential metabolite in gill tissues following LPS stimulation, and ELISA further confirmed that LTB4 levels were significantly elevated upon LPS challenge. A CXCR4 homolog designa…

The root extracellular trap (RET) is a network formed by root-associated, cap-derived cells (AC-DCs) and their mucilaginous secretions. The RET is located at the root tip where it establishes a protective interface against abiotic and biotic stresses. The RET mucilage is mainly composed of complex cell wall polysaccharides (including pectin and xyloglucan) and glycoproteins, that are entangled to each other within the network. Despite our knowledge of the composition of RET mucilage, the structural organization of the RET remains poorly understood. Here, we have investigated the role of pectin

Palisaded neutrophilic granulomatous dermatitis (PNGD) is an uncommon reactive granulomatous dermatosis frequently associated with rheumatoid arthritis (RA). Histopathologically, PNGD is characterized by neutrophilic infiltrates, degenerated collagen bundles, and surrounding palisading histiocytes; however, the contribution of neutrophils to lesion formation remains poorly understood. We report two patients with RA-associated PNGD who presented with chronic erythematous papules and nodules on the elbows. Histopathological examination revealed characteristic palisaded granulomatous inflammation

Microglia are key immune-competent cells responding immediately to manipulate post-stroke neuroinflammation to shape the prognosis of stroke … alleviate ischemic brain injury. Recombinant GPNMB (rGPNMB) administration could reduce neutrophil extracellular traps (NETs) formation, while knockout of GPNMB promoted NET formation in ischemic stroke. In addition, we found that CD44 functioned importantly in mediating the role of GPNMB inhibiting NET formation and alleviating ischemic brain injury. Depletion of n …

Pancreatic ductal adenocarcinoma (PDAC) is one of the most lethal malignancies, characterized by poor treatment response. A part of this dismal prognosis can be attributed to the immunosuppressive tumor microenvironment (TME) that renders tumors anti-immunogenic and resistant to immunotherapies. Neutrophil extracellular traps (NETs) are a frontline defense in pathogen clearance. However, NETs also play a destructive role in many chronic conditions, such as autoimmune disease and cancer. Recent evidence corroborates the impact of NETs on PDACs pathogenesis by promoting tumor progression and enh…

The interplay between neutrophils and macrophages is central to inflammatory regulation, yet how macrophage polarization governs the homeostasis of neutrophil extracellular traps (NETs) remains poorly understood. Here, we show that the macrophage polarizing environment is a critical determinant of NETs dynamics in human primary cell co-cultures. Using immunofluorescence, flow cytometry, kinetic extracellular DNA assays, and multiplex cytokine profiling, we demonstrate that IFN-γ/LPS (M1-polarized) conditions promote robust but transient NETs formation, accompanied by increased loss of neutroph…

BACKGROUND: Neutrophil extracellular traps (NETs) contribute to atherosclerosis progression and are linked to adverse clinical outcomes such as myocardial infarction and stroke. Although the triggers of NET formation in plaques are known, the mechanisms governing DNase-mediated NET clearance and how these are disrupted during atherosclerosis remain unclear. Moreover, the consequences of impaired NET clearance on disease progression are not known.
METHODS: Low-density lipoprotein receptor knockout (Ldlr-/-) mice with hematopoietic cell-specific deletion of DNase1 and DNase1L3 were fed a Wester…

Excessive production of neutrophil extracellular traps (NETs) contributes to immunothrombosis activation in ischemic stroke pathogenesis. The metabolic and mechanistic regulators of NET formation in relation to platelet activation during ischemic stroke remain poorly understood. In the present study, using multiple animal and clinical models of stroke, we examined the role of homocysteine and its biochemical modifications on NET formation and concomitant neuronal damage, platelet aggregation, motor and gait functions. Phosphoproteomics analysis of neutrophils in response to homocysteine reveal…

Neutrophil extracellular traps (NETs) have a dual role in the innate immune response to thermal injuries. NETs provide an early line of defence against infection. However, excessive NETosis can mediate the pathogenesis of immunothrombosis, disseminated intravascular coagulation (DIC) and multiple organ failure (MOF) in sepsis. Recent studies suggest that high interleukin-8 (IL-8) levels in intensive care unit (ICU) patients significantly contribute to excessive NET generation. This study aimed to determine whether IL-8 also mediates NET generation in patients with severe thermal injuries. IL-8…

Neutrophil extracellular traps (NETs), an important host defense mechanism, are assembled after the release of decondensed chromatin and other nuclear components by a process termed NETosis. However, excessive NET release destroys surrounding tissues, leading to conditions such as sepsis where platelets are implicated in the pathogenic switch of NETosis. Here, we show that platelets trigger iron accumulation and promote lipid peroxide production in neutrophils co-stimulated with lipopolysaccharide and platelets in vitro, resulting in the induction of NETosis. We also screened for compounds tha…

Ewing sarcoma (EWS) is a highly aggressive cancer with a survival rate of 70%-80% for patients with localized disease and under 30% for those with metastatic disease. Tumor-infiltrating neutrophils (TIN) can generate extracellular net-like DNA structures known as neutrophil extracellular traps (NETs). However, little is known about the presence and prognostic significance of tumor-infiltrating NETs in EWS. Herein, we investigated 46 patients diagnosed with EWS and treated in the Tel Aviv Medical Center between 2010 and 2021. TINs and NETs were identified in diagnostic biopsies of EWS by immuno…

Chronic rhinosinusitis (CRS) is characterized by inflammatory cell infiltration in the sinonasal mucosa. Eosinophil and neutrophil extracellular traps (EETs and NETs, respectively) are prominently found in CRS. This study aimed to investigate the effect of airborne fungi, Alternaria alternata and Aspergillus fumigatus, on EET and NET formation. Nasal epithelial cells, eosinophils, and neutrophils were isolated from eosinophilic CRS (ECRS), non-ECRS (NECRS), and healthy control. We determined eosinophil and neutrophil transepithelial migration after fungal treatment. We then determined the rele…

BACKGROUND: Complex interactions in the tumor microenvironment (TME) between bladder cancer (BLCA) and immune cells are critical for cancer progression. However, studies of neutrophil extracellular trap-associated long non-coding RNAs (NET-lncRNAs) in the TME of BLCA have not been reported. This study aims to screen for NET-lncRNAs in BLCA and to preliminarily explore their effects on BLCA development.
METHODS: The correlation of NET-related gene sets, which were identified from the cancer genome atlas (TCGA) BLCA datasets, with lncRNAs was analyzed and the prognosis-related genes were identi

In disease states with chronic inflammation, there is a crosstalk between mast cells and neutrophil granulocytes in the inflamed microenvironment, which may be potentiated by tryptase. In systemic mastocytosis (SM), mast cells are constitutively active and tryptase is elevated in blood. Mast cell activation in SM leads to symptoms from various organs depending on where the active mast cells reside, for example, palpitations, flush, allergic symptoms including anaphylactic reactions, and osteoporosis. Whether neutrophil function is altered in SM is not well understood. In the current study, we

OBJECTIVE: Gout flares that occur during urate-lowering therapy (ULT) are typically related to the shrinkage of tophi due to aggregated neutrophil extracellular traps (NETs) that have captured monosodium urate crystals in the tissues. The present study was undertaken to analyze the blocking effect of α1 -antitrypsin on neutrophil elastase, and it was found that α1 -antitrypsin induced rapid inflammation in the presence of unstable tophi.
METHODS: Cell-free DNA levels in serum samples were compared between patients who experienced a varying number of gout flares. We investigated whether cell-f…
