Preventive effect of flavonoids from Wushan Shencha (Malus doumeri leaves) on CCl4-induced liver injury.

Wushan Shencha (Malus doumeri leaf) is a unique tea-like drink. Herein, the effect of flavonoids from Wushan Shencha (FWSSC) on carbon tetrachloride-induced liver injury was studied. The serum and liver tissues of experimental mice were analyzed by kits, a slice technique, and qPCR assay. The liver index is a calculated liver-to-body weight ratio, and the experimental results showed that FWSSC reduced the liver index of the model group with liver injury, which was the highest. Sections stained with H&E showed that FWSSC reduced stem cell necrosis caused by liver injury. FWSSC reduced the serum…

Centrifugal partition chromatography elution gradient for isolation of sesquiterpene lactones and flavonoids from Anvillea radiata.

An innovative procedure coupling pressurized solvent extraction and centrifugal partition chromatography (CPC) used in linear gradient elution mode was developed to isolate two pure germacranolides (9α-hydroxyparthenolide and 9β-hydroxyparthenolide) and to separate flavonoids (nepetin, isorhamnetin and jaceosidin) and chlorophyll pigments from aerial parts of Anvillea radiata (Coss.&Durieu). The two main germacranolides recovered using this method represent 2 and 5% of the dried plant material respectively. These molecules were extracted using accelerated solvent extraction with chloroform. Af…

Dietary flavonoids fisetin and myricetin: dual inhibitors of Plasmodium falciparum falcipain-2 and plasmepsin II.

Malaria is one of the most devastating infectious diseases in the developing world. Until now, only one candidate malaria vaccine RTS,S/AS01 has shown modest protection in phase 3 trial in African infants. Hence the treatment of malaria still depends on the current chemotherapeutic drugs. Considering the resistance of malaria parasites to almost all used antimalarial drugs, aiming at multi-targets rather than a single target will be a more promising strategy. Previous studies have shown that myricetin and fisetin exhibited in vitro antimalarial activity against Plasmodium falciparum, but very

Oral metabolism and efficacy of Kalanchoe pinnata flavonoids in a murine model of cutaneous leishmaniasis.

Leishmaniasis is a parasitic disease that threatens 350 million people worldwide. In a search for new antileishmanial drugs, the in vitro activity of flavonoids from Kalanchoe pinnata (Crassulaceae) was previously demonstrated in infected cells. In order to demonstrate the safety and oral activity of K. pinnata, flavonoids were evaluated in vivo in a murine model of cutaneous leishmaniasis. Daily oral doses of quercetin 3-O-alpha-L-arabinopyranosyl (1-->2)-alpha-L-rhamnopyranoside, quercetin 3-O-alpha-L-rhamnopyranoside, and free quercetin (16 mg/kg body weight) all were able to control the le…

Antagonistic interactions between the flavonoids hesperetin and naringenin and beta-lactam antibiotics against Staphylococcus aureus.

Flavonoids are a group of polyphenolic plant compounds with a range of biological activities. This study shows that the flavonoids hesperetin and naringenin have antibacterial activity against methicillin-sensitive and methicillin-resistant Staphylococcus aureus isolates. Minimum inhibitory concentrations for hesperetin were 250 and 500 microg/mL, respectively, and for naringenin were 125 and 250 microg/mL, respectively. This effect was reversed by the beta-lactam antibiotics methicillin, penicillin and oxacillin, but not by cefoxitin. For bacteria growing in the presence of these antibiotics,…

Phytochemical Investigation of Stachys hissarica: Antioxidant Activity, Flavonoid Characterization, and In Silico Analysis.

Evaluating Stachys hissarica extracts via DPPH, FRAP, and ABTS assays revealed that the aqueous methanolic fraction exhibited strong, ascorbic acid-comparable antioxidant activity. This high potency confirms that hydrophilic phenolic compounds are the primary drivers of its radical-scavenging effect. HPLC-PDA/ESI-MS/MS analysis of the aqueous methanolic extract led to the tentative annotation of 35 metabolites, including hydroxycinnamic acid derivatives, flavonoid glycosides, acetylated and methoxylated flavones, consistent with the phytochemical profile of the Stachys genus. The antioxidant a

A Novel Ca2+-Dependent O-Methyltransferase Involved in Flavonoid Methylation in Tartary Buckwheat.

Methoxyflavonoids have attracted increasing attention because of their diverse biological activities and improved physicochemical properties. Although methylation is pivotal to methoxyflavonoid biosynthesis, the O-methyltransferases (OMTs) responsible for this process in Tartary buckwheat remain poorly characterized. In this study, the Tartary buckwheat cultivar "Y324" was used to systematically analyze the accumulation patterns and spatial distribution of methoxyflavonoids during achene development. Most methoxyflavonoids accumulated progressively during achene development, and they were spec…

BnaC03.MYB56 and BnaA09.MYB61 antagonistically regulate flavonoid metabolism in Brassica napus.

Flavonoid biosynthesis is negatively correlated with seed oil content in B. napus. However, the molecular mechanisms of flavonoid metabolism mediated by MYB repressor in B. napus are still not well understood. In this study, we identify an R2R3-MYB transcription factor, BnaC03.MYB56, which regulates flavonoid metabolism and oil content in B. napus. Overexpression of BnaC03.MYB56 reduced flavonoid metabolite levels and significantly increased seed oil content by 11.0%-11.4% compared to the wild-type, whereas the mutant lines exhibited the opposite phenotype. BnaC03.MYB56 directly binds to the "

Combinatorial Biosynthesis of Antithrombotic Flavonoid 6-O-Glycosides From Carthamus tinctorius.

Flavonoid 6-O-glycosides from Carthamus tinctorius (safflower) represent a unique class of potential antithrombotic agents. However, their pharmaceutical development has been hindered by limited natural resources. In this study, we characterized four key biosynthetic enzymes from C. tinctorius: Ct6OGT (6-O-glycosyltransferase), Ct7OGT (7-O-glycosyltransferase), Ct3OGT (3-O-glycosyltransferase), and CtFLS (flavonol synthase). Among them, Ct6OGT is the first identified high-regioselectivity flavonoid 6-O-glycosyltransferase. By combining these enzymes with upstream genes identified from safflowe

The Biosynthesis of a Fungal Flavonoid, Chlorflavonin, Precisely Controlled by Two Types of Equilibrium States.

Flavonoids are polyphenolic natural products predominantly isolated from plants and exhibit a diverse array of biological activities. Because they serve as important pharmaceuticals and nutraceuticals, there is a strong demand for their sustainable supply. Chlorflavonin is a rare fungal flavonoid with potent antitubercular activity. While its biosynthetic enzymes are expected to be valuable tools for application in fungal production of flavonoids, the biosynthetic pathway remains unknown. Here, we elucidate the complete biosynthetic pathway for chlorflavonin through detailed functional analysi…
