Lipases and hydrolases play important roles in diverse biological processes in fungi. Their roles in virulence in the phytopathogenic fungus Alternaria alternata remain poorly understood. The AaTgl1 protein of A. alternata is a putative lipase/hydrolase characterized by a triacylglycerol lipase domain, an α/β-hydrolase superfamily domain, and a catalytic triad (Ser353, Asp522, His548) within the conserved GxSxG motif. Sequence analysis revealed only 64-68% similarity to orthologs from other filamentous fungi, suggesting a specialized role in lipid metabolism that may contribute to the ecologic

Understanding the distribution of agaric fungi through molecular phylogeny is critical for biodiversity assessment. This comprehensive investigation documented the presence of Panaeolus subbalteatus, a previously unreported agaric fungus, addressing a significant gap in the country's knowledge of fungal diversity. This psilocybin-containing mushroom species, known for its neurotoxic properties, was collected from humus-rich soil in the Gujranwala district of Punjab province. The study employed integrated morpho-anatomical and molecular approaches, including detailed microscopic examination of

BACKGROUND: Three de novo designed low molecular weight cationic peptides (IJ2, IJ3 and IJ4) containing an unnatural amino acid α,β-didehydrophenylalanine (∆Phe) exhibited potent antifungal activity against fluconazole (FLC) sensitive and resistant clinical isolates of Candida albicans as well as non-albicans and other yeast and filamentous pathogenic fungi. In the present study, their synthesis, susceptibility of different fungi and the mechanism of anti-candidal action have been elucidated.
METHODS: The antimicrobial peptides (AMPs) were synthesized by solid-phase method and checked for ant

Recent expansion of immunocompromised population has led to significant rise in zygomycosis caused by filamentous fungus Rhizopus oryzae. Due to emergence of fungal resistance and side-effects of antifungal drugs, there is increased demand for novel drug targets. The current study elucidates molecular interactions of peptide drugs with G-6-P synthase (catalyzing the rate-limiting step of fungal cell wall biosynthetic pathway) of R.oryzae by molecular docking studies. The PDB structures of enzyme in R.oryzae are not known which were predicted using I-TASSER server and validated with PROCHECK. P

Aspergillus fumigatus is a human opportunistic fungal pathogen whose cell wall protects it from the extracellular environment including host defenses. Chitin, an essential component of the fungal cell wall, is synthesized from UDP-GlcNAc produced in the hexosamine biosynthetic pathway. As this pathway is critical for fungal cell wall integrity, the hexosamine biosynthesis enzymes represent potential targets of antifungal drugs. Here, we provide genetic and chemical evidence that glucosamine 6-phosphate N-acetyltransferase (Gna1), a key enzyme in this pathway, is an exploitable antifungal drug

Sporotrichosis is a polymorphic chronic infection of humans and animals classically acquired after traumatic inoculation with soil and plant material contaminated with Sporothrix spp. propagules. An alternative and successful route of transmission is bites and scratches from diseased cats, through which Sporothrix yeasts are inoculated into mammalian tissue. The development of a murine model of subcutaneous sporotrichosis mimicking the alternative route of transmission is essential to understanding disease pathogenesis and the development of novel therapeutic strategies. To explore the impact

Series of 4-(5-aryl-2H-tetrazol-2-yl)butan-2-ol, 1-(5-aryl-2H-tetrazol-2-yl)propan-2-ol and their acetates have been screened against Candida albicans. Among the tested compounds, (±)-1-[5-(2-chlorophenyl)-2H-tetrazol-2-yl]propan-2-yl acetate (E5) proved to be the most effective inhibitor of fungal growth and was further evaluated against young (adhesion phase) and mature biofilm in vitro. The activity exhibited by the tested tetrazole derivatives against C. albicans associated with minor cytotoxicity towards Vero epithelial cells make us suggest that E5 could be a promising structure in the d

The ABC transporter Cdr1 protein of Candida albicans, which plays a major role in antifungal resistance, has two transmembrane domains (TMDs) and two nucleotide-binding domains (NBDs). The 12 transmembrane helices of TMDs that are interconnected by extracellular and intracellular loops (ICLs) mainly harbor substrate recognition sites where drugs bind while cytoplasmic NBDs hydrolyze ATP which powers drug efflux. The coupling of ATP hydrolysis to drug transport requires proper communication between NBDs and TMDs typically accomplished by ICLs. This study examines the role of cytoplasmic ICLs of

Members of the pleiotropic drug resistance (PDR) family of ATP binding cassette (ABC) transporters consist of two homologous halves, each containing a nucleotide binding domain (NBD) and a transmembrane domain (TMD). The PDR transporters efflux a variety of hydrophobic xenobiotics and despite the frequent association of their overexpression with the multidrug resistance of fungal pathogens, the transport mechanism of these transporters is poorly understood. Twenty-eight chimeric constructs between Candida albicans Cdr1p (CaCdr1p) and Cdr2p (CaCdr2p), two closely related but functionally distin

Lobomycosis is a chronic cutaneous infection caused by the uncultivable fungus Paracoccidioides lobogeorgii. Lesions are often described as … modest differences in biglycan and decorin expression, with spatial redistribution of biglycan near fungal structures. Patients treated with itraconazole showed lesion stabilization. These findings indicate collagen loss rather than true keloid formation.

Five guanacastane-type diterpenoids, including a novel compound, guanacastepene V (1), and four known compounds (2-5) along with a novel diterpenoid moniliforminol C (6), were isolated from a marine-derived fungus, Coprinellus xanthothrix. The structures of the isolated compounds were elucidated using NMR spectroscopy and computational chemistry. In antifungal assays against Candida auris, an emerging fungal pathogen, compounds 1 and 2 exhibited inhibitory activity, with minimum inhibitory concentration values of 6.25 and 3.12 µg/mL, respectively. This antifungal effect was specific to C. auri

A 60-year-old immunocompromised patient developed rapidly progressive proptosis that was secondary to mucormycosis. This life-threatening fungal infection usually is associated with chemosis, proptosis, ophthalmoplegia, and visual loss. The fungus may invade ocular structures, sinuses, and extend into the brain. The standard of care includes correction of the underlying condition, administration of liposomal amphotericin B with posaconazole, and surgical debridement of infected and necrotic tissue. We present a case of unilateral proptosis due to mucormycosis in an immunocompromised patient. T

Primary pulmonary adenocarcinoma with metastatic spread to the brain stem was diagnosed at necropsy in an aged Bulldog. Initial clinical signs included coughing, decreased exercise tolerance, and personality changes. Primary differential diagnoses after thoracic radiography were multicentric metastatic neoplasia or fungal disease. Later clinical signs (inappetence, unilateral facial paralysis and decreased sensation, and hemiparesis) were referable to the effect of the tumor on cranial nerves V and VII and other brain stem structures. Primary lung tumors are uncommon, as are metastatic tumors

The impact of climate change induced habitat fragmentation on plant species and populations has been studied in numerous systems, but far less is known about how these processes shaped the population structure and demographic history of fungal symbionts. Here, we investigate the population structure and demographic history of ectomycorrhizal fungus Rhizopogon salebrosus, a symbiont of Pinus species, across the Madrean Sky Islands Archipelago (MSIA) of southern Arizona and northern Sonora. Rhizopogon salebrosus produces truffle-like sporocarps and depends on small mammals for spore dispersal. U

Indole-diterpenes are an important class of chemical compounds which can be unique to different fungal species. The highly complex lolitrem compounds are confined to Epichloë species, whilst penitrem production is confined to Penicillium spp. and Aspergillus spp. These fungal species are often present in association with pasture grasses, and the indole-diterpenes produced may cause toxicity in grazing animals. In this review, we highlight the unique structural variations of indole-diterpenes that are characterised into subgroups, including paspaline, paxilline, shearinines, paspalitrems, terpe
