Insights into functional pharmacology of α₁ GABA(A) receptors: how much does partial activation at the benzodiazepine site matter?

RATIONALE: Synthesis of ligands inactive or with low activity at α₁ GABA(A) receptors has become the key concept for development of novel, more tolerable benzodiazepine (BZ)-like drugs. WYS8, a remarkably (105 times) α₁-subtype selective partial positive modulator, may serve as a pharmacological tool for refining the role of α₁ GABA(A) receptors in mediation of BZs' effects. OBJECTIVES: Here, the effects of WYS8 on GABA-induced currents and on diazepam-induced potentiation of recombinant BZ-sensitive GABA(A) receptors were studied in more detail. In addition, the behavioral profile of WYS8 (0

Abnormal GABAA receptor-mediated currents in dorsal root ganglion neurons isolated from Na-K-2Cl cotransporter null mice.

We have recently disrupted Slc12a2, the gene encoding the secretory Na-K-2Cl cotransporter in mice (NKCC1) (Delpire et al., 1999). Gramicidin perforated-patch and whole-cell recordings were performed to study GABA-induced currents in dorsal root ganglion (DRG) neurons isolated from wild-type and homozygote NKCC1 knock-out mice. In wild-type DRG neurons, strong GABA-evoked inward current was observed at the resting membrane potential, suggesting active accumulation of Cl(-) in these cells. This GABA-induced current was blocked by picrotoxin, a GABA(A) receptor blocker. The strong Cl(-) accumula

Interaction of pentobarbital with gabaergic drugs acting on the Cl(-)-ATPase activity of the plasma membranes from bream brain (Abramis brama L.).

The present study was designed to investigate the role of the interaction of pentobarbital with gamma-aminobutyric acid (GABA)ergic drugs acting on the Cl(-)-adenosine triphosphate (ATP)ase activity of the plasma membranes fraction of bream brain. The preincubation and then incubation of the membranes with pentobarbital as well as with other GABAergic ligands was conducted at physiologic pH (7.4), i.e. at the condition where the Cl(-)-ATPase activity is not detected. Pentobarbital (1-100 microM) induces Cl(-)-ATPase activity, however at high concentration (1,000 microM) no effect of the ligand

Corticostriatal Projections Relying on GABA Levels Mediate Exercise-Induced Functional Recovery in Cerebral Ischemic Mice.

Stroke is a neurological disorder characterized by high disability and death worldwide. The occlusion of the middle cerebral artery (MCAO) supplying the cortical motor regions and its projection pathway regions can either kill the cortical neurons or block their projections to the spinal cord and subcortical structure. The cerebral cortex is the primary striatal afferent, and the medium spiny neurons of the striatum have been identified as the major output neurons projecting to the substantia nigra and pallidum. Thus, disconnection of the corticostriatal circuit often occurs in the model of MC

Astrocytic GABAergic signaling in perioperative neurocognitive disorders: mechanisms and intervention strategies.

Perioperative neurocognitive disorders (PND) are a major burden for older surgical patients, yet little is known about the cellular events that set them in motion, and no therapy currently changes their course. Anesthesia and amnesia after etomidate, sevoflurane, or isoflurane depend mainly on gamma-aminobutyric acid type A receptors (GABA(A)Rs), and an increasing body of work shows that astrocytes also respond to these drugs. This review examines an astrocyte-centered GABAergic cascade that may link anesthetic exposure to PND and grades the strength of the evidence at each node. When anesthet

The GABA(B1) receptor within the infralimbic cortex is implicated in stress resilience and vulnerability in mice.

Resilience is the capacity to maintain normal psychological and physical functions in the face of stress and adversity. Understanding how one can develop and enhance resilience is of great relevance to not only promoting coping mechanisms but also mitigating maladaptive stress responses in psychiatric illnesses such as depression. Preclinical studies suggest that GABA(B) receptors (GABA(B1) and GABA(B2)) are potential targets for the treatment of major depression. In this study, we assessed the functional role of GABA(B) receptors in stress resilience and vulnerability by using a chronic unpre

Attenuation of dopamine-induced GABA release in problem gamblers.

INTRODUCTION: We have previously shown that an interaction between medial prefrontal and parietal cortices is instrumental in promoting self-awareness via synchronizing oscillations in the gamma range. The synchronization of these oscillations is modulated by dopamine release. Given that such oscillations result from intermittent GABA stimulation of pyramidal cells, it is of interest to determine whether the dopaminergic system regulates GABA release directly in cortical paralimbic regions. Here, we test the hypothesis that the regulation of the GABA-ergic system by the dopaminergic system bec

Nociceptive spinal neurons expressing NK1 and GABAB receptors are located in lamina I.

The nociceptive nature of spinal dorsal horn neurons expressing NK1 and gamma-aminobutyric acid (GABA)(B) receptors was evaluated in the rat. Immunodetection of the Fos protein, induced by noxious mechanical stimulation of the skin, was combined with immunocytochemistry for NK1 or GABA(B) receptors (double-immunostaining study) or both receptors (triple-immunostaining study). Neurons double-labeled for Fos and for each receptor largely prevailed in lamina I. The proportions of Fos-positive cells immunostained for NK1 or GABA(B) receptors were higher in lamina I than in the remaining spinal lam

Women with PTSD have a changed sensitivity to GABA-A receptor active substances.

RATIONALE: The use of benzodiazepines in treating anxiety symptoms in patients with posttraumatic stress disorder (PTSD) has been debated. Studies on other anxiety disorders have indicated changed sensitivity to GABA-A receptor active substances. OBJECTIVE: In the present study, we investigated the GABA receptor sensitivity in PTSD patients. METHODS: Injections of allopreganolone, diazepam, and flumazenil were carried out, each on separate occasions, in 10 drug naïve patients with PTSD compared to 10 healthy controls. Effects were measured in saccadic eye velocity (SEV) and in subjective rat

GABA concentration sets the conductance of delayed GABAA channels in outside-out patches from rat hippocampal neurons.

GABAA channels were activated by GABA in outside-out patches from rat cultured hippocampal neurons. They were blocked by bicuculline and potentiated by diazepam. In 109 of 190 outside-out patches, no channels were active before exposure to GABA (silent patches). The other 81 patches showed spontaneous channel activity. In patches containing spontaneous channel activity, rapid application of GABA rapidly activated channels. In 93 of the silent patches, channels could be activated by GABA but only after a delay that was sometimes as long as 10 minutes. The maximum channel conductance of the chan

Functional pharmacology of GABA(A) receptors containing the chicken brain gamma 4 subunit.

The functional pharmacology of receptors composed of the chicken brain GABA(A) receptor gamma 4 subunit and the mammalian GABA(A) receptor alpha 3 and beta2 subunits was studied by heterologous expression in Xenopus laevis oocytes using the two electrode voltage-clamp technique. GABA-evoked currents had an EC(50) of 180+/-30 microM. Responses were blocked by the competitive and non-competitive GABA(A) receptor antagonists, bicuculline methochloride and picrotoxin. Sodium pentobarbital reversibly potentiated the current several-fold, and Zn(2+) ions blocked the current with high potency (IC50=2

Pharmacologic Treatment with GABA(B) Receptor Agonist of Methamphetamine-Induced Cognitive Impairment in Mice.

Methamphetamine (METH) is a highly addictive drug, and addiction to METH has increased to epidemic proportions worldwide. Chronic use of METH causes psychiatric symptoms, such as hallucinations and delusions, and long-term cognitive deficits, which are indistinguishable from paranoid schizophrenia. The GABA receptor system is known to play a significant role in modulating the dopaminergic neuronal system, which is related to behavioral changes induced by drug abuse. However, few studies have investigated the effects of GABA receptor agonists on cognitive deficits induced by METH. In the presen

Activation of the dorsal vagal nucleus increases pancreatic exocrine secretion in the rat.

Pancreatic secretion is regulated by the dorsal vagal nucleus (DVN) which is modulated by several neurotransmitters and diverse synaptic inputs. The inhibitory neurotransmitter GABA is a major modulator of the vagal output to the gastrointestinal tract. The present study investigated the effects of GABA(A) receptor blockade in the DVN, using bicuculline methiodide (BIM, GABA(A) receptor antagonist, 100 pmol/25 nl), on pancreatic exocrine secretion (PES). Male Sprague-Dawley rats anaesthetised with isoflurane were used in all experiments. PES was collected from the common bile-pancreatic duct a

Neurobiological background for the development of new drugs in schizophrenia.

Psychopharmacology of schizophrenia has remained static for many years because the mechanisms explored have been basically monoaminergics, primarily focused toward the modification of dopaminergic function and, later on, serotonergic. In fact, most of the antipsychotics introduced in clinical practice in the last years have been antagonists or selective agonists of these receptors (D(2)/5-HT(2)). The exploration of other receptor pathways, and in particular those additionally involved in the action of the paradigmatic "atypical" antipsychotic clozapine (ie, cholinergic and noradrenergic), has

Development of 1,2,4-oxadiazole-containing isoxazoline insecticides with improved honeybee compatibility.

BACKGROUND: Spodoptera frugiperda (S. frugiperda) and Plutella xylostella (P. xylostella) are major constraints to staple and cruciferous crops, respectively, and have evolved widespread resistance to anthranilic amide insecticides. Isoxazolines, acting as γ-aminobutyric acid (GABA) receptor modulators, are promising alternatives for controlling resistant populations. However, many members of this class exhibit high toxicity toward ecologically important pollinators. Here, we report a series of new isoxazoline analogues featuring a 1,2,4-oxadiazole motif designed by a ring-closure strategy to

Is there a physiological role for the neurosteroid THDOC in stress-sensitive conditions?

Endogenous neurosteroids affect brain excitability during physiological states such as pregnancy and the menstrual cycle, and during conditions of acute and chronic stress. The neurosteroid allotetrahydrodeoxycorticosterone (THDOC) is an allosteric modulator of the GABA(A) receptor. Although the role of THDOC within the brain is undefined, recent studies indicate that stress induces THDOC to levels that can activate GABA(A) receptors. These results might have significant implications for human stress-sensitive conditions such as epilepsy, post-traumatic stress disorder and depression.

Synergistic Enhancement of Fecal GABA Content by Bifidobacterium adolescentis 4-2 and Mannooligosaccharides in a Human Intestinal Flora Model.

BACKGROUND: Gamma-aminobutyric acid (GABA) is the principal inhibitory neurotransmitter in the central nervous system and exerts multiple health-promoting effects, including antidepressant, hypotensive, immunomodulatory, and antidiabetic actions. Previous studies have reported a positive correlation between the abundance of Bifidobacterium species in the gut and fecal GABA levels. However, the direct contribution of specific GABA-producing Bifidobacterium strains and the associated interaction with prebiotics remain insufficiently characterized. METHODS: This study aimed to elucidate the dire

Striatal, pallidal, and pars reticulata evoked inhibition of nigrostriatal dopaminergic neurons is mediated by GABA(A) receptors in vivo.

Dopaminergic neurons express both GABA(A) and GABA(B) receptors and GABAergic inputs play a significant role in the afferent modulation of these neurons. Electrical stimulation of GABAergic pathways originating in neostriatum, globus pallidus or substantia nigra pars reticulata produces inhibition of dopaminergic neurons in vivo. Despite a number of prior studies, the identity of the GABAergic receptor subtype(s) mediating the inhibition evoked by electrical stimulation of neostriatum, globus pallidus, or the axon collaterals of the projection neurons from substantia nigra pars reticulata in v

Participation of the GABAergic system on the glutamate release of frontal cortex synaptosomes from Wistar rats with experimental autoimmune encephalomyelitis.

We previously found that the glutamate release was decreased in synaptosomes from rat cerebral cortex during the development of experimental autoimmune encephalomyelitis (EAE), the animal model of multiple sclerosis. Various other reports have shown a deficit in the expression of proteins associated with GABAergic neurotransmission in the neocortex of patients with multiple sclerosis and it was also demonstrated that the activation of GABAA receptors leads to an inhibition of glutamate release. Now, in order to evaluate the events that may affect the neuronal function in EAE synaptosomes, we a

Classic benzodiazepines modulate the open-close equilibrium in alpha1beta2gamma2L gamma-aminobutyric acid type A receptors.

BACKGROUND: Classic benzodiazepine agonists induce their clinical effects by binding to a site on gamma-aminobutyric acid type A (GABAA) receptors and enhancing receptor activity. There are conflicting data regarding whether the benzodiazepine site is allosterically coupled to gamma-aminobutyric acid binding versus the channel open-close (gating) equilibrium. The authors tested the hypothesis that benzodiazepine site ligands modulate alpha1beta2gamma2L GABAA receptor gating both in the absence of orthosteric agonists and when the orthosteric sites are occupied. METHODS: GABAA receptors were r
