Cocaine-induced immediate-early gene expression in the nucleus accumbens: roles of separate cAMP sensors.

Immediate-early gene (IEG) induction after administration of amphetamine or cocaine has been used to trace the signaling pathways that mediate neuronal plasticity required for the short- and long-term behavioral effects of these psychostimulants. We recently reported that a novel cyclic AMP (cAMP)-dependent Rap guanine nucleotide exchange factor-2 (RapGEF2)-ERK signaling pathway is required for Egr-1 induction in D1 medium-spiny neurons (MSNs) of the nucleus accumbens (NAc) after cocaine treatment, and that its deletion from the NAc neurons attenuates cocaine-induced locomotor sensitization an

RRR-alpha-tocopheryl succinate inhibits EL4 thymic lymphoma cell growth by inducing apoptosis and DNA synthesis arrest.

RRR-alpha-tocopheryl succinate (vitamin E succinate, VES) treatment of murine EL4 T lymphoma cells induced the cells to undergo apoptosis. After 48 hours of VES treatment at 20 micrograms/ml, 95% of cells were apoptotic. Evidence for the induction of apoptosis by VES treatments is based on staining of DNA for detection of chromatin condensation/fragmentation, two-color flow-cytometric analyses of DNA content, and end-labeled DNA and electrophoretic analyses for detection of DNA ladder formation. VES-treated EL4 cells were blocked in the G1 cell cycle phase; however, apoptotic cells came from a

MOLECULAR MECHANISMS OF DIABETIC PERIODONTITIS: IDENTIFICATION OF KEY OXIDATIVE STRESS-RELATED GENES AND POTENTIAL THERAPEUTIC ROLE OF METFORMIN THROUGH MMP14 AND PXDN.

AIM: Periodontitis and diabetes mellitus (DM) exhibit a bidirectional relationship, with oxidative stress potentially serving as a crucial pathological bridge. However, the underlying molecular mechanisms remain unclear. METHODS: We identified differentially expressed genes (DEGs) related to oxidative stress in periodontitis and diabetes. Consensus clustering was performed to identify disease subtypes. Machine learning algorithms, including Support Vector Machine, LASSO regression, and Random Forest, were employed to identify key regulatory genes. Molecular docking was conducted to explore th

Identification of Key Genes and Immune Infiltrate in Nonalcoholic Steatohepatitis: A Bioinformatic Analysis.

Источник пометил публикацию как отозванную

BACKGROUND: Nonalcoholic steatohepatitis (NASH) can progress to cirrhosis and hepatic carcinoma and is closely associated with changes in the neurological environment. The discovery of new biomarkers would aid in the treatment of NASH. METHODS: Data GSE89632 were downloaded from the Gene Expression Omnibus (GEO) database, and R package "limma" was used to identify differentially expressed genes (DEGs) for NASH vs. normal tissues. The STRING database was used to construct a protein-protein interaction (PPI) network, and the Cytoscape software program (Version 3.80) was used to visualize the PP

Perineuronal nets in cerebellar nuclei neurons orchestrate social behaviour via regulation of neuronal activity in circuits innervated by the cerebellum.

Certain types of neurons in the central nervous system are wrapped in extracellular matrix protein complexes named perineuronal nets (PNNs). While it is known that PNNs regulate neuronal activity by modulating synaptic plasticity, their pathophysiological role in psychiatric disorders has not been sufficiently clarified. Here, we demonstrate that the expression of PNNs in autism-spectrum-disorder (ASD)-associated mice, valproic acid-injected mice and chromodomain helicase DNA-binding protein 8 (Chd8) gene haploinsufficient mice was decreased in cerebellar nuclei neurons. The pharmacological di

A bioinformatics analysis of potential cellular communication networks in non-alcoholic steatohepatitis and colorectal adenoma using scRNA-seq and bulk-seq.

BACKGROUND: Non-alcoholic fatty liver disease (NAFLD) is the global most common chronic liver disease. Non-alcoholic steatohepatitis (NASH), an inflammatory subtype of NAFLD, has been shown to significantly increase the risk of colorectal adenoma (CRA). Therefore, from the perspective of bioinformatics analysis, the potential mechanisms of NASH/NAFLD-CRA can be explored. METHODS: In this study, we screened the differentially expressed genes (DEGs) and core effect pathways between NASH and CRA by analyzing the single-cell data of CRA patients and the high-throughput sequencing data (GSE37364 a

Effects of dietary long-chain polyunsaturated fatty acids on osmoregulation and salinity adaptation in red tilapia (Oreochromis mossambicus♀ × O. niloticus♂) under salinity stress.

The roles of long-chain polyunsaturated fatty acids (LC-PUFA) in osmoregulation and salinity adaptation in teleost fish were investigated in red tilapia (Oreochromis mossambicus♀ × O. niloticus♂). Groups of juvenile fish were reared initially in freshwater (FW, 0 ppt) or seawater (SW, 32 ppt) and fed diets containing either LC-PUFA-rich fish oil (FO) or rapeseed oil (VO) that lacks LC-PUFA, but is rich in C18 PUFA. After 8 weeks of feeding, the four groups of fish were subjected to salinity stress by directly changing to brackish water (BW, 16 ppt). Fish were sampled after 0 h, 3 h, 6 h, 1 d,

A fructo-oligosaccharide and sea buckthorn complex ameliorates glucose and lipid metabolism in type 2 diabetic (T2DM) rats via metabolic and anti-inflammatory pathways.

This study investigated the effects and underlying mechanisms of a fructo-oligosaccharide (FOS) and sea buckthorn complex (FS) on blood glucose and lipid metabolism in type 2 diabetic mellitus (T2DM) rats. The T2DM model was induced by a high-fat diet (HFD) combined with alloxan administration. Prior to rat experiments, the synergistic hypoglycemic and hypolipidemic effects of FOS and sea buckthorn have been verified by zebrafish experiments. FS intervention significantly reduced fasting blood glucose, triglycerides (TG), total cholesterol (TC), and low-density lipoprotein cholesterol (LDL-C)

Sesamolin Protects Mice From Ovariectomized Bone Loss by Inhibiting Osteoclastogenesis and RANKL-Mediated NF-κB and MAPK Signaling Pathways.

This article was submitted to Experimental Pharmacology and Drug Discovery, a section of the journal Frontiers in Pharmacology. Postmenopausal osteoporosis (PMOP), which increases the risk of fracture, is the most common bone disease in women. PMOP not only increases the risk of death but also imposes a financial burden on countless families. At present, most of the drugs used to treat osteoporosis have significant side effects, so it is important to find effective anti-osteoporosis medications without major side effects. Sesamolin (Ses) is a kind of natural lignan extracted from sesame oil. M

Activator protein-1 transactivation of the major immediate early locus is a determinant of cytomegalovirus reactivation from latency.

Human cytomegalovirus (HCMV) is a ubiquitous pathogen that latently infects hematopoietic cells and has the ability to reactivate when triggered by immunological stress. This reactivation causes significant morbidity and mortality in immune-deficient patients, who are unable to control viral dissemination. While a competent immune system helps prevent clinically detectable viremia, a portrait of the factors that induce reactivation following the proper cues remains incomplete. Our understanding of the complex molecular mechanisms underlying latency and reactivation continues to evolve. We prev

Increased expression of NGFI-A mRNA in the rat striatum following burst stimulation of the medial forebrain bundle.

Using in situ hybridization, we examined the mRNA expression for several immediate early genes in dopamine-innervated brain areas following electrical burst vs. regular stimulation of the medial forebrain bundle in anaesthetized rats. Two hours after 5 Hz burst stimulation, the expression of the nerve growth factor-inducible clone A (NGFI-A) mRNA was increased in the medial part of the striatum. This increase was prevented by pretreatment with the dopamine-D1 receptor antagonist, SCH23390 (0.1 mg/kg i.p.). After 8 Hz burst stimulation, NGFI-A mRNA expression was increased in the medial, centra

The Crucial Role of Cerebellar Crus I in Electroacupuncture Pretreatment for Attenuating Myocardial Ischemia-Reperfusion Injury.

BACKGROUND: Electroacupuncture pretreatment (EA-pre) has been shown to help reduce myocardial ischemia-reperfusion injury (MIRI), but the underlying mechanism remains unclear. Our previous studies indicated that EA activates the cerebellar cortex, specifically the Crus Ⅰ. However, whether activation of the Crus Ⅰ contributes to the attenuation of MIRI induced by EA-pre remains unclear. This study investigated the possible relationship between EA-induced relief of MIRI and the activation of Crus Ⅰ. METHODS: Electrocardiogram recording, echocardiography, and cardiac histology staining were used

Optogenetic stimulation of medial prefrontal cortex excites GABAergic cells in the nucleus accumbens and hippocampus of Wistar-Kyoto rats exposed to chronic mild stress.

BACKGROUND: High frequency optogenetic stimulation (OGS) of prelimbic cortex (PLC) has been reported to exert antidepressant-like effects in the chronic mild stress model of depression in Wistar Kyoto (WKY) rats, which are non-responsive to antidepressant drugs. Here we have examined the effect of OGS on activity in the PLC and in two other regions implicated in depression, the nucleus accumbens (NAc) and hippocampus (HPC). METHOD: OGS was applied to the PLC of WKY rats using the same stress schedule, and the identical placement, virus infection and stimulation parameters, used in the earlier

Amygdala stimulation ameliorates memory impairments and promotes c-Fos activity in fimbria-fornix-lesioned rats.

Recently we provided data showing that amygdala stimulation can ameliorate spatial memory impairments in rats with lesion in the fimbria-fornix (FF). The mechanisms for this improvement involve early gene expression and synthesis of BDNF, MAP-2, and GAP43 in the hippocampus and prefrontal cortex. Now we have studied which brain structures are activated by the amygdala using c-Fos as a marker of neural activation. First, we studied neuronal activation after tetanic stimulation to the amygdala in intact rats. We then carried out a second study in FF-lesioned rats in which the amygdala was stimul

Afferents from vocal motor and respiratory effectors are recruited during vocal production in juvenile songbirds.

Learned behaviors require coordination of diverse sensory inputs with motivational and motor systems. Although mechanisms underlying vocal learning in songbirds have focused primarily on auditory inputs, it is likely that sensory inputs from vocal effectors also provide essential feedback. We investigated the role of somatosensory and respiratory inputs from vocal effectors of juvenile zebra finches (Taeniopygia guttata) during the stage of sensorimotor integration when they are learning to imitate a previously memorized tutor song. We report that song production induced expression of the imme

D2-40 expression in primary scarring and nonscarring alopecia.

The pathogenesis of scarring alopecia is believed to be related to damage to stem cells within, and outside the confines of, the follicular bulge region. Scattered reports indicate that D2-40, a monoclonal antibody that specifically detects a fixation-resistant epitope on podoplanin, highlights the basal cell layer of the outer root sheath. Given this, we sought to ascertain involvement of the same in a total of 52 cases of alopecia (33 scarring and 19 nonscarring). D2-40 expression, noted in the peripheral layer of the outer root sheath (above and below attachment of the pilar muscle), was si

The effects of noxious dental heating on the jaw-opening reflex and trigeminal Fos expression in the ferret.

Previous studies have established that the activation of peripheral nociceptors alters the central processing of nociceptive stimuli. In this study, we examined whether noxious heating of the dental pulp enhances the nociceptive jaw-opening reflex (JOR) and the expression of the immediate early gene c-fos in chloral hydrate/pentobarbital-anesthetized ferrets. We hypothesized that the application of noxious heat to the dental pulp, a procedure that evokes a preferential activation of pulpal C-fibers, will enhance JOR responses to electrical stimulation of the tooth pulp and that this enhanced r
