Causal effects of gut microbiota on chalazion: a two-sample Mendelian randomization study.

PURPOSE: To investigate the causal relationship between gut microbiota (GM) and chalazion through Mendelian randomization (MR) analysis. METHODS: GM-related genome-wide association studies (GWAS) were obtained from the International Consortium MiBioGen. Genetic data for chalazion were sourced from the MRC Integrative Epidemiology Unit (IEU) Open GWAS database. Five MR methods, including inverse variance weighted (IVW), were employed to estimate causal relationships. Cochran's Q test was used to detect heterogeneity, the MR-Egger intercept test and MR-PRESSO regression were utilized to detect

A Mendelian Randomization Study of Plasma Homocysteine Levels and Cerebrovascular and Neurodegenerative Diseases.

Background: Homocysteine (Hcy) is a toxic amino acid and hyperhomocysteinemia (HHcy) was reported to be associated with both cerebrovascular disease and neurodegenerative disease. Our aim was to assess the causal link between plasma Hcy level and cerebrovascular and neurodegenerative diseases through a Mendelian randomization (MR) study. Methods: A two-sample MR study was performed to infer the causal link. We extracted the genetic variants (SNPs) associated with plasma Hcy level from a large genome-wide association study (GWAS) meta-analysis. The main MR analysis was performed using the inver

Decoding the lipid-migraine link: a genetic and lipidomic investigation of migraine subtypes.

BACKGROUND: Migraine frequently co-occurs with cardiovascular and metabolic diseases. Observational studies examining the association between conventional lipid profiles and migraine risk have yielded inconsistent results and cannot establish causality. This study aimed to investigate the causal effects of specific lipid species on migraine and its primary subtypes: migraine with aura (MA) and without aura (MO). METHODS: Using a Mendelian randomization (MR) methodology, this study analyzed genome-wide association study (GWAS) data from the UK Biobank and FinnGen Consortium. Exposures comprise

Multivariable Mendelian randomization: the use of pleiotropic genetic variants to estimate causal effects.

A conventional Mendelian randomization analysis assesses the causal effect of a risk factor on an outcome by using genetic variants that are solely associated with the risk factor of interest as instrumental variables. However, in some cases, such as the case of triglyceride level as a risk factor for cardiovascular disease, it may be difficult to find a relevant genetic variant that is not also associated with related risk factors, such as other lipid fractions. Such a variant is known as pleiotropic. In this paper, we propose an extension of Mendelian randomization that uses multiple genetic

Genetic counselling in ALS: facts, uncertainties and clinical suggestions.

The clinical approach to patients with amyotrophic lateral sclerosis (ALS) has been largely modified by the identification of novel genes, the detection of gene mutations in apparently sporadic patients, and the discovery of the strict genetic and clinical relation between ALS and frontotemporal dementia (FTD). As a consequence, clinicians are increasingly facing the dilemma on how to handle genetic counselling and testing both for ALS patients and their relatives. On the basis of existing literature on genetics of ALS and of other late-onset life-threatening disorders, we propose clinical sug

Obstetrical Challenges in Robinow Syndrome.

Robinow syndrome is a genetically heterogenous syndrome that exhibits great pleiotropy, involving skeletal genital, cardiac, and craniofacial developmental anomalies. Fertility is not always compromised, and many individuals may be able to have a healthy pregnancy. Similar to other more common skeletal dysplasias and growth disorders such as achondroplasia, there are several challenges to be addressed in managing physiologic differences that occur in the context of pregnancy, and published literature centers on pregnant people with achondroplasia. We present a patient with Robinow syndrome (RO

Biologically informed instrument selection for dietary Mendelian randomization using chemosensory receptor variants.

BACKGROUND: Mendelian randomization (MR) is increasingly used for causal inference in nutritional epidemiology; however, dietary MR studies often rely on instruments statistically selected from genome-wide association studies of self-reported intake, which are vulnerable to pleiotropy and reverse causation and may violate core MR assumptions. We aimed to develop and evaluate a biologically informed framework for selecting valid genetic instruments for dietary exposures, based on genes encoding taste and olfactory receptors that mediate chemosensory inputs and shape food preferences and dietary

Type 1 diabetes mellitus increases the risk of sudden sensorineural hearing loss: A two-sample Mendelian randomization study.

OBJECTIVES: Diabetes mellitus is closely associated with sudden sensorineural hearing loss (SSNHL), but no definitive evidence has established a causal relationship between type 1 diabetes mellitus (T1DM) and SSNHL. This study aims to investigate the impact of T1DM on SSNHL from a genetic perspective, providing insights for risk prediction and treatment strategies. METHODS: Genetic data related to exposure (T1DM) and outcome (SSNHL) were obtained from publicly available genome-wide association studies (GWAS). Instrumental variables were selected, and Mendelian randomization (MR) analysis was

Refining Genetic Instruments for Dietary Intake Mendelian Randomization Using Phenome-Wide Association Studies.

Most Mendelian randomization (MR) of dietary intake use the full set of genome-wide significant (GWS) variants in the instrumental variable (IV), likely biasing causal estimates due to pleiotropy. To characterize the common methods to handle pleiotropy in dietary intake MR, we conducted a scoping review of the literature on dietary intake MR studies. We extracted information on IV construction, assessment of pleiotropy, and sensitivity analyzes revealing that only 20% of studies used an IV with functional plausibility. In the absence of functionally-informed IVs, we tested if two-sample MR usi

Antagonistic pleiotropy at the stem solidness 1 locus balances wheat stem architecture and yield components under terminal drought.

Stem structural investment enhances grain size and yield under drought, challenging the traditional view of vegetative-reproductive trade-offs. Antagonistic pleiotropy at the SSt1 locus reveals a genetic constraint between stem diameter and solidness. Incorporating genotype-by-environment into genomic selection models improves prediction accuracy by 46.5%. Terminal drought is the primary environmental constraint on wheat productivity in the Mediterranean basin, a challenge intensifying under climate change. While stem biomass investment is known to promote grain-filling under stress, its under

Pleiotropic roles of cold shock proteins with special emphasis on unexplored cold shock protein member of Plasmodium falciparum.

The cold shock domain (CSD) forms the hallmark of the cold shock protein family that provides the characteristic feature of binding with nucleic acids. While much of the information is available on bacterial, plants and human cold shock proteins, their existence and functions in the malaria parasite remains undefined. In the present review, the available information on functions of well-characterized cold shock protein members in different organisms has been collected and an attempt was made to identify the presence and role of cold shock proteins in malaria parasite. A single Plasmodium falci

The causal effects of primary biliary cholangitis on irritable bowel syndrome: a mendelian randomization study.

BACKGROUND: The gut-liver axis indicates a potential relationship between the primary biliary cholangitis (PBC) and irritable bowel syndrome (IBS). Nonetheless, the causality of this association remains unclear. This study performed a two-sample Mendelian randomization (MR) approach to investigate the causal relationship between the PBC and IBS. METHODS: We used public genome-wide association study (GWAS) datasets for PBC and IBS, treating PBC as the exposure and IBS as the outcome, in an MR analysis primarily employing the inverse-variance weighted (IVW) method.
