High-velocity resistance training improves the force-velocity profile in middle-aged and older adults.

The main goal of the present study was to examine the effects of a high-velocity resistance training (HVRT) program on the force-velocity (F-V) profile of different groups of aging adults. An additional aim was to explore whether individual baseline F-V profiles were associated with training-induced improvements in muscle power and functional capacity. Middle-aged and older adults with and without mobility limitations (n = 36, 61 ± 13 years) completed a 12-week HVRT program (2 days/week, 40-60% 1-RM). F-V profiles were determined using leg press exercises at loads ranging from 30-90% 1-RM, and

Enhancing fall risk assessment in the oldest old: The interplay of cognition and gait speed.

Individuals 90 + years have a high prevalence of falls, often with devastating consequences. Fall risk assessment and early intervention can prevent falls, yet factors associated with fall risk are understudied in this high-risk group. We aimed to evaluate whether slow gait, impaired cognition, or a combination of the two are associated with elevated fall risk in the oldest old. Participants were from The 90 + Study, a longitudinal study with evaluations every six months. We tested the association of 'slow gait' (Four-Meter Walk Test, below sex-specific median), 'impaired cognition' (cognitive

Effects of whole-body vibration on cognitive function: a systematic review and meta-analysis.

Whole-body vibration exercise (WBV) is a low-intensity intervention for improving cognitive function, particularly among individuals who are older or have physical limitations. However, quantitative evidence regarding its effectiveness is limited. This meta-analysis aimed to quantify the effects of WBV on cognitive function and explore major moderating variables and differences across cognitive domains. Sixteen randomized controlled trials (RCTs) were included, yielding 37 effect sizes (Hedges' g) related to cognitive outcomes. Effect sizes were estimated for overall cognitive function and fou

Clonal hematopoiesis mutation is associated with risk of incident lung cancer and death.

Clonal hematopoiesis (CH) has emerged as a potential risk factor with leukemia and some solid tumors, but the association between CHIP and lung cancer risk or death remains to be elucidated. This study utilized a longitudinal cohort design derived from the UK Biobank, incorporating whole-exome sequencing to identify CH mutations. The analysis included a nested 1:5 case-control of 3750 incident lung cancer cases and 18,687 controls matched on age and year at blood draw, sex, race, and smoking status. Mediation analyses were employed to explore the role of inflammatory markers and Mendelian rand

The influence of aging on graded peripheral venous return and complex blood flow features through human veins.

Aging is associated with reduced venous compliance and altered muscle pump function, though how these features impact venous return during volume-stressed conditions remains poorly characterized. We examined lower-limb venous flow dynamics and muscle pump efficiency between healthy younger (YA) and older adults (OA) under two volume-stressed conditions. Fourteen YA (7F, 26 ± 4 years) and fourteen OA (9F, 76 ± 6 years) underwent assessments at head-down tilt (HDT, -3°) and head-up tilt (HUT, + 20°) positions. Participants performed isometric plantarflexion contractions at 8 kg and 50% maximum v

Metabolic signatures, vitamin D, and red cell distribution width in dementia risk: UK Biobank insights.

Red cell distribution width (RDW), reflecting erythrocyte size variability, and serum vitamin D are emerging biomarkers for dementia. We examined the independent and joint associations of RDW and 25-hydroxyvitamin D [25(OH)D] with incident dementia and the potential mediating role of plasma metabolomic profiles in a large prospective cohort study. We analyzed data from 162,606 UK Biobank participants aged ≥ 50 years and dementia-free with RDW and 25(OH)D measurements at baseline. Incident dementia was identified via electronic health records for ≤ 15 years. Cox proportional hazards models adju

Psychosocial adversity captured by ICD-10 Z-codes and mortality risk: evidence from the UK Biobank.

Psychosocial adversity is a major determinant of health, but the mortality associations of its clinical documentation via ICD-10 Z55-Z65 codes remain unclear. We conducted a matched cohort study of UK Biobank participants with linked Hospital Episode Statistics and mortality records. Participants with ≥ 1 ICD-10 Z55-Z65 code recorded between 2010 and 2020 were propensity score-matched (1:1) to hospitalized participants without Z-codes. Mortality follow-up occurred from 2020 to 2023. Associations with all-cause mortality were estimated using Cox proportional hazards models, and cause-specific m

Dietary composition and metabolic stress differentially affect the ovarian microenvironment in mice.

The ovarian microenvironment is a dynamic target of aging and highly responsive to metabolic and dietary stressors. Western diets (WD) and high-fat diets (HFD) promote obesity and systemic metabolic dysfunction. However, whether these diets induce remodeling of the ovarian microenvironment remains unclear. In this study, we investigated how distinct dietary patterns influence metabolic homeostasis, estrous cyclicity, ovarian reserve, and stromal remodeling in female mice. Three-month-old C57BL/6 mice (n = 48) were fed a control diet (CTL), low-calorie Western diet (LcWD), WD, or HFD for 17 wee

Sex- and age-specific patterns in bone, muscle, and fat across adulthood and their relation to osteosarcopenic adiposity phenotype: a cross-sectional study.

Evidence suggests that age-associated differences in body composition-bone/muscle/adipose tissue-are nonlinear, but their timing, sex-specific features, and relation to osteosarcopenic adiposity (OSA) phenotype remain poorly defined. OSA is characterized by the coexistence of osteopenia/osteoporosis, sarcopenia, and excess or redistributed adiposity. We examined sex- and age-specific patterns and inflection points for bone, muscle, fat, and intramuscular adipose tissue (IMAT) in cross-sectional data from 9717 Caucasian adults aged 20-90 years. Body composition was assessed using BIA-ACC, a non

Multimorbidity as a predictor of mortality in companion dogs.

Multimorbidity, the presence of two or more conditions, is associated with a higher risk of death as individuals age. However, modeling multimorbidity in laboratory animals is difficult, if not impossible, because specific conditions are seldom individually diagnosed and treated in these settings. Because of their shared environment, physiology, and genetic diversity, and because they are medically managed as individuals, companion dogs have potential to serve as a translational multimorbidity model. Yet it is unknown how diagnoses accumulate over time, how these diagnoses are associated with

Feasibility of home-based, remotely delivered exercise training to improve physical function in older sepsis survivors: a pilot randomized controlled trial.

Sepsis survivors frequently experience post-discharge physical function decline and encounter logistical barriers to participation in traditional outpatient rehabilitation. This pilot study examined the feasibility, safety, and preliminary efficacy of a remotely delivered, home-based exercise program on physical function in older sepsis survivors. In this single-blinded, randomized controlled trial (RCT), sepsis survivors aged ≥ 55 years were randomized to (a) 12 weeks of avatar-guided, progressive exercise training group (EX) or (b) a standard care control group (CO). Primary outcomes were fe

Is morbidity compressing or expanding? Cross-national trends in midlife multimorbidity across cohorts.

While life expectancy has increased globally, it remains uncertain whether recent generations are entering midlife with a greater or lower burden of chronic disease and whether these cohort patterns differ by gender. Moreover, cross-national evidence comparing cohort differences in morbidity remains limited, constraining our understanding of how health trajectories vary across national contexts. Using harmonized longitudinal data from the Health and Retirement Study and 3 international partner studies (13 countries in total), we compared two midlife birth cohorts-adults aged 50-58 surveyed in

Lifetime non-relational traumatic experiences are associated with biological ageing.

Exposure to non-relational trauma, such as serious accidents, war or life-threatening illness, is linked to poor mental and physical health. Its relationship with biological ageing markers, however, remains underexplored. This study's aim was to examine associations between non-relational trauma and multiple biological ageing markers, and to assess whether associations vary by trauma burden, trauma type and sex. We analysed UK Biobank data from 152,863 participants (mean age = 56.4 years; 56.5% female). Lifetime exposure to six non-relational traumatic experiences was assessed. Biological agei

Identification and phenotypic profiling of subgroups with distinct cognitive aging trajectories.

Cognitive aging is shaped by genetic variation, environmental factors, and health-related conditions. Until now, it is largely unclear why some individuals maintain their cognitive function, and others show progressive cognitive decline. This study uncovered determinants of distinct cognitive aging trajectories in the older population. To approach the inter-individual variability in cognitive aging, we clustered n = 696 dementia-free individuals from the prospective TREND study based on their longitudinal changes in comprehensive cognitive testing every 2 years over 13 years on average. Development of a multimorbidity health conditions outcome index for caloric restriction interventional studies in older adults: a preliminary investigation in an observational cohort study.

Using multiple sources, we provide the conceptual justification and statistical support for a multimorbidity outcome associated with obesity-related conditions, which we term the Health Conditions Index (HCI). This index was designed to capture the health effects of multi-year studies of caloric restriction for older adults with BMIs in the overweight or obesity classification. We used a subset of participants in the Health, Aging and Body Composition Cohort Study to evaluate multiple aspects of the index and its components over 5 years of follow-up. The 937 participants in the subset had an a

PTUPB improves cognitive function in Alzheimer's disease associated with enhancing cerebral vascular myogenic response and attenuating vascular remodeling.

Genetic studies have linked EPHX2 (encoding soluble epoxide hydrolase, sEH) and PTGS2 (encoding cyclooxygenase-2, COX-2) to Alzheimer's disease (AD). Elevated levels of sEH and COX-2 found in AD patients and animals suggest their involvement in neurodegeneration, glial activation, vascular dysfunction, and inflammation. This study evaluated the effects of a new dual sEH/COX-2 inhibitor, PTUPB, on cerebrovascular function and cognition in TgF344-AD rats. The rats received oral PTUPB (2 mg/kg/day) for 25 days. Body weight, plasma glucose, and HbA1c levels remained stable between PTUPB- and vehic

Tracking the eyes to track decline: Global eye movement similarity as a novel neuropsychological marker of mild cognitive impairment.

INTRODUCTION: Across two studies, we investigated whether global eye movement similarity reflects visuospatial memory impairment in older adults, aiming to identify a novel neuropsychological marker of mild cognitive impairment (MCI). METHODS: In Study 1, 35 participants completed a global eye movement similarity memory (GEMS-M) task, viewing indoor scene images with three objects and identifying changes during the recognition phase. In Study 2, 60 participants (30 healthy controls, 30 with MCI) completed a passive version of the task without explicit memory judgment. RESULTS: Study 1 showed

Licochalcone a enhances cognitive resilience in APP/PS1 Mice by modulating glucose metabolism, Aβ burden, and neuroinflammation.

Alzheimer's disease (AD) is a complex neurodegenerative disorder. Current therapeutic approaches targeting a single pathway have shown limited efficacy, highlighting the need for multi-target interventions. Licochalcone A (LCA), a chalcone of licorice root, has demonstrated anti-inflammatory and antidiabetic properties. However, its potential neuroprotective mechanisms in AD remain unclear. The present study aims to elucidate the beneficial effect of LCA against cognitive decline in an AD mouse model. For this purpose, five-month-old APPswe/PS1dE9 (APP/PS1) mice received intraperitoneal LCA (1

Hypomorphic STING1/TMEM173 variants may support immuno-metabolic resilience in aging people living with HIV.

Antiretroviral therapy (ART) has significantly increased life expectancy of people living with HIV (PLWH). Nonetheless, despite effective virological control, PLWH are faced with accelerated aging, characterized by higher prevalence of age-related comorbidities than the general population. Persistent inflammation and activation of innate immunity seem to drive this immunosenescent phenotype. The cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING) pathway is increasingly recognized as a key player in the aging process and in age-related diseases, like cardiovascular and metabo

Autologous cytokine-induced NK cells as candidate cellular senolytics: evidence, obstacles, and the experiments still needed.

Cellular senescence and chronic low-grade inflammation occupy a central, mechanistically coupled position among the hallmarks of aging. Natural killer (NK) cells decline in per-cell cytotoxicity with age, and lower NK cell cytotoxicity has been associated with higher cancer incidence and infection-related mortality in older adults. NK cells are also endogenous effectors of senescent-cell clearance, engaging stress-induced NKG2D and DNAM-1 ligands and depending on perforin-mediated cytolysis. These literatures meet at a hypothesis: that autologous cytokine-induced NK cells, manufactured from a
