A salivary protein NlSP2 engages the OsCBSX3-SNAC3 module to enhance rice immunity.

The molecular dialogue between rice and the brown planthopper (BPH; Nilaparvata lugens) is mediated by insect salivary effectors, yet the molecular mechanisms underlying plant perception and immune activation remain largely unclear. Here, we characterized the BPH salivary protein NlSP2, which is secreted into rice tissues during feeding. Silencing of NlSP2 in BPH impairs salivary sheath formation, feeding ability, and survival, whereas its ectopic expression in rice enhances resistance to BPH via MAPK activation, H2O2 accumulation, and induction of defense-related genes. We identify the rice c

Silencing PsASH2 affects embryo development in the cotton mealybug.

Mealybugs are highly aggressive pests that infest various plants and cause substantial economic losses. Histone lysine methyltransferases (KMT) are evolutionarily conserved and proposed to be essential in early embryo development in animals. However, few KMTs have been reported in mealybugs. Here, we identified a novel KMT gene, PsASH2, in the cotton mealybug, Phenacoccus solenopsis Tinsley. This gene was highly expressed in the ovary of female adults. Through RNA interference (RNAi) of PsASH2 by dsRNA microinjection, we found a reduction in the number of male embryos and total embryos in the

Plant Virus-Induced Inheritable Apoptosis Drives Reproductive Costs in Female Insect Vectors to Balance Viral Biparental Transmission.

Insect-borne pathogens often reduce the reproductive fitness of insect vectors. Rice gall dwarf virus (RGDV) is biparentally transmitted to the offspring of its leafhopper vector. However, maternal transmission of RGDV decreases female fecundity and disrupts egg development via an unknown mechanism. This study reveals that RGDV induces mitochondria-dependent apoptosis in leafhopper ovaries, promoting viral infection but impairing ovary development. This apoptosis is transmitted to eggs during maternal RGDV transmission, promoting viral infection while harming eggs. RGDV in the ovary activates

Differentially spliced mitochondrial CYP419A1 contributes to ethiprole resistance in Nilaparvata lugens.

The brown planthopper Nilaparvata lugens is one of the most economically important pests of cultivated rice in Southeast Asia. Extensive use of insecticide treatments, such as imidacloprid, fipronil and ethiprole, has resulted in the emergence of multiple resistant strains of N. lugens. Previous investigation of the mechanisms of resistance to imidacloprid and ethiprole demonstrated that overexpression and qualitative changes in the cytochrome P450 gene CYP6ER1 lead to enhanced metabolic detoxification of these compounds. Here, we present the identification of a secondary mechanism enhancing e

Immunolocalization of Beet Curly Top Virus (BCTV) and GroEL Chaperon Protein of Endosymbionts in Beet Leafhopper (Circulifer tenellus) Vector Tissue.

Beet curly top virus (BCTV, curtovirus, geminiviridae) causes one of the most economically significant viral diseases in crops in the Western United States and is transmitted only by the beet leafhopper (Circulifer tenellus) in a non-propagative circulative manner. A better understanding of how this virus overcomes insect vector cellular barriers is essential to understanding virus-vector interactions. The distribution of BCTV in its beet leafhopper vector was investigated using immunofluorescence confocal laser scanning microscope analysis (iCLSM) on the whole-mount-dissected organs of leafho

Nutrient supplementation by genome-eroded Burkholderia symbionts of scale insects.

Hemipterans are known as hosts to bacterial or fungal symbionts that supplement their unbalanced diet with essential nutrients. Among them, scale insects (Coccomorpha) are characterized by a particularly large diversity of symbiotic systems. Here, using microscopic and genomic approaches, we functionally characterized the symbionts of two scale insects belonging to the Eriococcidae family, Acanthococcus aceris and Gossyparia spuria. These species host Burkholderia bacteria that are localized in the cytoplasm of the fat body cells. Metagenome sequencing revealed very similar and highly reduced …

Boron deficiency mediates plant-insect (Diaphorima citri) interaction by disturbing leaf volatile organic compounds and cell wall functions.

UNLABELLED: Nutritional enhancement has been reported to effectively relieve infected symptoms of Huanglongbing, one of the most destructive diseases of citrus. However, few studies focused on the role of plant nutrition in citrus plant-vector (Asian citrus psyllid; Diaphorina citri Kuwayama) interactions, which is regarded as an important part to develop an effective management strategy. METHOD: In the present study, a hydroponic culture was carried out to evaluate the effects of boron deficiency on psyllid feeding process to decode the molecular/biochemical basis of host-psyllid interaction

Insights into the synergistic mechanism of target resistance: A case study of N. lugens RDL-GABA receptors and fipronil.

It is known that a single mutation exerts moderate resistance to pesticide, while double mutations (DM) cause severe resistance problem through synergistic effect, and even result in failure application of pesticides. However, little is known about how double mutations would synergistically cause much high resistance level. In this work, computational studies were performed on the interaction of fipronil with N. lugens RDL-GABA receptors, to see how single and double mutations changed receptor structure properties and then conferred distinct resistance levels. The A2'S mutation displayed relat

Evidence for transovarial transmission of tomato yellow leaf curl virus by its vector, the whitefly Bemisia tabaci.

The whitefly Bemisia tabaci is the only vector of the tomato yellow leaf curl geminivirus (TYLCV). The insect transmits the virus in a persistent-circulative manner. TYLCV DNA was detected by polymerase chain reaction and by Southern blot hybridization in progeny (eggs, first and second instars, adults) of single viruliferous whiteflies that developed on eggplant or on cotton (two TYLCV nonhost plants). Furthermore, TYLCV DNA was present in the progeny of insects that had acquired the virus through the egg. The adult progeny of the viruliferous insects and their own progeny were able to infect

Attachment of Metarhizium anisopliae to the southern green stink bug Nezara viridula cuticle and fungistatic effect of cuticular lipids and aldehydes.

In this paper we examined the conidial attachment of Metarhizium anisopliae on the southern green stink bug, Nezara viridula, using the exuvia and nymphal stage of the host as a substrate for M. anisopliae conidiospores. Initial studies using fluorescein isothiocyanate (FITC)-labeled conidia examined the differential binding of conidia to various sites on the cuticle. Both the topography and the chemistry of the cuticle affected conidial adhesion. Conidia were trapped in areas containing large numbers of setae (e.g., antennal tips, apical portions of tibia and tarsi). Chemical treatments to re

[The sensory neuropile of the nerve chain of Eurygaster integriceps P].

Structure of the sensory neuropil of the abdominal nervous chain has been studied in the Eurygaster integriceps P. by means of methylene blue and paraldehyde fuchsin with phloxin staining. The sensory neuropil consists of three parts: 1) main sensory neuropil, including terminal, T-shaped, antero- and posterconnective fibers; 2) lateral sensory neuropils of the extremities, that in the insect studied, as in the dragon-fly larva, have the form of the overturned eight; 3) system of thick T-shaped fibers getting into the first thoracic ganglion and running along the second nerves, as well as alon

Mitochondrial Genomes Reveal Global Population Structure, Lineage Divergence, and Candidate Diagnostic Markers of Diaphorina citri.

The Asian citrus psyllid, Diaphorina citri Kuwayama (Hemiptera: Liviidae), is the primary vector of Candidatus Liberibacter asiaticus, the causal agent of huanglongbing, the most devastating disease of citrus worldwide. Understanding its population structure and developing lineage-specific markers are important for surveillance, quarantine and pest management. Here, 133 complete mitochondrial genomes were analyzed, comprising 77 previously published sequences, 13 newly assembled genomes and 43 metagenome-assembled mitogenomes. Phylogenetic analysis revealed two highly differentiated mitochondr…

Repellency and toxicity of long-lasting insecticide-treated bed nets (LLINs) to bed bugs.

Vector control is essential for eliminating malaria, a vector-borne parasitic disease responsible for over half a million deaths annually. Success of vector control programs hinges on community acceptance of products like long-lasting insecticide-treated nets (LLINs). Communities in malaria-endemic regions often link LLIN efficacy to their ability to control indoor pests such as bed bugs (Cimex lectularius L. and Cimex hemipterus (F.)) (Hemiptera: Cimicidae). Despite this, little is known about the potential repellent effects and toxicity of LLINs to bed bugs. Herein, we demonstrate for the fi…

No Interactions of Stacked Bt Maize with the Non-target Aphid Rhopalosiphum padi and the Spider Mite Tetranychus urticae.

In the agroecosystem, genetically engineered plants producing insecticidal Cry proteins from Bacillus thuringiensis (Bt) interact with non-target herbivores and other elements of the food web. Stacked Bt crops expose herbivores to multiple Cry proteins simultaneously. In this study, the direct interactions between SmartStax® Bt maize producing six different Cry proteins and two herbivores with different feeding modes were investigated. Feeding on leaves of Bt maize had no effects on development time, fecundity, or longevity of the aphid Rhopalosiphum padi (Hemiptera: Aphididae), and no effects…

High diversity in CMA3/DAPI-banding patterns in Heteropterans.

Heteroptera is the most numerous and diverse suborder of Hemiptera, with about 38,000 species. This diversity also involves cytogenetic features, including chromosome number and a sex determining system. Information about heterochromatin occurrence and distribution is scarce in heteropterans, but still, there is some evidence of variability. We determined the chromosome number and CMA3/DAPI-banding pattern of 179 individuals of 25 heteropteran species from Brazil. Eight species of Pentatomidae exhibited a constant chromosome number (2n = 12 + XY), but in Coreidae (12 species), Largidae (1 spec…

Silencing LsAtg8 reduces female fecundity and elevates Wolbachia density in embryos of the rice planthopper Laodelphax striatellus.

BACKGROUND: Autophagy is essential for insect reproduction and endosymbiont homeostasis, with Atg8 serving as a key autophagy-related gene. However, the specific role of LsAtg8 in reproduction and endosymbiont regulation in Laodelphax striatellus has not been fully examined. RESULTS: In this study, we discovered that LsAtg8 is expressed at higher levels in the fat body and ovaries of L. striatellus compared to other autophagy-related genes, regardless of endosymbiont Wolbachia infection status. Knockdown of LsAtg8 using RNA interference significantly reduced female fecundity and delayed oocyt

Single and Multiple Infestation by Coccoid Pests in Coffee Shape Plant Chemical Defenses and Interactions Across Trophic Levels.

Crop plants are attacked by multiple pests, inducing metabolic changes that shape interactions across trophic levels and influence the effectiveness of biological control. We examined how infestation by the coccoids Coccus viridis (scales) and Planococcus citri (mealybugs) modulates phytohormones, secondary metabolites, herbivore infestation, and behavior of the predator Cryptolaemus montrouzieri in Coffea arabica plants. Infestation by either scales or mealybugs activated the salicylate pathway but differentially regulated jasmonates and selected secondary metabolites. Scale infestation reduc

Microbiome-Epigenome Interplay Impacts Microbial Symbiosis and Stress Adaptations in the Brown Planthopper (Nilaparvata lugens).

The gut microbiota and epigenetic processes both contribute to insect survival and adaptation; however, the relationship between these two systems remains largely unexplored. In this study, we used the brown planthopper (Nilaparvata lugens; BPH) to explore microbiome-epigenome interactions and evaluate its impact on BPH survivability under environmental stress. Disruption of the gut microbiome using antibiotics significantly altered the epigenetic profile of various stress-responsive genes in the BPH. Similarly, perturbations in the epigenome induced by 5-azacytidine resulted in an altered mic

RNA interference targeting tyrosine hydroxylase and dopa decarboxylase inhibits the reproduction and survival in the small brown planthopper Laodelphax striatellus: A promising strategy for pest control.

The small brown planthopper Laodelphax striatellus is a serious insect vector in rice and causes substantial economic losses. Dopamine (DA) plays an important role in the multiple physiological and behavioral processes of insects, and tyrosine hydroxylase (TH) and dopa decarboxylase (DDC) are crucial enzymes in regulating DA synthesis. Although numerous studies have investigated the roles of TH and DDC in various insects, their functions in rice planthoppers remain poorly understood. Here, we cloned LsTH and LsDDC from L. striatellus. LsTH was predominantly expressed at the late stage of 3rd-5
