Potassium and magnesium balance are frequently altered by common pathological conditions. Isolated disturbances of potassium balance do not produce secondary abnormalities in magnesium homeostasis. In contrast, primary disturbances in magnesium balance, particularly magnesium depletion, produce secondary potassium depletion. This appears to result from an inability of the cell to maintain the normally high intracellular concentration of potassium, perhaps as a result of an increase in membrane permeability to potassium and/or inhibition of Na+-K-ATPase. As a result, the cells lose potassium, w…

Disorders of potassium homeostasis can be life-threatening. This is especially true in critically-ill patients, in whom concurrent disorders may exacerbate the adverse effects of both hyper- and hypokalemia. Alterations in potassium balance are usually multifunctional in origin. Four broad categories encompass the most common diseases and syndromes seen in the intensive care setting: pharmacologic agents, acid-base disturbances, hypomagnesemia, and renal insufficiency. The various influences that disturb the serum potassium concentration may either potentiate or counter-balance one another. Th…

A general model is proposed casting aspects of ontogeny in quantitative terms amenable to genetic analysis. Its primordial construct is a chain of cells (termed a "pursuer") growing under the influence of a signal towards a fixed structure termed a "target." There is provision for graduated correction of the direction of growth of the pursuer. The determinants of scale include the size of the cells and the distance from the target. The minimum number of parameters is two: the initial angle of growth; and the force of the correction of errors of direction. Both are potentially of genetic intere

A survey is given on disturbances in tissue homeostasis induced by hypovolemic shock conditions. Fluid shifts taking place between the extra- and intravascular fluid compartments are important early compensatory responses following hypovolemia. Usually the supply-to-demand ratio of oxygen in most tissues can however, not be kept up if the hypovolemic insult is severe due to deterioration of the microcirculation. Cellular hypoxia will ensue and may with time affect the integrity of the cells. Cellular functional disturbances occur earlier and are more pronounced in peripheral non-vital tissues …

Magnesium deficiency (MD) may be either latent: i.e. without disturbances in blood Mg, calcium and intracellular potassium (Ki) or patent, with a rich symptomatology: i.e. with hypomagnesemia, hypocalcemia and decreased Ki. These two aspects are also found with magnesium excess. The discrepancy between these two opposite clinical forms leads one to postulate the existence of a control of Mg homeostasis, efficient in latent forms and deficient in patent forms of Mg disturbances. 4 'endocrine glands', (i.e.) adrenal medulla, parathyroids, thyroid and beta-islets in the pancreas are involved main…

Antiserum specific for purified canine renal renin was used to inhibit this enzyme in trained, conscious dogs. The antiserum did not affect blood pressure in sodium-replete dogs but decreased plasma renin activity and blood pressure in sodium-depleted animals. The antiserum also reduced blood pressure to control levels concomitant with suppression of plasma renin activity in uninephrectomized dogs with acute renovascular hypertension. These observations establish the role of the renin-angiotensin system in the maintenance of blood pressure in the sodium-depleted state as well as in the initiat

The effects of phosphate depletion on magnesium (Mg) homeostasis were evaluated in rats fed a diet containing 0.03% phosphorus for periods up to 8 wk. Plasma phosphorus fell significantly (P < 0.01) from 10.1+/-0.27 (SE) to 5.0+/-0.54 mg/100 ml within 1 day and continued to fall gradually to a level of 1.2+/-0.21 mg/100 ml by the end of the 8th wk. A significant (P < 0.01) increment in urinary Mg excretion (UMgV) from 46+/-2.7 to 126+/-24 mueq/24 h occurred during the 1st day of phosphate depletion; UMgV reached a peak of 300+/-24 mueq/24 h by the 3rd day and remained high ranging between 150-…

Mutations in the innate immune receptor NOD2 are the greatest single genetic risk factor for Crohn's disease, yet the mechanisms by which NOD2 regulates intestinal homeostasis remain unclear. We used a CRISPR-generated zebrafish model to determine the impacts of NOD2 deficiency on intestinal health. In cellular, molecular, and transcriptomic studies, we uncovered substantial effects of NOD2 deficiency on epithelial and immune compartments, including deregulated expression of developmental pathways that establish and maintain the gut epithelium, and an unexpected increase in the expression of m…

BACKGROUND: Synthetic polymers are widely used in medical and dental appliances, including clear aligners, retainers, splints, and sleep devices. Despite their widespread use, little is known about the release of microplastics from traditional thermoformed polymers compared with recently developed direct-printed photopolymers. Plastic particles have been detected in a variety of human tissues such as, lung, liver, spleen and kidneys. Each of these sites contain local resident macrophages tasked with maintaining homeostasis by clearing cell debris and foreign bodies. Like these organs, the oral…

Osteoporosis (OP) is primarily characterized by reduced bone mass, microarchitectural deterioration, and an increased susceptibility to frag … and mineralization-associated calcium signaling. Emerging evidence indicates that disruption of ER homeostasis regulates osteogenic differentiation through the three canonical branches of the unfolded protein response (UPR), including PERK-eIF2α-ATF4, IRE1α-XBP1, and ATF6 signaling pathways. In parallel, selective ER autophagy (ER-phagy) dynamically regulate …

Cohen syndrome (CS) is a rare autosomal recessive neurodevelopmental disorder characterized by intellectual disability, microcephaly, retinal dystrophy, and neutropenia. We previously demonstrated that VPS13B mediates phosphatidylinositol 4-phosphate (PI4P) transport to promote mitochondrial fission. Here, we identify VPS13B as a regulator of lysosomal homeostasis. VPS13B knockout (KO) HeLa cells exhibited aberrant lysosomal distribution and reduction in LAMP1-positive lysosomes. Bulk RNA sequencing revealed coordinated downregulation of lysosome-related genes, including genes required for aci…

Mitochondrial regulation in mesenchymal stem cells (MSCs) serves as a critical determinant of bone formation and skeletal homeostasis. While dietary nitrate and its transporter Sialin are implicated in systemic homeostasis, their specific roles in MSCs' function remain unclear. Here, we demonstrate that Sialin deficiency impairs MSCs' function and disrupts bone homeostasis. Gain- and loss-of-function studies reveal that Sialin localizes to the mitochondrial membrane and promotes osteogenic differentiation by maintaining mitochondrial bioenergetic integrity. Mechanistically, Sialin recruits pST…

The Hippo signaling pathway, a highly conserved signaling cascade from Drosophila to mammals, plays a critical role in mammals in regulating cell proliferation, senescence and apoptosis. In the pancreas, this pathway has emerged as a key regulator in various processes, including development, cell fate determination, and regeneration following injury. Consequently, aberrant the Hippo signaling pathway disrupts pancreatic tissue homeostasis, leading to uncontrolled cell proliferation, oncogenic transformation, and the subsequent development of pancreatic diseases. In this review, we comprehensiv…

Immune alterations, such as neutrophil dysfunction, significantly affect the progression and outcome of periodontitis, a prevalent inflammat … of neutrophils. These findings underscore the critical role of the CD300lf/MyD88 axis in neutrophil homeostasis and suggest that modulation of CD300lf through ceramide presents a promising therapeutic strategy for periodontitis.
