Parasite interactions in natural populations: insights from longitudinal data.

The physiological and immunological state of an animal can be influenced by current infections and infection history. Consequently, both ongoing and previous infections can affect host susceptibility to another parasite, the biology of the subsequent infection (e.g. infection length) and the impact of infection on host morbidity (pathology). In natural populations, most animals will be infected by a succession of different parasites throughout the course of their lives, with probably frequent concomitant infections. The relative timing of different infections experienced by a host (i.e. the se

Mast Cell Response to Parasites: from Recognition and Activation to Host Defense Modulation.

Parasites represent a diverse and widely distributed group of pathogens that cause diseases with significant global health implications. The interaction between parasite and host is characterized by a high degree of complexity, with both parties continuously adapting to changes in the other. The successful host invasion is largely attributable to the evasion strategies employed by parasites to ensure their survival in immunocompetent individuals. In turn, the host's defense mechanisms utilize a variety of structures and processes, ranging from primary barriers to the most sophisticated ones, t

Protozoan parasites in group-living primates: testing the biological island hypothesis.

A series of articles by W.J. Freeland published in the 1970s proposed that social organization and behavioral processes were heavily influenced by parasitic infections, which led to a number of intriguing hypotheses concerning how natural selection might act on social factors because of the benefits of avoiding parasite infections. For example, Freeland [1979] showed that all individuals within a given group harbored identical gastrointestinal protozoan faunas, which led him to postulate that social groups were akin to "biological islands" and suggest how this isolation could select specific t

Sialylated N-glycans mediate monocyte uptake of extracellular vesicles secreted from Plasmodium falciparum-infected red blood cells.

Glycoconjugates on extracellular vesicles (EVs) play a vital role in internalization and mediate interaction as well as regulation of the host immune system by viruses, bacteria, and parasites. During their intraerythrocytic life-cycle stages, malaria parasites, Plasmodium falciparum (Pf) mediate the secretion of EVs by infected red blood cells (RBCs) that carry a diverse range of parasitic and host-derived molecules. These molecules facilitate parasite-parasite and parasite-host interactions to ensure parasite survival. To date, the number of identified Pf genes associated with glycan synthes

Distinct transcriptomic signatures define febrile malaria depending on initial infective states, asymptomatic or uninfected.

BACKGROUND: Cumulative malaria parasite exposure in endemic regions often results in the acquisition of partial immunity and asymptomatic infections. There is limited information on how host-parasite interactions mediate the maintenance of chronic symptomless infections that sustain malaria transmission. METHODS: Here, we determined the gene expression profiles of the parasite population and the corresponding host peripheral blood mononuclear cells (PBMCs) from 21 children (< 15 years). We compared children who were defined as uninfected, asymptomatic and those with febrile malaria. RESULTS:…

The Biology of Malaria Parasite Liver Infection.

The Plasmodium liver stages of infection-initiated when mosquito-transmitted sporozoites invade hepatocytes-are asymptomatic and essential for initial parasite replication and progression to blood stage infection. This paper provides an overview of liver stage biology, from sporozoite invasion mechanisms and host hepatocyte remodeling to intracellular development, nutrient acquisition, and evasion of host defenses. We highlight recent insights into cellular and molecular host-parasite interactions, and innate immune pathways that mediate liver stage clearance. The paper also reviews the evolut…

Glycan remodelling of live adult Schistosoma mansoni worms by chemical inhibition of α-mannosidases.

Schistosomiasis is a neglected tropical disease affecting over 250 million people worldwide. Schistosoma parasites can survive in the human host for years due to their capability to evade and modulate host immune responses. Glycans and glycoproteins produced by schistosomes are thought to play an important role in shaping parasite-host interactions. In addition, N-linked glycans are vital post-translational protein modifications involved in fundamental cellular and developmental processes like protein folding and cell-cell interactions. In this study, we generated live Schistosoma mansoni adul

Virulence in the three-spined stickleback specific parasite Schistocephalus solidus is inherited additively.

Parasite virulence is a key trait in host-parasite interactions and plays a crucial role in infection dynamics. Our study system offers the rare opportunity to study the virulence of an individual macroparasite (Schistocephalus solidus) in its vertebrate fish host (Gasterosteus aculeatus). The size of the tapeworm in the fish can be regarded as a good proxy for individual parasite virulence, as parasite size correlates negatively with fitness traits of the stickleback host (i.e. the bigger the parasite, the lower the host's reproductive success) as well as directly with the number of parasite …

Interactions between bacterial microbiota and protozoan parasites: an intimate relationship.

Interactions between parasites and the gut microbiota play a key role in immune responses and susceptibility to zoonotic diseases; however, many aspects of how protozoan infections alter microbial diversity and how these changes influence parasite pathogenicity and host defense remain poorly understood. This review integrates recenst studies using murine models of the Muroidea superfamily, highlighting patterns in microbial diversity, functional interactions, and potential immunometabolic mechanisms underlying these relationships. Both consistent findings and discrepancies among studies are id

T cell exhaustion in parasitic infections.

T cell exhaustion is a state of progressive functional impairment that occurs upon persistent antigenic stimulation and is a key mechanism by which chronic parasitic infections evade host immunity. As research into T cell exhaustion continues to deepen, immunotherapies targeting exhausted T cells have emerged as a prominent area of interest, particularly in cancers and chronic viral infections. However, T cell exhaustion caused by parasitic infections exhibits significant differences from classic T cell exhaustion observed in chronic viral infections, and understanding how parasites with their

A biological invasion modifies the dynamics of a host-parasite arms race.

By imposing novel selection pressures on both participants, biological invasions can modify evolutionary 'arms races' between hosts and parasites. A spatially replicated cross-infection experiment reveals strong spatial divergence in the ability of lungworms (Rhabdias pseudosphaerocephala) to infect invasive cane toads (Rhinella marina) in Australia. In areas colonized for longer than 20 years, toads are more resistant to infection by local strains of parasites than by allopatric strains. The situation reverses at the invasion front, where super-infective parasites have evolved. Invasion-induc

Disentangling specific and unspecific components of innate immune memory in a copepod-tapeworm system.

Evidence that the innate immune system can respond with forms of memory upon reinfection has been accumulating over the past few years. These phenomena of "immune priming" in invertebrates, and "trained immunity" in vertebrates, are contrary to previous belief that immune memory and specificity are restricted to the adaptive immune system. However, while trained immunity is usually a response with rather low specificity, immune priming has shown highly specific responses in certain species. To date, it is largely unknown how specificity in innate immune memory can be achieved in response to di

Selection on an extreme-yet-conserved larval life-history strategy in a tapeworm.

Evolutionary stasis characterizes many phenotypes, even ones that seem suboptimal. Among tapeworms, Schistocephalus solidus and its relatives have some of the shortest developmental times in their first intermediate hosts, yet their development still seems excessively long considering they can grow faster, larger, and safer in the next hosts in their complex life cycles. I conducted 4 generations of selection on the developmental rate of S. solidus in its copepod first host, pushing a conserved-but-counterintuitive phenotype toward the limit of known tapeworm life-history strategies. Faster pa

The interplay of helminthic neuropeptides and proteases in parasite survival and host immunomodulation.

Neuropeptides comprise a diverse and broad group of neurotransmitters in vertebrates and invertebrates, with critical roles in neuronal signal transduction. While their role in controlling learning and memory in the brains of mammals is known, their extra-synaptic function in infection and inflammation with effects on distinct tissues and immune cells is increasingly recognized. Helminth infections especially of the central nervous system (CNS), such as neurocysticercosis, induce neuropeptide production by both host and helminth, but their role in host-parasite interplay or host inflammatory r
