Disease incidence is related to bleaching extent in reef-building corals.

Recent outbreaks of coral bleaching and disease have contributed to substantial declines in the abundance of reef-building coral. Significant attention has been paid to both phenomena in order to determine their effect on reef trajectories. Although each is positively correlated with high temperatures, few studies have explored the potential links between bleaching and disease. A longitudinal study of corals in the Florida Keys was therefore conducted during the 2005 Caribbean bleaching event to quantify bleaching extent and disease incidence in corals, and to determine whether they were relat

Co-evolution and exploitation of host cell signaling pathways by bacterial pathogens.

Bacterial pathogens have evolved by combinations of gene acquisition, deletion, and modification, which increases their fitness. Additionally, bacteria are able to evolve in "quantum leaps" via the ability to promiscuously acquire new genes. Many bacterial pathogens - especially Gram-negative enteric pathogens - have evolved mechanisms by which to subvert signal transduction pathways of eukaryotic cells by expressing genes that mimic or regulate host protein factors involved in a variety of signaling cascades. This results in the ability to cause diseases ranging from tumor formation in plants

Fire blight: applied genomic insights of the pathogen and host.

The enterobacterial phytopathogen Erwinia amylovora causes fire blight, an invasive disease that threatens a wide range of commercial and ornamental Rosaceae host plants. The response elicited by E. amylovora in its host during disease development is similar to the hypersensitive reaction that typically leads to resistance in an incompatible host-pathogen interaction, yet no gene-for-gene resistance has been described for this host-pathogen system. Comparative genomic analysis has found an unprecedented degree of genetic uniformity among strains of E. amylovora, suggesting that the pathogen ha

Host-Candida auris interactions in the skin.

Candida auris is an emerging, multidrug-resistant fungal pathogen that causes healthcare-associated outbreaks and life-threatening systemic infections. Unlike other Candida species, C. auris exhibits a distinct capacity for persistent skin colonization. In this review, we summarize our current understanding of clinical risk factors and host-microbe interactions that underlie C. auris skin colonization and infection. We discuss fungal determinants, including the unique mannan outer layer, fungal adhesins, the protein kinase Hog1, and other pathways in C. auris that govern adaptation in the skin

Serum lipidome remodeling in viral pneumonia: from pathophysiology to therapeutics.

Viral pneumonia remains a preeminent global health threat, frequently culminating in acute respiratory distress syndrome (ARDS) and systemic organ failure. While traditional paradigms have centered on protein-based inflammatory cascades, this review synthesizes a vast body of emerging evidence to redefine viral pneumonia as a profound systemic metabolic crisis, specifically characterized by the radical remodeling of the host lipidome. Based on a meta-synthesis of clinical and mechanistic data, we delineate a consistent "metabolic crash" during severe infection, where a precipitous decline in s

Xanthomonas effectors: nutritional hijacking for a long running battle.

Xanthomonas spp. are devastating plant pathogens that cause significant agricultural losses worldwide. Their virulence relies on type III secretion system (T3SS) effectors that manipulate host cells, but the functions of many effectors remain unknown. A recent report demonstrated that the AvrBs2 effector from the rice pathogen Xanthomonas oryzae pv. oryzicola manipulates host metabolism to synthesize a unique metabolite, xanthosan, from host sugars. The bacteria then import and utilize this metabolite via specific transporters. Notably, transgenic rice plants engineered to degrade this metabol

Anti-phage defense systems in bacteria: molecular mechanisms and their role in shaping phage therapy strategies.

Bacteria and phages are engaged in a persistent evolutionary struggle. To survive constant phage predation, bacteria have evolved a highly diverse and multi-layered immune arsenal that determines the success of therapeutic phage infection. Bacterial defenses include receptor blockade, DNA restriction systems, CRISPR-Cas adaptive immunity, and secondary messenger signaling systems that induce effector-mediated cell death. Recent mechanistic advances have elucidated systems such as Thoeris (gcADPR-activated SIR2 effectors depleting NAD⁺), CBASS (cyclic nucleotide-activated effectors disrupting c

Differential roles of three subclass III C6 transcription factors in the insect-pathogenic lifecycle of Beauveria bassiana.

Six subclass III C6 transcription factors (TFs) with unique Zn(II)2Cys6 binuclear cluster DNA-binding (C6 or GAL4) domain not locating at N-terminus are underexplored in Beauveria bassiana, a wide-spectrum mycoinsecticide. Here, we report that three C6 TFs with the GAL4 domain locating at C-terminus (TF1), between C-terminal and central regions (TF2) and between central and N-terminal regions (TF3) are largely differentiated in function. TF1 was proven to localize in both nucleus and cytoplasm and regulate host immune repression in the course of host hemocoel colonization instead of cuticular

Small impact of the host plant physiological state on the genome formula of a nanovirus.

The genome formula refers to the specific and reproducible pattern of relative accumulation of genome segments in within-host populations of viruses with segmented genomes. It has been proposed that the genome formula may have an adaptive function by allowing rapid adaptation of viral genes' expression, through variation in their relative copy number, in response to a changing environment. Consistently, the faba bean necrotic stunt virus (FBNSV) genome formula has been demonstrated to be strongly dependent on the host genotype, since it changes significantly when the same viral isolate moves f

Autophagy in emerging and highly concerned severe zoonotic infectious diseases.

Autophagy is a conserved cellular process that mediates degradation of damaged organelles, misfolded proteins, and invading pathogens, playing critical roles in intracellular homeostasis and immune regulation. Given that over 70% of infectious diseases and 60% of emerging infectious diseases are zoonotic, posing a major threat to global health, this review aims to summarize the cellular and molecular mechanisms underlying the crosstalk between autophagy and key zoonotic pathogens. We comprehensively retrieved relevant research literature from the PubMed, Web of Science, and Scopus databases (w

Morphophysiological disorders and metabolic reprogramming in Physalis peruviana infected with the physalis rugose mosaic virus.

Physalis peruviana L. (Cape gooseberry) is susceptible to several pathogens, among which the sobemovirus physalis rugose mosaic virus (PhyRMV) is most prominent. This study investigated the anatomical, morphophysiological, and metabolic responses of P. peruviana to PhyRMV over the course of the infection cycle. P. peruviana plants were grown under greenhouse conditions (24 ± 2 ℃) and mechanically inoculated with buffer (mock) or PhyRMV inoculum. The local and systemic leaves were collected at 0, 3, 7, 14, 21, and 42 days after inoculation (DAI) and viral infection was confirmed by RT-qPCR. Mor

Study of Lawsonia intracellularis interactions with cell culture mitochondria.

Lawsonia intracellularis is an important obligate intracellular bacterium. It localizes in close proximity to mitochondria within host cells. Mitochondria regulate diverse physiological processes in host cells, thereby significantly affecting the intracellular proliferation of bacteria. However, how L. intracellularis interacts with mitochondria to promote its own proliferation remains unclear. In this study, the mitochondrial morphology of McCoy cells infected with L. intracellularis was examined by indirect immunofluorescence assay (IFA), and L. intracellularis infection was found to induce

Contributions of intestinal protists on the human gut landscape through the lens of Entamoeba spp.

Intestinal protists represent an underappreciated yet functionally significant component of the human gut microbiome. Historically dismissed as parasites or transient contaminants, many of these microbial eukaryotes, particularly Entamoeba spp., are now recognized as integral to gut ecosystem function and host immune homeostasis. This review examines the complex roles of Entamoeba spp. in the mammalian gut, positioning them as dynamic microbiome "landscapers" that influence host-pathogen interactions, immune tone, and microbial diversity. We explore the evolutionary adaptation of Entamoeba to …

Arbovirus Infections and Epigenetic Mechanisms; a Potential Therapeutic Target.

Arboviruses are a group of arthropod-borne viral pathogens that pose a significant threat to the public health system. The clinical manifestations associated with these viruses range from self-limiting infections to life-threatening disorders. As a group of systemic viral infections, arboviruses can affect various parts of human organ systems, such as the nervous system. In the nervous system, epigenetic mechanisms are involved in various mechanisms including adult neurogenesis, neuronal-glial differentiation, the regulation of neural behaviour and neural plasticity, as well as other brain fun

Chemosensory protein 3 is a brain host factor for the induction of enhanced-locomotory activity in the BmNPV-silkworm infection model.

Although it is reported that the protein tyrosine phosphatase gene of baculovirus (group I nucleopolyhedrovirus) can induce enhanced locomotory activity (ELA) in caterpillars, our understanding of the host factors that are involved in the regulation of the behavioral change is still limited. Previously, single-nucleus RNA sequencing (snRNA-seq) was used to identify 19 distinct clusters representing Kenyon cell, glial cell, olfactory projection neuron, optic lobes neuron, hemocyte, muscle cell types and other unannotated cells in the silkworm larvae brains. Immunity responses as checkpoints for efficient transmission of begomoviruses by whiteflies.

Begomoviruses are a group of single stranded DNA plant viruses exclusively transmitted by the sweet potato whitefly Bemisia tabaci in a persistent, circulative manner. After acquisition from plant phloem, this group of viruses circulate and are retained within the whitefly, interacting with tissues, cells and molecular pathways for maintaining the safety of the infective intact virions, by exploiting cellular mechanisms and avoiding degradation by the insect immune responses. During retention, the virions are internalized in the midgut cells, exit and spend hours-days in the hemolymph and cros

Pathogenicity study of ascomycetous opportunistic yeasts from fresh feces of pigeons using Tenebrio molitor larvae as a host model.

The choice of model to assess host-pathogen interactions is crucial for the analysis of microbial pathogenicity. Insects are increasingly chosen as model hosts for the study of microbial infections. Although the immune system responses of insects are not specific, they have a complex, evolutionarily conserved innate immune system with cellular and humoral elements. In this study, we investigated in vivo the virulence of potentially pathogenic ascomycetous yeasts found in fresh feces of street pigeons using the larval host model of Tenebrio molitor. The results were arranged in survival curves …

Multi-omics analysis reveals the pathobiome-host interactions in the bleaching disease of the seaweed Saccharina japonica.

BACKGROUND: The emerging concept of the pathobiome has revolutionized our understanding of disease etiology by emphasizing the complex interactions between multiple pathogens and their hosts during disease progression. Although significant advancements have been made in characterizing the pathobiome in human, animal, and plant diseases, the pathobiome associated with seaweed diseases remains unexplored. Saccharina japonica, a commercially important farmed seaweed, has increasingly suffered from bleaching disease during its nursery stage, severely compromising the supply of healthy sporelings a
