Association Between Abdominal Obesity and Hyperuricemia in Peruvian Adults with Type 2 Diabetes Mellitus.

BACKGROUND: Abdominal obesity (AO) is associated with hyperuricemia (HU) in people with type 2 diabetes (T2DM), but most evidence comes from Asian studies and Latin American data are scarce. This study addresses that gap in a Peruvian setting. We aimed to evaluate the association between AO and HU in adult Peruvians diagnosed with T2DM. METHODS: We carried out a cross-sectional observational study in adults with T2DM in a private clinic in Lima - Peru. We defined HU as serum uric acid ⩾ 6 mg/dL and defined AO as waist circumference ⩾ 94 cm in men and ⩾88 cm in women. In addition, we estimated…

Hyperuricemia impairs endothelial function through SMS2-dependent activation of the endoplasmic reticulum stress response.

Hyperuricemia is a recognized risk factor for cardiovascular diseases, including peripheral arterial disease (PAD), though molecular mechanisms linking uric acid to endothelial dysfunction remain unclear. This study investigates the role of sphingomyelin synthase 2 (SMS2) and endoplasmic reticulum (ER) stress in uric acid-induced endothelial impairment. Human umbilical vein endothelial cells were exposed to physiologically relevant concentrations of uric acid, with SMS2 function modulated through siRNA knockdown and ER stress inhibited using 4-phenylbutyric acid. Uric acid exhibited concentrat…

[Anti-hyperuricemia activity and its mechanism of flavonoid extract from saffron floral bio-residues].

A hyperuricemic rat model induced by adenine and ethambutol was established to investigate the anti-hyperuricemia activity and its mechanism of the flavonoid extract from saffron floral bio-residues. Sixty-seven SD rats were randomly divided into control group, model group, positive control group, and flavonoid extract groups(with 3 doses), respectively, and each group contained 11 or 12 rats. The hyperuricemic model was established by continuous oral administration of adenine(100 mg·kg~(-1)) and ethambutol(250 mg·kg~(-1)) for 7 days. At the same time, the positive control group was given allo…

Comparison of different insulin resistance surrogates to predict hyperuricemia among U.S. non-diabetic adults.

PURPOSE: Although it has been well-acknowledged that insulin resistance (IR) plays a critical role in the development of hyperuricemia (HU), specific relationship between IR and HU in non-diabetic patients remains rarely studied, and there is still no large-scale research regarding this issue. This study aims to explore the association between triglyceride glucose (TyG), TyG with body mass index (TyG-BMI), the ratio of triglycerides divided by high-density lipoprotein cholesterol (TG/HDL-C), metabolic score for insulin resistance (METS-IR), and the risk of HU in non-diabetic patients in The Un…

Longitudinal associations between metabolic score for visceral fat and hyperuricemia in non-obese adults.

BACKGROUND AND AIMS: The assessment of visceral adiposity is of great significance for the prevention of hyperuricemia (HUA), especially in non-obese individuals. The metabolic score for visceral fat (METS-VF) is a newly proposed surrogate of visceral obesity. We aimed to evaluate the longitudinal associations of METS-VF with the risk of HUA in non-obese adults. METHODS AND RESULTS: A total of 16,058 non-obese adults without HUA were included for this retrospective cohort analyses. The crude incidence rate of HUA in non-obese women and men were 20.9 and 69.6 per 1000 person-years, respectivel…

High-Protein Diet Induces Hyperuricemia in a New Animal Model for Studying Human Gout.

Hyperuricemia is a central risk factor for gout and increases the risk for other chronic diseases, including cardiometabolic disease, kidney disease, and hypertension. Overproduction of urate is one of the main reasons for hyperuricemia, and dietary factors including seafoods, meats, and drinking are contributed to the development of it. However, the lack of a suitable animal model for urate metabolism is one of the main reasons for the delay and limitations of hyperuricemia research. Combining evolutionary biological studies and clinical studies, we conclude that chicken is a preferred animal…

HPRT-related hyperuricemia with a novel p.V35M mutation in HPRT1 presenting familial juvenile gout.

Unlike complete deficiency of hypoxanthine phosphoribosyltransferase (HPRT) (i.e., Lesch-Nyhan syndrome), partial HPRT deficiency causes HPRT-related hyperuricemia without neurological symptoms. Herein, we describe a 22-year-old man without neurological symptoms that presented gout, hyperuricemia (serum urate level, 12.2 mg/dL), multiple renal microcalculi, and a family history of juvenile gout that was exhibited by his brother and grandfather. Genetic testing revealed a novel missense mutation, c.103G>A (p.V35M), in the HPRT1 gene, and biochemical testing (conducted using the patient's erythr…

New Rice-Derived Short Peptide Potently Alleviated Hyperuricemia Induced by Potassium Oxonate in Rats.

Gout that caused by hyperuricemia affects human health seriously and more efficient drugs are urgently required clinically. In this study, a novel peptide named RDP1 (AAAAGAKAR, 785.91 Da) was identified from the extract of shelled fruits of Oryza sativa. Our results demonstrated that RDP1 (the minimum effective concentration is 10 μg/kg) could significantly reduce the serum uric acid and creatinine and alleviate hyperuricemic nephropathy in rats by intragastric administration. RDP1 inhibited xanthine oxidase, which also was verified at the animal level. Results from molecular docking indicate…

Cichorium intybus L. promotes intestinal uric acid excretion by modulating ABCG2 in experimental hyperuricemia.

BACKGROUND: This paper describes a protocol for determining the incidence of serious fall injuries for Strategies to Reduce Injuries and Develop Confidence in Elders (STRIDE), a large, multicenter pragmatic clinical trial with limited resources for event adjudication. We describe how administrative data (from participating health systems and Medicare claims) can be used to confirm participant-reported events, with more time- and resource-intensive full-text medical record data used only on an "as-needed" basis. METHODS: STRIDE is a pragmatic cluster-randomized controlled trial involving 5451

Mild hyperuricemia induces vasoconstriction and maintains glomerular hypertension in normal and remnant kidney rats.

BACKGROUND: Hyperuricemia has been associated with renal disease. Because glomerular hemodynamic alterations critically contribute to initiation and progression of renal disease, we evaluated the effect of mild hyperuricemia in glomerular microcirculatory changes in rats under normal conditions and with renal injury induced by subtotal renal ablation (RK). METHODS: Hyperuricemia was induced in normal and remnant kidney (RK) rats on a normal sodium diet by administration of oxonic acid (OA). To prevent hyperuricemia, allopurinol (AP) was administered concomitantly. Glomerular hemodynamics were…

Association between hyperuricemia and kidney stones in Southern China: a multicentre cross-sectional study.

BACKGROUND: Hyperuricemia has been identified as a significant independent risk factor for kidney stones. However, a paucity of research has been conducted on the correlation between hyperuricemia in the general population and the prevalence of kidney stones. Southern China has a high incidence of kidney stones, and analysis of data from health check-ups can help to identify those at risk of developing kidney stones. This is of positive clinical significance for the prevention of kidney stones in hyperuricemia populations. METHODS: A multicentre cross-sectional study was conducted using data …
