The female reproductive system is strongly influenced by nutrition and energy balance. It is well known that food restriction or energy depletion can induce suppression of reproductive processes, while overnutrition is associated with reproductive dysfunction. However, the intricate mechanisms through which nutritional inputs and metabolic health are integrated into the coordination of reproduction are still being defined. In this review, we describe evidence for essential contributions by hormones that are responsive to food intake or fuel stores. Key metabolic hormones-including insulin, the

Carboxypeptidase H (CPH) is a peptide-processing enzyme thought to be involved in the synthesis of many neuropeptides, including vasopressin (VP) and oxytocin (OT). In this study, employing in situ hybridization histochemistry, we have shown that CPH mRNA is abundantly expressed in the magnocellular paraventricular and supraoptic nuclei of the hypothalamus, the primary sites of OT and VP synthesis. Since this enzyme is copackaged in secretory vesicles and hence coreleased with the neurohypophysial hormones, enzyme stores are depleted in parallel with the peptide hormones during states of hyper

Hypodipsic hypernatremia developed in association with hypopituitarism and hydrocephalus in a 7-month-old cat. Initial clinical signs (generalized weakness, cervical ventroflexion) were related to a hypernatremia-induced polymyopathy. Forced water intake and dietary sodium restriction corrected the hypernatremia and signs of muscle dysfunction. After restoration of eunatremia, secretion of pituitary hormones normalized. It was speculated that hypothalamic dysfunction, possibly related to hydrocephalus, induced both hypodipsia and transient hypopituitarism.

To specify interrelations between clinical and hormonal aspects of reproductive dysfunctions appearing during therapy with atypical antipsychotic agents (clozapine, olanzapine, quetiapine), 131 female patients with paranoid schizophrenia and schizoaffective disorder have been studied. Distinctive features of dynamics of prolactin as well as hypothalamic-pituitary-gonadal hormones levels for each medication studied and correlations between hormone indices and clinical presentations of reproductive dysfunctions (galactorrhea, disturbances of menstrual cycle and libido) were found.

Authors present a case of 28-year old female with anterior hypopituitarism and diabetes insipidus, with properly functioning anterior pituitary cells as showed by means of measuring pituitary hormones in response to neurohormones i.v. injections. Magnetic resonance imaging revealed neoplastic tissue in the pituitary stalk destroying supraopticohypophysial and paraventriculohypophysial tracts, as well as portal blood system, thus preventing release of vasopressin and these hypothalamic neurohormones from accessing anterior pituitary.

Growth hormone deficiency can be congenital or acquired. While acquired cases are related to organic causes that affect the hypothalamic-pituitary axis, structural abnormalities may or may not be detected in congenital cases. Hence, brain magnetic resonance imaging (MRI) is paramount in this context since it reflects diverse pathological processes with prognostic importance. This article will discuss the neuroradiologist's perspective on pediatric growth hormone deficiency assessment. The most common hypothalamic-pituitary findings will be explored, based on a brief overview of the pituitary d

The insulin tolerance test is considered the gold standard for assessing the hypothalamic-pituitary-adrenal and growth hormone (GH) axes, but its use varies considerably among different endocrine units. We recommend using the insulin tolerance test to assess the hypothalamic-pituitary-adrenal axis within 3 months of pituitary surgery, where adrenocorticotropic hormone 1-24 testing is equivocal, and to assess for GH deficiency where the patient is being considered for GH replacement therapy. We also discuss safety issues, how to ensure adequate hypoglycaemia and possible alternative tests, such

The last few years have seen the isolation, characterization and synthesis of two new hypothalamic peptides, the growth-hormone releasing factor (GRF) and the corticotrophin-releasing factor (CRF). GRF selectively stimulates pituitary growth hormone. It is interesting to note that GRF was isolated from two pancreatic tumors which were responsible for an acromegalic condition in the two patients. Hypothalamic GRF (1-44) has been found to be identical to this pancreatic GRF. CRF was isolated first from ovine hypothalamus and later characterized in several species and in the human. CRF specifical

Between 1980 and 1989, five infants (four males and one female) with congenital hypopituitarism were born within a relatively sparsely populated area of central Massachusetts, an incidence 25-fold greater than expected. In all infants the pituitary stalk was undetectable on magnetic resonance imaging, and a bright spot on T-1 weighted imaging, probably representing posterior pituitary tissue, was found ectopically located inferior to the median eminence. The height of the anterior pituitary remnant on magnetic resonance imaging varied from undetectable to 4 mm, and similarly, on hormonal testi

Gonadotropin-inhibitory hormone (GNIH) was discovered in quail with the ability to reduce gonadotropin expression/secretion in the pituitary. There have been few studies on GNIH orthologs in teleosts (LPXRFamide (Lpxrfa) peptides), which have provided inconsistent results. Therefore, the goal of this study was to determine the roles and modes of action by which Lpxrfa exerts its functions in the brain-pituitary axis of zebrafish (Danio rerio). We localized Lpxrfa soma to the ventral hypothalamus, with fibers extending throughout the brain and to the pituitary. In the preoptic area, Lpxrfa fibe

Circulating resistin stimulates endogenous glucose production (GP). Here, we report that bi-directional changes in hypothalamic resistin action have dramatic effects on GP and proinflammatory cytokine expression in the liver. The infusion of either resistin or an active cysteine mutant in the third cerebral ventricle (icv) or in the mediobasal hypothalamus stimulated GP independent of changes in circulating levels of glucoregulatory hormones. Conversely, central antagonism of resistin action markedly diminished the ability of circulating resistin to enhance GP. We also report that centrally me

OBJECTIVE: Hypothalamic dysfunction is a rare condition and can be encountered in patients who have been diagnosed or treated for a suprasellar brain tumor. Due to its rarity, the signs and symptoms of hypothalamic dysfunction may be difficult to recognize, leading to delayed diagnosis of the suprasellar brain tumor or to difficulties in finding the health-care expertise for hypothalamic dysfunction after tumor treatment. To improve the care and outcome of patients with acquired hypothalamic dysfunction, professionals are required to understand the patient's needs.
DESIGN: A worldwide online

BACKGROUND: Autism is a complex, heterogeneous, behaviorally-defined disorder characterized by disruptions of the nervous system and of other systems such as the pituitary-hypothalamic axis. In a previous genome wide screen, we reported linkage of autism with a 1.2 Megabase interval on chromosome 5q31. For the current study, we hypothesized that 3 of the genes in this region could be involved in the development of autism: 1) paired-like homeodomain transcription factor 1 (PITX1), which is a key regulator of hormones within the pituitary-hypothalamic axis, 2) neurogenin 1, a transcription facto

Obesity is one of the most common metabolic diseases and the greatest threats of the health because of possibility of numerous complications. In order to design effective drugs or apply the helpful surgical procedure it is essential to understand physiology of appetite control and pathophysiology of obesity. According to the first law of thermodynamics, the energy input in the form of food, equals energy expenditure through exercise, basal metabolism, thermogenesis and fat biosynthesis. The control of body weight actually concerns the control of adipose tissue with the key role of hypothalamus

Growth hormone deficiency affects roughly between one in 3000 and one in 4000 children with most instances of growth hormone deficiency being idiopathic. Growth hormone deficiency can also be associated with genetic diseases or chromosome abnormalities. Association of growth hormone deficiency together with hypothalamic-pituitary axis malformation and Cat-Eye syndrome is a very rare condition. We report a family with two brothers presenting with growth delay due to a growth hormone deficiency associated with a polymalformation syndrome. They both displayed pre-auricular pits and tags, imperfor

WHAT IS KNOWN AND OBJECTIVE: Insulin resistance impairs the impact of levothyroxine on thyroid autoimmunity and hypothalamic-pituitary-thyroid axis activity. Both metformin and myo-inositol were found to improve insulin sensitivity and to reduce thyrotropin levels in individuals with hypothyroidism. The aim of the present study was to compare the effect of levothyroxine on thyroid autoimmunity and hypothalamic-pituitary-thyroid axis activity between women receiving metformin and myo-inositol.
METHODS: The study included two groups of women with autoimmune hypothyroidism, treated for at least

Polycystic ovary syndrome (PCOS) is a hypothalamic-pituitary-gonadal (HPG) axis disorder. PCOS symptoms most likely result from a disturbance in the complex feedback regulation system of the HPG axis, which involves gonadotrophic hormones and ovarian steroid hormones. However, the nature of this complex and interconnecting feedback regulation makes it difficult to dissect the molecular mechanisms responsible for PCOS phenotypes. Global estrogen receptor α (ERα) knockout (KO) mice exhibit a disruption of the HPG axis, resulting in hormonal dysregulation in which female ERα KO mice have elevated

Obesity and anxiety disorders are frequently comorbid, yet the neural circuit mechanisms linking metabolic and affective states remain incompletely understood. Here, in male mice exposed to long-term high-fat diet, we identify a hypothalamic circuit comprising arcuate nucleus agouti-related protein neurons and paraventricular nucleus corticotropin-releasing hormone neurons that regulates anxiety-like behavior under metabolic stress, with downstream recruitment of lateral hypothalamic area glutamatergic neurons to modulate feeding in a state-dependent manner. In vivo fiber photometry and ex viv

BACKGROUND/AIM: Hypothalamic-pituitary (HT-P) dysfunction is one of the most common endocrine late effects following cranial radiotherapy. However, there are currently no specific data describing this complication in adult-onset cancer patients after whole brain radiotherapy (WBRT). The present cohort study aims to establish the prevalence of HT-P axis dysfunction in this group of patients.
PATIENTS AND METHODS: Twenty-six cancer patients previously treated with WBRT (median follow-up=20.5 months) received standardized endocrine check-up focusing on HT-P function.
RESULTS: In 50% of the pati

PURPOSE: Obesity is a common consequence in patients with tumors of the hypothalamic region and of related treatment in children. Much less information is available on adult patients and long-term survivors. The aims of this study were to estimate the prevalence of obesity in adult patients with acquired structural hypothalamic damage and to define the characteristics of patients at greatest risk of obesity.
METHODS: A retrospective study was conducted of 52 patients (25 women; median age at diagnosis, 44 years; range, 17 to 78 years) with tumors involving the hypothalamic region. These inclu
