Pro/con: vaccination and waning immunity - presents contrasting perspectives on the durability of vaccine-induced protection and the potential need for booster strategies.

PURPOSE OF REVIEW: mpox vaccination remains the only effective drug-based tool against mpox. Currently, there are two third-generation vaccines: LC16m8 and MVA-BN. Immunogenicity, observational and modelling data have demonstrated their efficacy in preventing mpox. However, evidence of waning immunity from 1 year after mpox vaccination has emerged, while mpox is still circulating worldwide, and there is a threat of a more contagious and virulent clade. RECENT FINDINGS: We review the available data on immunogenicity of mpox vaccines, after primary and booster dose. There are pros and cons to r

Impact of pre-Omicron COVID-19 vaccine boosters on the risk of Omicron variant infections: A systematic review and meta-regression.

BACKGROUND: An argument against COVID-19 vaccine boosters is that immune imprinting may impair immune responses to new SARS-CoV-2 variants, as some epidemiological studies found a paradoxical increase in Omicron variant infections correlated with the number of prior pre-Omicron vaccine doses. However, substantial variability between studies has left the true impact of boosters uncertain, warranting further investigation. METHODS: We systematically reviewed available data and applied meta-regression to identify sources of heterogeneity among studies that examined the impact of pre-Omicron COVI

Bistability and complex bifurcation diagrams generated by waning and boosting of immunity.

We investigate an epidemiological model that incorporates waning of immunity at the individual level and boosting of the immune system upon re-exposure to the pathogen. When immunity is fully restored upon boosting, the system can be expressed as an SIRS-type model with discrete and distributed delays. We conduct a numerical bifurcation analysis varying the boosting force and the maximum period of immunity (in the absence of boosting), while keeping other parameters fixed at values representative of a highly infectious disease like pertussis. The stability switches of the endemic equilibrium,

An overview of the BCG vaccine and its future scope.

Despite intense elimination efforts, tuberculosis (TB) still poses a threat to world health, disproportionately affecting less developed and poorer countries. The Bacillus Calmette-Guérin (BCG) vaccine, the only anti-TB authorized vaccine can partially stop TB infection and transmission, however, its effectiveness ranges from 0 to 80%. As a result, there is an urgent need for a more potent TB vaccination given the widespread incidence of the disease. Enhancing BCG's effectiveness is also important due to the lack of other licensed vaccinations. Recently, fascinating research into BCG revaccina

Bayesian inference for prediction of survival probability in prime-boost vaccination regimes.

We focus on Bayesian inference for survival probabilities in a prime-boost vaccination regime in the development of an Ebola vaccine. We are interested in the heterologous prime-boost regimen (unmatched vaccine deliverys using the same antigen) due to its demonstrated durable immunity, well-tolerated safety profile, and suitability as a population vaccination strategy. Our research is motivated by the need to estimate the survival probability given the administered dosage. To do so, we establish two key relationships. Firstly, we model the connection between the designed dose concentration and

T cell reactivity to Bordetella pertussis is highly diverse regardless of childhood vaccination.

The incidence of whooping cough due to Bordetella pertussis (BP) infections has increased recently. It is believed that the shift from whole-cell pertussis (wP) vaccines to acellular pertussis (aP) vaccines may be contributing to this rise. While T cells are key in controlling and preventing disease, nearly all knowledge relates to antigens in aP vaccines. A whole-genome mapping of human BP-specific CD4+ T cell responses was performed in healthy vaccinated adults and revealed unexpected broad reactivity to hundreds of antigens. The overall pattern and magnitude of T cell responses to aP and no

Preferences and willingness of accepting COVID-19 vaccine booster: Results from a middle-income country.

The recent wave of COVID-19 cases has led to the potential need for booster doses. We surveyed 6,294 people and found that 87.6% reported willingness to take a booster dose, with vaccine efficacy rate being the most common reason cited to accept booster dose. Differences in acceptance rates were noted among those working in non-health related sectors, different ethnic groups as well as those who had taken viral vector vaccines.

Novel vaccine prime and selective BCG boost: a new tuberculosis vaccine strategy for infants of HIV-infected mothers.

Novel tuberculosis vaccination strategies hinge on BCG priming, yet newborn BCG vaccination may cause BCG disease in HIV-infected infants. Viral-vectored or subunit prime vaccine, followed by delayed BCG boost only for HIV-uninfected infants, may be a safe alternative for all newborns, regardless of maternal HIV infection. This approach should be tested using new tuberculosis vaccine candidates. If safety and immunogenicity of a novel vaccine prime is established in infants of HIV-infected mothers, for whom newborn BCG carries unacceptable risk, this strategy might then be compared to conventi

Carotid artery dissection as a possible severe complication of pertussis in an adult: clinical case report and review.

Adults can experience typical symptoms of pertussis. Complications due to severe paroxysmal coughing have been reported in this age group. We report carotid artery dissection as a possible severe complication of pertussis in a previously healthy adult. Adult booster immunization against pertussis should be considered to protect adults themselves from this illness and its potential complications.

[Cytogenetic study of blood lymphocytes in children revaccinated against smallpox].

As the result of primary re-immunization with small-pox vaccine of eight years' old children chromosome aberrations were observed in their peripheral blood lymphocytes, the frequencies being 5.3% and 7.9% on the seventh day and in a month after the reimmunization respectively. Chromosome aberrations were significantly more rare in children with a high level of immunity retained after the preceding immunization as compared to those whose immunity was weakened with time. After the elapse of 6 months following the re-immunization the frequency of chromosome aberrationd did not exceed the initial

Overcoming barriers to polio eradication in challenging subnational geographies: lessons from South Khyber Pakhtunkhwa, Pakistan, 2022-2024.

The global polio program has relied on supplementary immunization activities (SIA) in countries where routine immunization (RI) coverage is suboptimal. During these SIAs, oral poliovirus vaccine (OPV) is administered to all children under the age of 5 years. However, implementing SIAs in areas with insecurity and conflict, such as the South Khyber Pakhtunkhwa (South KP) region of Pakistan is very challenging because of attacks against vaccination teams and community boycotts among other reasons. These challenges sometimes lead to cancelation, delays, and poor quality of immunization campaigns.
