Salvigenin alleviates diabetic nephropathy through Nrf2-mediated modulation of renal inflammation and ferroptosis.

BACKGROUND: Diabetic nephropathy (DN) is a leading cause of global end-stage renal disease, driven by oxidative stress, persistent inflammation and ferroptosis. Salvigenin, a natural flavonoid with anti-inflammatory and antioxidant activity, displays protective effects against oxidative injury. This study investigated its renoprotective actions in DN and the underlying mechanism. METHODS: In vitro high glucose-stimulated HK-2 cell model and in vivo db/db DN mouse model were adopted. HK-2 cells were treated with gradient salvigenin for 24 h. We assessed cell viability, proliferation, inflammat

Spatially selective clearance of inflammation-mediated reactive oxygen species in injured kidney cells.

Rationale: Kidney injury is characterized by the accumulation of reactive oxygen species (ROS). Current ROS scavengers utilized in the treatment of clinical kidney injury lack the capacity for spatially selective ROS scavenging within the injured cells, often resulting in irreversible damage to healthy tissues. Methods: We developed a spatially selective therapeutic strategy using platelet-shipped tetrahedral framework nucleic acids (TFNAs@PLT). Results: In this system, platelets achieve spatial targeting of injured kidney cells by responding to inflammatory signals and releasing TFNAs followi

The mechanism of different microglial receptors mediating central inflammation in chronic migraine: a meta-analysis.

BACKGROUND: Microglial activation is increasingly recognized as a critical contributor to both the initiation and chronification of migraine. Recent advances have highlighted the importance of receptor-mediated signaling pathways in regulating microglial responses; however, their integrated role in central inflammation remains insufficiently characterized. This review aimed to evaluate the mechanistic involvement and therapeutic relevance of microglial receptors in chronic migraine. METHODS: A comprehensive systematic search was conducted in PubMed, Web of Science, and Scopus to identify studi

Systemic inflammation as a mediator between food preferences and metabolic syndrome: a cross-sectional study.

BACKGROUNDS: Both dietary preferences and systemic inflammation are associated with metabolic syndrome (MetS); however, the role of inflammation in the association between dietary preferences and MetS remains largely unclear. METHODS: A total of 33,728 participants were included from the Henan Rural Cohort Study (n = 39,259). Food preferences were collected using a questionnaire. Generalized linear models were used to investigate the independent and joint associations of food preferences, specifically for sour or spicy foods, and inflammatory biomarkers [such as systemic immune-inflammatory i

The impact of adverse childhood experiences on gut microbiota and markers of inflammation is mediated by obesity and depression.

BACKGROUND: Adverse childhood experiences (ACEs) are associated with poor health outcomes in adulthood including obesity, psychiatric symptoms, and elevated levels of inflammatory markers. Our previous work found ACEs are associated with altered gut microbiota composition. In the present work, we examined ACE associations with gut microbiota and peripheral measures of inflammation in pregnant women with or without obesity, and explored potential modifying factors including diet and depressive symptoms. METHODS: Female participants were recruited in the third trimester of pregnancy as part of

Loss of Progranulin Expression Decreases NLRP3 Inflammasome-Mediated Inflammation and Enhances Bone Anabolism.

Progranulin (PGRN), a glycosylated protein, is expressed in most tissues, including bones, and its level is elevated in the serum and joints of individuals with inflammatory bone loss disorders such as rheumatoid arthritis (RA). Previously, using global and macrophage-specific Grn deletion mice, we demonstrated that loss of PGRN protects against age-related bone loss selectively in females, suggesting a sex-dependent role for macrophage-derived PGRN in skeletal homeostasis. Here, we investigated the role of PGRN in the regulation of macrophage-mediated inflammation and bone formation. Immune-p

Distinct Inflammation-Associated Microbiome Signatures in Pediatric Non-IgE-Mediated Food Allergy.

Non-IgE-mediated food allergy is characterized by delayed gastrointestinal manifestations and the absence of reliable non-invasive biomarkers. Increasing evidence suggests that gut microbiota may contribute to this disease pathogenesis. The aim of this study was to characterize the gut microbiome composition in thirty pediatric patients diagnosed with non-IgE-mediated food allergy, in comparison to fifteen healthy controls children, and to investigate its association with fecal calprotectin, eosinophil-derived neurotoxin (EDN) and IgA. Gut microbiota profiling was performed by 16S rRNA gene se

Protein Phosphatase 2A Modulates AKT-Mediated Macrophage Polarization in Adipocyte-Induced Inflammation.

Obesity and type 2 diabetes mellitus (T2DM) are associated with chronic low-grade inflammation that sustains insulin resistance and metabolic dysfunction. Adipose tissue, through the secretion of adipokines and cytokines, plays a central role in shaping macrophage polarization and maintenance of an inflammatory microenvironment. While kinase-mediated signaling such as PI3K-AKT has been extensively studied, the contribution of phosphatases, particularly protein phosphatase 2A (PP2A), to macrophage polarization remains poorly defined. In this study, RAW264.7 macrophages exposed to inflammatory a

Estrogen receptor beta limits T cell-mediated inflammation to maintain immune homeostasis.

Many autoimmune diseases exhibit a female sex bias in prevalence and severity, yet the mechanisms for this remain unclear. 17β-estradiol (E2) is a steroid sex hormone with established immunomodulatory roles in CD4+ T cells, which express the nuclear estrogen receptors ERα and ERβ. Autoimmune disease patients exhibit reductions in ERβ expression, suggesting that dysregulated E2 signaling contributes to inflammation. We previously identified a novel role for ERβ in promoting the TGF-β-dependent differentiation of Foxp3+ Tregs, supporting the idea that ERβ has anti-inflammatory functions. In this

Succinate Ameliorates Sepsis-Associated Acute Kidney Injury by Mediating Inflammation via GPR91.

Succinate is a key metabolic intermediate in the tricarboxylic acid (TCA) cycle, which participates in energy metabolism. It accumulates under various pathological conditions and exerts regulatory effects through binding to its specific receptor GPR91. However, its specific function in sepsis-associated acute kidney injury (SA-AKI) remains unknown. Herein, we utilized GPR91 knockout (GPR91-/-) mice and LPS-induced SA-AKI mouse models to elucidate the role and mechanisms of the succinate-GPR91 axis in SA-AKI. The results demonstrated significantly elevated succinate levels in the circulation of

Inhibiting HuR-Mediated mRNA Stabilization Attenuates Psoriasis-Like Inflammation in Preclinical Models.

PURPOSE: Psoriasis is a chronic inflammatory skin disease characterized by hyperproliferative keratinocytes and activated Th17/Th1 lymphocytes, which sustain a self-perpetuating cycle by releasing pro-inflammatory cytokines. The RNA-binding protein (RBP) Human Antigen R (HuR) stabilizes many of these cytokine mRNAs, contributing to disease progression. This study aims to investigate the role of the HuR protein in psoriasis and modulate its activity to develop novel therapeutic strategies. PATIENTS AND METHODS: We evaluated the effects of Human Antigen R (HuR) silencing on psoriatic markers in

Macrophage-Mediated Transport of Insoluble Indirubin Induces Hepatic Injury During Intestinal Inflammation.

Many plant-derived bioactive molecules with low solubility and permeability can cause hepatocyte injury. However, the mechanism by which they induce hepatic damage without passive diffusion into the hepatic circulatory system remains unclear. This study demonstrate that indirubin, the main component of indigo naturalis with poor aqueous solubility, predisposes mice with chronic colitis to hepatic injury. This closely mimics the hepatic damage commonly observed in ulcerative colitis patients treated with indigo naturalis. Upon administration, indirubin is detected in the plasma, Peyer's patches

The inflammatory architecture reflects the effects of pharmacological and genetic interventions on resolution of TLR2-mediated inflammation.

INTRODUCTION: Immune cells form defined pro- and anti-inflammatory regions around a pathogen during an innate immune response. These include, in Toll-like receptor (TLR)-2-induced inflammation, a core region containing the pathogen, an adjacent pro-inflammatory (PI) region and a surrounding anti-inflammatory (AI) region. Interventions targeting specific immune cells or signaling pathways disrupt this architecture and affect the resolution of inflammation. Here, we investigated, which changes in the inflammatory architecture may favor an increased resolution of inflammation. METHODS: Immune ce

Inflammation mediation of the association between brominated flame retardants and psoriasis among U.S. adults.

BACKGROUND: Brominated flame retardants (BFRs), widely applied in fire prevention, are persistent environmental pollutants. Accumulating evidence indicates that BFRs can trigger inflammatory responses in humans, potentially contributing to various diseases. Given the central role of inflammation in psoriasis pathogenesis, investigating the association between BFR exposure and psoriasis risk is of considerable importance. METHODS: We analyzed data from the 2005-2006 and 2009-2014 cycles of the National Health and Nutrition Examination Survey (NHANES). concentrations of BFRs were natural logari

Poly-T-Modified Gold Nanorods Suppress Macrophage-Mediated Inflammation for Periodontitis Therapy.

Traditional treatments for periodontitis are limited by their inability to adequately modulate the immune response and control inflammation. Recently, nucleic acid-modified nanomaterials have attracted significant attention for their potential in regulating inflammation. Among these, most nanomaterials, such as spherical nucleic acids, tend to exhibit pro-inflammatory effects. In this study, we identified for the first time that poly-T sequence-modified gold nanorods (PTM AuNRs) possess significant anti-inflammatory properties. The PTM AuNRs demonstrated excellent biocompatibility and efficacy

Exosome-mediated modulation of macrophage polarization and inflammation in early Klebsiella pneumoniae lung infections.

Klebsiella pneumoniae is a major pathogen responsible for severe pulmonary infections, yet the early mechanisms of infection remain incompletely understood. This study investigates the role of exosomes derived from K. pneumoniae in polarizing macrophages to the M1 phenotype, thereby facilitating early lung infections. Utilizing single-cell Raman spectroscopy, we rapidly detected K. pneumoniae within host cells and observed significant lipid expression changes. Metabolomic analysis of exosomes from infected epithelial cells uncovered an elevation of phosphatidylcholine, which disrupted endothel
