Toll-like receptor cascade and gene polymorphism in host-pathogen interaction in Lyme disease.

Lyme disease (LD) risk occurs in North America and Europe where the tick vectors of the causal agent Borrelia burgdorferi sensu lato are found. It is associated with local and systemic manifestations, and has persistent posttreatment health complications in some individuals. The innate immune system likely plays a critical role in both host defense against B. burgdorferi and disease severity. Recognition of B. burgdorferi, activation of the innate immune system, production of proinflammatory cytokines, and modulation of the host adaptive responses are all initiated by Toll-like receptors (TLRs

Internalization-dependent recognition of Mycobacterium avium ssp. paratuberculosis by intestinal epithelial cells.

Mycobacterium avium ssp. paratuberculosis (MAP) is the causative agent of Johne's disease, a highly prevalent chronic intestinal infection in domestic and wildlife ruminants. The microbial pathogenesis of MAP infection has attracted additional attention due to an association with the human enteric inflammatory Crohn's disease. MAP is acquired by the faecal-oral route prompting us to study the interaction with differentiated intestinal epithelial cells. MAP was rapidly internalized and accumulated in a late endosomal compartment. In contrast to other opportunistic mycobacteria or M. bovis, MAP

Screening for C-type lectin receptor (CLR)/bacteria interactions using a bovine CLR-Fc fusion protein library reveals recognition of Pasteurella multocida B:2 by MICL.

Pattern recognition receptors (PRRs) are an essential component of the innate immune system. Myeloid C-type-lectin receptors (CLRs) serve as PRRs and play a crucial role in pathogen recognition. While the role of CLRs has been mainly studied in mice and humans, their function in cattle is poorly understood. To address this gap, we generated a novel bovine CLR-hFc fusion protein library, enabling high-throughput screening of bovine CLR/pathogen interactions. The functionality of the bovine CLR-hFc fusion proteins was validated with known CLR ligands using ELISA- and flow cytometry-based binding

Bifunctional glycolipids targeting TLR4·MD-2 and short pentraxins.

Innate immune detection of pathogen- and danger-associated molecular patterns (PAMPs/DAMPs) centres on pattern-recognition receptors, with the TLR4/MD-2 complex being uniquely sensitive to trace levels of lipopolysaccharide (LPS) as well as infection-triggered endogenous ligands. While this axis rapidly induces protective cytokine production and upregulation of co-stimulatory molecules, its malfunction can cause pathological hyperinflammation culminating in systemic inflammatory response syndrome (SIRS), highlighting the importance of the development of TLR4 antagonists for the management of i

Molluscum Contagiosum Virus Protein MC005 Inhibits NF-κB Activation by Targeting NEMO-Regulated IκB Kinase Activation.

Molluscum contagiosum virus (MCV), the only known extant human-adapted poxvirus, causes a long-duration infection characterized by skin lesions that typically display an absence of inflammation despite containing high titers of live virus. Despite this curious presentation, MCV is very poorly characterized in terms of host-pathogen interactions. The absence of inflammation around MCV lesions suggests the presence of potent inhibitors of human antiviral immunity and inflammation. However, only a small number of MCV immunomodulatory genes have been characterized in detail. It is likely that many

The ZFP36 Family as a Post-Transcriptional Immune Checkpoint in Immunity and Disease: Molecular Mechanisms and Functional Implications.

The zinc finger protein 36 (ZFP36) family, including ZFP36/tristetraprolin (TTP), ZFP36 CCCH-type-like 1 (ZFP36L1), and ZFP36 CCCH-type-like 2 (ZFP36L2), consists of conserved CCCH-type tandem zinc-finger RNA-binding proteins. These proteins recognize AU-rich elements (AREs) in target mRNAs and promote deadenylation, decay, and translational repression. In this review, we use the term post-transcriptional immune checkpoint in a restricted conceptual sense: ZFP36 family proteins are intracellular, RNA-level negative regulators that tune the magnitude, duration, and resolution of immune effector

Newcastle Disease Virus V Protein Degrades Mitochondrial Antiviral Signaling Protein To Inhibit Host Type I Interferon Production via E3 Ubiquitin Ligase RNF5.

Paramyxovirus establishes an intimate and complex interaction with the host cell to counteract the antiviral responses elicited by the cell. Of the various pattern recognition receptors in the host, the cytosolic RNA helicases interact with viral RNA to activate the mitochondrial antiviral signaling protein (MAVS) and subsequent cellular interferon (IFN) response. On the other hand, viruses explore multiple strategies to resist host immunity. In this study, we found that Newcastle disease virus (NDV) infection induced MAVS degradation. Further analysis showed that NDV V protein degraded MAVS t

Protein Posttranslational Modifications in Immunity: Molecular Mechanisms and Therapeutic Targets.

The immune response is a highly dynamic and precisely controlled process, and relies on an intricate network of protein interactions to maintain its homeostasis. Protein posttranslational modifications (PTMs), by covalent addition of chemical groups or peptide chains, or by proteolytic cleavage, directly alter the structure, activity, electrical charge, thermal stability, and function of immune-related proteins. Here, we enumerate the main PTMs in the immune system and highlight their global regulatory roles across innate immunity, T-cell-mediated cellular immunity, and B-cell-mediated humoral

Brucella-infected macrophage-derived microparticles enhance the host anti-Brucella acquired immune response.

Microparticles (MPs) serve as critical mediators of intercellular communication by shuttling bioactive molecules, holding significant potential for clinical diagnostics and next-generation vaccine development. Brucella, an intracellular pathogen responsible for severe zoonosis, evades host immunity through persistent infection; however, the role of Brucella-induced MPs in modulating adaptive immunity remains poorly defined. In this study, we demonstrate that the infection of RAW264.7 macrophages with the attenuated Brucella melitensis strain M5 triggers robust MP release. These MPs enhance hos

BR-SIGNALING KINASE1 physically associates with FLAGELLIN SENSING2 and regulates plant innate immunity in Arabidopsis.

Pathogen-associated molecular pattern (PAMP)-trigged immunity (PTI) is the first defensive line of plant innate immunity and is mediated by pattern recognition receptors. Here, we show that a mutation in BR-SIGNALING KINASE1 (BSK1), a substrate of the brassinosteroid (BR) receptor BRASSINOSTEROID INSENSITIVE1, suppressed the powdery mildew resistance caused by a mutation in ENHANCED DISEASE RESISTANCE2, which negatively regulates powdery mildew resistance and programmed cell death, in Arabidopsis thaliana. A small secreted protein triggers a TLR2/4-dependent inflammatory response during invasive Candida albicans infection.

Candida albicans can switch from commensal to pathogenic mode, causing mucosal or disseminated candidiasis. The host relies on pattern-recognition receptors including Toll-like receptors (TLRs) and C-type lectin receptors (CLRs) to sense invading fungal pathogens and launch immune defense mechanisms. However, the complex interplay between fungus and host innate immunity remains incompletely understood. Here we report that C. albicans upregulates expression of a small secreted cysteine-rich protein Sel1 upon encountering limited nitrogen and abundant serum. Sel1 activates NF-κB and MAPK signali

Zoonotic orthopoxviruses encode a high-affinity antagonist of NKG2D.

NK and T lymphocytes express both activating and inhibiting receptors for various members of the major histocompatibility complex class I superfamily (MHCISF). To evade immunologic cytotoxicity, many viruses interfere with the function of these receptors, generally by altering the displayed profile of MHCISF proteins on host cells. Using a structurally constrained hidden Markov model, we discovered an orthopoxvirus protein, itself distantly class I-like, that acts as a competitive antagonist of the NKG2D activating receptor. This orthopoxvirus MHC class I-like protein (OMCP) is conserved among

Oncofetal RNA-binding proteins of IGF2BP family suppress IRF-3 and NF-kB dependent transcription downstream of cytosolic RNA sensors.

Activation of innate inflammatory signaling and tumor-specific antigen presentation in cancer cells provides a foundation for anti-cancer immunotherapies. Here, we show that the insulin-like growth factor 2 mRNA-binding proteins (IGF2BP1, IGF2BP2, and IGF2BP3), which are upregulated across diverse human malignancies, including acute myeloid leukemia (AML), suppress RNA-sensing pattern recognition receptor signaling and downstream ISRE- and NF-κB-dependent transcription. Among these pathways, RIG-I signaling is particularly sensitive to IGF2BP-mediated inhibition. This suppressive effect is str

Microbiological mechanisms of immune response modulation: clinical implications.

The human microbiota is a major regulator of immune development, mucosal barrier integrity, and systemic immune tone. Its effects are mediated by microbe-associated molecular patterns, pattern-recognition receptors, microbial metabolites such as short-chain fatty acids, tryptophan-derived indoles, secondary bile acids and vitamin-pathway metabolites, and direct interactions with epithelial and immune cells. A balanced microbiota supports regulatory T-cell responses, secretory IgA production, innate-like lymphocyte maturation and epithelial barrier function, whereas dysbiosis may promote barrie

Non-IgE mediated mast cell activation.

Mast cells (MCs) are innate immune cells that are scattered in tissues throughout the organism being particularly abundant at sites exposed to the environment such as the skin and mucosal surfaces. Generally known for their role in IgE-mediated allergies, they have also important functions in the maintenance of tissue integrity by constantly sensing their microenvironment for signals by inflammatory triggers that can comprise infectious agents, toxins, hormones, alarmins, metabolic states, etc. When triggered their main function is to release a whole set of inflammatory mediators, cytokines, c

MAPK Activation Is Essential for Waddlia chondrophila Induced CXCL8 Expression in Human Epithelial Cells.

BACKGROUND: Waddlia chondrophila (W. chondrophila) is an emerging agent of respiratory and reproductive disease in humans and cattle. The organism is a member of the order Chlamydiales, and shares many similarities at the genome level and in growth studies with other well-characterised zoonotic chlamydial agents, such as Chlamydia abortus (C. abortus). The current study investigated the growth characteristics and innate immune responses of human and ruminant epithelial cells in response to infection with W. chondrophila. METHODS: Human epithelial cells (HEp2) were infected with W. chondrophil

Decoding epithelial-immune cell crosstalk in uropathogenic Escherichia coli urinary tract infections.

Urinary tract infections (UTIs) pose a significant global health burden, characterized by a spectrum of clinical outcomes shaped by intricate epithelial-immune interactions. Uropathogenic Escherichia coli (UPEC) is the predominant etiological agent, accounting for the majority of cystitis cases. Upon breaching urothelial barriers, UPEC triggers a robust innate immune response involving cytokine secretion, antimicrobial peptide production, and recruitment of neutrophils and other immune cells. This review synthesizes current understanding of the dynamic crosstalk between urothelial and immune c

Activation of innate host defense mechanisms by Borrelia.

Borrelia is the causative agent of Lyme disease, a widespread disease with important health consequences. Immune-mediated mechanisms are believed to play a major role in both host defense and in late complications of Lyme disease. Recognition of Borrelia and the initial activation of the innate immune system are important for host defense, as well as modulation of adaptive responses. Several classes of pattern recognition receptors (PRRs) have been suggested to be involved in the recognition of Borrelia: Toll-like receptors (TLRs), NOD-like receptors (NLRs) and C-type lectin receptors (CLRs).

Innate immunity and disorders of the female reproductive tract.

Sexually transmitted infections, and their associated sequelae, such as tubal infertility, ectopic pregnancy and preterm labour, are a major worldwide health problem. Chlamydia trachomatis infection is thought to be the leading global cause of tubal infertility and tubal ectopic pregnancy. Preterm birth occurs in around 10% of all deliveries, and nearly 30% of preterm deliveries are associated with intrauterine infection. The mucosal innate immune system of the female reproductive tract has evolved to eliminate such sexually transmitted pathogens whilst maintaining its ability to accommodate s
