Exome sequencing in Jewish and Arab patients with rhabdomyolysis reveals single-gene etiology in 43% of cases.

BACKGROUND: Rhabdomyolysis is a clinical emergency that may cause acute kidney injury (AKI). It can be acquired or due to monogenic mutations. Around 60 different rare monogenic forms of rhabdomyolysis have been reported to date. In the clinical setting, identifying the underlying molecular diagnosis is challenging due to nonspecific presentation, the high number of causative genes, and current lack of data on the prevalence of monogenic forms. METHODS: We employed whole exome sequencing (WES) to reveal the percentage of rhabdomyolysis cases explained by single-gene (monogenic) mutations in o

Glucose-6-phosphate dehydrogenase deficiency and borderline deficiency: association with neonatal hyperbilirubinemia.

OBJECTIVE: To characterize the occurrence of glucose-6-phosphate dehydrogenase (G6PD) deficiency and its association with neonatal hyperbilirubinemia. STUDY DESIGN: This study involved an evaluation of G6PD data for 2656 newborns from a universal newborn screening program. RESULTS: Mean G6PD activity was 14.2 ± 3.3 U/g Hb. Some 2.71% of the newborns were G6PD-deficient, and 1.77% had borderline G6PD activity, with male and female predominance, respectively. G6PD deficiency was more prevalent in newborns of Sephardic Jew and Muslim Arab backgrounds. The infants with G6PD deficiency had higher

Complications of circumcision in Israel: a one year multicenter survey.

BACKGROUND: In Israel, virtually all children undergo circumcision in the neonatal period. Traditionally, it is commonly performed by a "Mohel" (ritual circumciser) but lately there is an increasing tendency among the educated secular population to prefer a medical procedure performed by a physician and with local anesthetic injection. OBJECTIVES: To evaluate the outcome of this procedure and to compare the complication rate following circumcisions performed by ritual circumcisers and by physicians. METHODS: In 2001, of the 19,478 males born in four major medical centers in Israel 66 had cir

Lesion topography in two cases of nifedipine-related pemphigus.

Similar lesion topography which resembled an inverted fir tree in two patients with nifedipine-related pemphigus vulgaris is described. The possible relation of the lesion pattern to the offending drug is discussed.

Martsolf's syndrome in a non-Jewish boy.

Martsolf's syndrome has been described in Jewish people. We describe a patient of non-Jewish ancestry who has minor differences from other patients. The possible pattern of inheritance is discussed.

Aggression and Victimization Among Employees in Departments of Social Services in Israel.

This study examined whether employee stances of hostile attribution, perceived hostile climate, attitudes toward aggression, subjective norms, and perceived control affected workplace aggression and victimization in departments of social services. We also explored differences in workplace aggression and victimization in social services in Israel and compared departments of social services of mixed and nonmixed cities (Arabs and Jews). A sample of 470 social workers employed by social services in Israel completed self-report measures. We used regression analysis to explore levels of aggression …

Co-occurrence of autoimmune diseases and metabolic disorders in familial mediterranean fever patients: review of literature and case reports.

Familial Mediterranean Fever (FMF) is an autosomal recessive autoinflammatory disease predominantly affecting populations from the Mediterranean region, with a high prevalence reported among Armenians, Turks, Arabs, and Jews. The condition results from mutations in the MEFV gene, which encodes pyrin, a key regulator of inflammation. Both genetic and epigenetic factors contribute to the diverse clinical manifestations of FMF, which are characterized by recurrent episodes of fever, abdominal pain, chest pain, arthritis, and erysipelas-like erythema. In addition to these symptoms, individuals wit…

A Review of Gaucher Disease Pathophysiology, Clinical Presentation and Treatments.

Gaucher disease (GD, ORPHA355) is a rare, autosomal recessive genetic disorder. It is caused by a deficiency of the lysosomal enzyme, glucocerebrosidase, which leads to an accumulation of its substrate, glucosylceramide, in macrophages. In the general population, its incidence is approximately 1/40,000 to 1/60,000 births, rising to 1/800 in Ashkenazi Jews. The main cause of the cytopenia, splenomegaly, hepatomegaly, and bone lesions associated with the disease is considered to be the infiltration of the bone marrow, spleen, and liver by Gaucher cells. Type-1 Gaucher disease, which affects the …

Familial Creutzfeldt-Jakob disease with the E200K mutation: longitudinal neuroimaging from asymptomatic to symptomatic CJD.

Familial Creutzfeldt-Jakob disease (fCJD) in Jews of Libyan ancestry is caused by an E200K mutation in the PRNP gene. While carriers are born with this mutation, they usually remain asymptomatic until middle age. Early detection of conversion is crucial for understanding and eventually for the treatment of the disease. The aim of this study was to report longitudinal MRI data in E200K individuals who eventually converted from healthy mutation carriers to clinically symptomatic CJD. As a part of a prospective study, asymptomatic E200K mutation carriers were scanned annually until their conversi…

The founder Ashkenazi Jewish mutations in the MSH2 and MSH6 genes in Israeli patients with gastric and pancreatic cancer.

The genetic basis for gastric and pancreatic cancer is largely undetermined. These cancers are overrepresented in hereditary non polyposis c … astric (n = 23) and pancreatic (n = 120) cancer. The majority of cases (100/143-70%) were Ashkenazi Jews, and 10% (n = 16)--of mixed Ashkenazi-non Ashkenazi Jewish ancestry, and most participants (n = 96-67.1%) had a positive family history of cancer. Genotyping the MSH2*A636P mutation was performed by PCR followed by restriction enzyme digest, and the …

Vitamin D Receptor (VDR) polymorphic variants in patients with cancer and Gaucher disease.

BACKGROUND: An increased risk has been reported in Gaucher disease genotype N370S/N370S for multiple myeloma, hematologic and other malignancies, as well as skeletal involvement. The purpose of this study is to ascertain whether VDR genotype which is involved in the risk for many cancers including multiple myeloma is also correlated with cancers in patients with Gaucher disease. METHODS: VDR polymorphisms (ApaI, TaqI, BsmI, FokI) were analyzed by standard methods from 29 Gaucher patients (some on enzyme therapy) with various cancers and from 70 healthy Ashkenazi Jews. Associations between VDR…

Two founder mutations in the SEC23B gene account for the relatively high frequency of CDA II in the Italian population.

Congenital Dyserythropoietic Anemia type II is an autosomal recessive disorder characterized by unique abnormalities in the differentiation … frequency of E109K is similar in Italy and in the rest of Europe (and is also prevalent in Moroccan Jews), the relative frequency of R14W is significantly higher in Italy (26.3% vs. 10.7%). By haplotype analysis we demonstrated that both are founder mutations in the Italian population. By using the DMLE+ program our estimate for the age of the E109K mu …

The molecular basis of autosomal recessive diseases among the Arabs and Druze in Israel.

The Israeli population mainly includes Jews, Muslim and Christian Arabs, and Druze In the last decade, data on genetic diseases present in the population have been systematically collected and are available online in the Israeli national genetic database ( http://www.goldenhelix.org/server/israeli ). In the non-Jewish population, up to 1 July 2010, the database included molecular data on six diseases relatively frequent in the whole population: thalassemia, familial Mediterranean fever (FMF), cystic fibrosis, deafness, phenylketonuria and congenital adrenal hyperplasia, as well as data on 195 …
