[Molecular pathology of the erythrocyte membrane].

The erythrocyte membrane, in the broad sense of the term, comprises a lipid bilayer and below it the erythrocyte skeleton responsible for the mechanical properties of red cells. The lipid bilayer contains numerous proteins including the anion transporter. The underlying portion contains spectrin, actin and protein 4.1. At the interface between these two portions, ankyrin, protein 4.2 and other proteins are found. The structure and mode of expression of the corresponding genes are roughly known, as are the structure-function relationships within the proteins. In the wake, or sometimes ahead of

Decoding the Membrane Interactions of Antimicrobial Peptoids via Quartz Crystal Microbalance Nano-Viscometry.

The rise in antimicrobial resistance underscores the urgent need for novel therapeutics. Membrane-disrupting agents offer a possible solution. Natural antimicrobial peptides are membrane-active but suffer from proteolytic instability and poor bioavailability. Sequence-defined peptoids have emerged as promising antimicrobial peptide mimetics, offering enhanced stability, tunable side-chain chemistry, and potent activity against drug-resistant pathogens. Here, we employed quartz crystal microbalance (QCM)-based viscoelastic fingerprinting, complemented by dye leakage assays, to characterize the

Effects of charged drugs on the phase transition temperatures of phospholipid bilayers.

An approach is presented which allows the description of drug binding to lipid bilayers, when the drug is present in both charged and uncharged forms. Binding is described by Langmuir adsorption isotherms, with the maximum number of binding sites being 1/60 A2. An estimate of the change in drug pK on binding is necessary, and is close to zero for most drugs binding to dipalmitoyl phosphatidylcholine, although delta pK = 1.0 for procaine. From the binding curves it is possible to calculate the drug-induced decreases in lipid phase transition temperature, assuming ideal behaviour. Good fits betw…

Perturbed voltage-gated channel activity in perturbed bilayers: implications for ectopic arrhythmias arising from damaged membrane.

The ceaseless opening and closing of the voltage-gated channels (VGCs) underlying cardiac rhythmicity is controlled, in each VGC, by four mobile voltage sensors embedded in bilayer. Every action potential necessitates extensive packing/repacking of voltage sensor domains with adjacent interacting lipid molecules. This renders VGC activity mechanosensitive (MS), i.e., energetically sensitive to the bilayer's mechanical state. Irreversible perturbations of sarcolemmal bilayer such as those associated with ischemia, reperfusion, inflammation, cortical-cytoskeleton abnormalities, bilayer-disruptin

Antimicrobial peptides at (lipid) interfaces: Insights from monolayer models.

Antimicrobial peptides (AMPs) are key effectors of innate immunity that, beyond their canonical activity, exhibit promising therapeutic potential against cancer and cellular senescence. Their efficacy relies on selective membrane disruption driven by specific lipid signatures, yet quantifying these interactions in complex bilayer systems remains challenging. Lipid monolayers serve as powerful reductionist models to isolate the physicochemical determinants of this selectivity, effectively mimicking the outer leaflet of bacterial, cancerous, or senescent membranes. This review provides a critica

Curvature dynamics of alpha-synuclein familial Parkinson disease mutants: molecular simulations of the micelle- and bilayer-bound forms.

Alpha-synuclein remains a protein of interest due to its propensity to form fibrillar aggregates in neurodegenerative disease and its putative function in synaptic vesicle regulation. Herein, we present a series of atomistic molecular dynamics simulations of wild-type alpha-synuclein and three Parkinson disease familial mutants (A30P, A53T, and E46K) in two distinct environments. First, in order to match recent NMR experiments, we have simulated each protein bound to an SDS detergent micelle. Second, in order to connect more closely to the true biological environment, we have simulated the pro

Lanthanide chelates as bilayer alignment tools in NMR studies of membrane-associated peptides.

Theequimolar complex, consisting of the lipid-like, amphiphilic chelating agent 1,11-bis[distearylamino]-diethylenetriamine pentaacetic acid (DTPA-18) and Tm(3+), is shown by deuterium ((2)H) NMR to be useful in aligning bicelle-like model membranes, consisting of dimyristoylphosphatidylcholine (DMPC) and dihexanoylphosphatidylcholine (DHPC). As shown previously (1996, R. S. Prosser et al., J. Am. Chem. Soc. 118, 269-270), in the absence of chelate, the lanthanide ions bind loosely with the lipid phosphate groups and confer the membrane with a sufficient positive magnetic anisotropy to result

Amantadine-induced reorganization of model SARS-CoV-2 lipid envelopes.

Amantadine attracted renewed interest during the COVID-19 pandemic because of its known antiviral activity against influenza A. In this work, we investigated how amantadine affects a simplified model of the SARS-CoV-2 lipid envelope composed of DOPC : DMPS : PI (50 : 35 : 15), as well as monolayers of the individual lipids: DOPC, DMPS and PI. Langmuir experiments showed that the effect of the drug depends on the lipid type. In DOPC and PI monolayers, amantadine increased the area per molecule, suggesting its incorporation into more fluid layers. In contrast, in more compact DMPS monolayers, st

Unraveling How Membrane Nanostructure Changes Impact the Eye Irritation of Nonionic Alkyl Ethoxylate Surfactants.

Nonionic surfactants used in agri-spraying processes may cause varying degrees of corneal irritation when they come in direct contact with farmers' eyes, and the exact irritations are thought to be determined by how surfactants interact with corneal cell membranes. However, how nonionic surfactants interact with cell membranes at the molecular and nano levels remains largely unexplored. In this study, the interactions between nonionic surfactants (alkyl ethoxylate, C12Em) and lipid membranes were examined by membrane permeability measurement, quartz crystal microbalance with dissipation, dual

Miscibility of ternary mixtures of phospholipids and cholesterol in monolayers, and application to bilayer systems.

We investigate miscibility transitions of two different ternary lipid mixtures, DOPC/DPPC/Chol and POPC/PSM/Chol. In vesicles, both of these mixtures of an unsaturated lipid, a saturated lipid, and cholesterol form micron-scale domains of immiscible liquid phases for only a limited range of compositions. In contrast, in monolayers, both of these mixtures produce two distinct regions of immiscible liquid phases that span all compositions studied, the alpha-region at low cholesterol and the beta-region at high cholesterol. In other words, we find only limited overlap in miscibility phase behavio

Mechanosensitive membrane channels in action.

The tension-driven gating process of MscL from Mycobacterium tuberculosis, Tb-MscL, has been addressed at near-atomic detail using coarse-grained molecular dynamics simulations. To perform the simulations, a novel coarse-grained peptide model based on a thermodynamic parameterization of the amino-acid side chains has been applied. Both the wild-type Tb-MscL and its gain-of-function mutant V21D embedded in a solvated lipid bilayer have been studied. To mimic hypoosmotic shock conditions, simulations were performed at increasing levels of membrane tension approaching the rupture threshold of the

Differential control of band 3 lateral and rotational mobility in intact red cells.

Measurements of integral membrane protein lateral mobility and rotational mobility have been separately used to investigate dynamic protein--protein and protein-lipid interactions that underlie plasma membrane structure and function. In model bilayer membranes, the mobilities of reconstituted proteins depend on the size of the diffusing molecule and the viscosity of the lipid bilayer. There are no direct tests, however, of the relationship between mechanisms that control protein lateral mobility and rotational mobility in intact biological membranes. We have measured the lateral and rotational

Membrane protein structural validation by oriented sample solid-state NMR: diacylglycerol kinase.

The validation of protein structures through functional assays has been the norm for many years. Functional assays perform this validation for water-soluble proteins very well, but they need to be performed in the same environment as that used for the structural analysis. This is difficult for membrane proteins that are often structurally characterized in detergent environments, although functional assays for these proteins are most frequently performed in lipid bilayers. Because the structure of membrane proteins is known to be sensitive to the membrane mimetic environment, such functional as…

Membrane interactions of non-membrane targeting antibiotics: The case of aminoglycosides, macrolides, and fluoroquinolones.

Numerous antibiotics are known to target intracellular pathways, such as protein translation or DNA replication. Membrane transporters typically regulate drug uptake; however, little is known about direct interactions between these antibiotics and the cell membranes. Here, we studied the interactions between different aminoglycosides (kanamycin, gentamicin, streptomycin, neomycin), macrolides (azithromycin, clarithromycin, erythromycin), and fluoroquinolones (ciprofloxacin, levofloxacin) with bacterial membrane mimics to determine drug partitioning and potential drug-induced membrane disruptio

Human cells and cell membrane molecular models are affected in vitro by chlorpromazine.

This study presents evidence that chlorpromazine (CPZ) affects human cells and cell membrane molecular models. Human SH-SY5Y neuroblastoma cells incubated with 0.1 mM CPZ suffered a decrease of cell viability. On the other hand, phase contrast microscopy observations of human erythrocytes indicated that they underwent a morphological alteration as 1 microM CPZ changed their discoid normal shape to stomatocytes, and to hemolysis with 1 mM CPZ. X-ray diffraction experiments performed on dimyristoylphosphatidylcholine (DMPC) and dimyristoylphosphatidylethanolamine (DMPE) bilayers, classes of the

Control of the depth of molecules within membranes by polar groups: determination of the location of anthracene-labeled probes in model membranes by parallax analysis of nitroxide-labeled phospholipid induced fluorescence quenching.

The location of anthracene-labeled molecules incorporated into model membranes was measured by fluorescence quenching. The depth of the anthracene group was calculated from the degree of quenching by lipids carrying a nitroxide at different depths, using the parallax analysis (Chattopadhyay & London (1987) Biochemistry 26, 39-45). A series of anthracene derivatives was examined in order to determine what polar functional groups would anchor at the membrane surface, and at what depth anchoring would occur. An anthracene with only a methyl group was not anchored at the membrane surface, but deri

Concentration-Dependent Modes of Melittin-Induced Membrane Destabilization: An Integration of Experimental and Computational Tools.

Melittin is a well-known antimicrobial peptide whose mechanism of action is still not entirely understood. In this study, we combine complementary experimental techniques with molecular dynamics simulations to determine melittin-membrane interactions at different scales. Using 1,2-dioleoyl-sn-glycero-3-phosphoethanolamine (DOPE): 1,2-dipalmitoyl-sn-glycero-3-phospho-(1'-rac-glycerol) (DPPG) membranes that mimic the Escherichia coli inner membrane, this integrative approach reveals a clear transition in melittin's mode of action with increasing peptide loading. At low concentrations, melittin p

Force Field and Membrane Patch Size Effects on Atomistic Models of Aquaporin-7.

Molecular dynamics (MD) simulations are a powerful tool for characterizing membrane-protein dynamics, yet their predictive accuracy critically depends on the choice of force field and membrane representation. Here, we present a systematic benchmark of the AMBER 14SB and CHARMM 36 m force fields across multiple bilayer sizes, using human aquaporin-7 (aquaglyceroporin-7; hAQP7) as a representative membrane protein system. Both force fields maintained global structural integrity, but differed markedly in their dynamic profiles: CHARMM 36 m sampled a broader conformational space and produced more

Cold-selective topological bias and the emergence of the first membranes.

The emergence of the first membranes requires more than the synthesis of amphiphiles; it requires a mechanism that stabilizes bilayer-forming geometries within chemically diverse prebiotic mixtures. We present a minimal thermodynamic model coupling amphiphile topology to self-assembly, incorporating packing, curvature, and steric contributions. Changes in temperature alter the balance between ordering and disorder within amphiphile aggregates, thereby modifying the relative stability of lamellar, micellar, and dispersed states. The model identifies a bounded temperature regime in which geometr
