Untargeted plasma lipidomic profiling associated with pediatric metabolic dysfunction-associated steatohepatitis (MASH).

BACKGROUND: Altered lipid metabolism is central to the pathogenesis of MASH, yet lipidomic data in pediatric populations are limited. We conducted a cross-sectional analysis using high-resolution lipidomics to identify lipid alterations associated with pediatric MASH in youth enrolled in NASH CRN studies. METHODS: Clinical assessments included anthropometrics, lipid panels, liver enzymes, and markers of glucose metabolism. MASLD status was determined by liver biopsy or MRI-PDFF. Plasma untargeted lipidomics was performed using UHPLC-MS/MS. Linear models identified differentially abundant lipid…

Engineering Nanoplatforms for Alzheimer's Disease Detection via Biomolecular Corona Proteomic and Lipidomic Profiling.

The escalating global prevalence of Alzheimer's disease (AD) requires the development of sensitive, non-invasive diagnostic strategies capable of detecting pathological changes previous the clinical onset. By exploiting the spontaneous formation of the biomolecular corona (BC) around nanoparticles (NPs), it's possible to capture a rich, disease-specific fingerprint from the plasma that reflects systemic alterations often invisible to traditional diagnostic assays. In the present study, we demonstrate the efficacy of an innovative nanotechnological platform utilizing a synergistic dual-silica N

Comprehensive Lipidomic Analysis of Streptomyces by NPLC-HRMS Reveals a Previously Unknown Phospholipid Class.

Studies on lipids in Streptomyces bacteria are limited and primarily rely on thin-layer chromatography (TLC) and off-line mass spectrometry. In this article, we present an in-depth analysis of Streptomyces lipids, performed using normal-phase liquid chromatography coupled with high-resolution mass spectrometry (NPLC-HRMS), applied to various Streptomyces strains in recent collaborations. All identified lipid classes detected in the extracts are described. Numerous mass spectra are provided, offering a detailed view of the distribution of molecular species and their structural characteristics.

PFOA Exposure Elicits Quantitative Lipidomic Changes in the Pancreas in a Mouse Model of Pancreatic Cancer.

Pancreatic cancer is the fourth leading cause of cancer deaths in the US and predicted to rise to second by 2030. Several risk factors have been identified as potential contributors to pancreatic cancer development including lifestyle factors and long-term exposure to occupational and environmental carcinogens. In mice, exposure to perfluorooctanoic acid (PFOA) leads to diabetic outcomes and pancreatic cancer, while epidemiologic studies have linked PFOA exposure to type 1, type 2, and gestational diabetes, obesity, and pancreatic cancer. While multiple studies have evaluated changes in serum

Lipidomic profiling reveals medium-chain acylcarnitines alterations as metabolic signatures in IgA nephropathy.

Immunoglobulin A nephropathy (IgAN), the most common primary glomerulonephritis worldwide, exhibits highly variable clinical progression, ranging from asymptomatic urinary abnormalities with potential for spontaneous resolution to progressive kidney failure over 10–20 years. The limited understanding of IgAN pathogenesis continues to constrain the development of effective therapeutic strategies. To elucidate key pathogenic mechanisms, we performed both untargeted and quantitative lipidomic profiling in patients with IgAN. Our results revealed significant disruptions in carnitine metabolism, pa

Eicosapentaenoic acid ethyl ester, cardiac metabolomic and lipidomic signatures, and cardioprotection in myocardial infarction.

BACKGROUND AND AIMS: The cardiovascular benefits of eicosapentaenoic acid ethyl ester (EPA-E) in MACE reduction might extend beyond its triglyceride (TG)-lowering effects. However, the mechanism behind remains unknown. This study investigated whether EPA-E exerts cardioprotection in the setting of myocardial infarction (MI). METHODS: Hypertriglyceridaemia (HTG) was induced in male rats by a high-sucrose diet (>175 mg·dL-1; baseline). Hypertriglyceridaemia rats were administered for 2 weeks high-dose EPA-E supplementation (HTG + EPA-E; n = 21) or placebo (HTG; n = 27) in combination with the d

Postprandial extracellular vesicle lipidomes: links between dietary fat quality and cardiometabolic signalling.

Dietary fat intake acutely induces a dynamic postprandial state characterized by transient elevations in triacylglycerol-rich lipoproteins, circulating non-esterified fatty acids, and lipid mediators, superimposed on habitual diet and individual metabolic status. Within this context, extracellular vesicles (EVs) have emerged as lipid-bilayer nanoparticles whose lipid composition may reflect cellular lipid handling and contribute to interorgan signaling. This narrative review integrates mechanistic evidence, omics studies, and human feeding trials to examine how dietary lipids and postprandial

Air pollution and hypertension in rural versus urban children: Lipidomic insights into PM2.5 impacts.

Fine particulate matter and its impact on blood pressure (BP) in children remain a concern, with the role of lipid metabolism as a potential mediator not fully elucidated. We conducted a cohort study of 2239 urban subjects and 2194 rural subjects at baseline in China from 2014 to 2024 and a nested case-control study with lipidomics analyses. Analysis results showed that higher fine particulate matter smaller than 2.5 μm (PM2.5) exposure associated with high density lipoprotein cholesterol (HDL-C), non-HDL-C, and higher systolic blood pressure (SBP), partially mediated by HDL-C/non-HDL-C change…
