A Ca(2+) threshold for induction of spike-timing-dependent depression in the mouse striatum.

The striatum is the principal input nucleus of the basal ganglia, receiving glutamatergic afferents from the cerebral cortex. There is much interest in mechanisms of synaptic plasticity in the corticostriatal synapses. We used two-photon microscopy and whole-cell recording to measure changes in intracellular calcium concentration ([Ca(2+)](i)) associated with spike-time-dependent plasticity in mouse striatum. Uncaging glutamate adjacent to a dendritic spine caused a postsynaptic potential at the soma and a rise in spine [Ca(2+)](i). Action potentials elicited at the soma raised both dendrite a

Sparse Coding and Temporal Pattern Learning Co-Mediated by Dual Spike-Timing-Dependent Plasticity in a Multilayer Excitatory-Inhibitory Spiking Network.

Excitatory-inhibitory (E-I) local circuits play a central role in synaptic plasticity and neural coding, yet their multilayer learning dynamics remain poorly understood. We constructed a multilayer feedforward spiking neural network with intra-layer E-I connectivity, using Izhikevich neurons to model regular spiking (RS) and fast spiking (FS) cells, and examined cooperative learning under excitatory and inhibitory spike-timing-dependent plasticity (eSTDP and iSTDP). FS-mediated lateral inhibition alleviates the long-term depression bias arising from RS firing rate adaptation via winner-take-al

hUC-MSCs transplantation promotes neurological recovery by ameliorating spinal cord injury and hippocampal synaptic remodeling in female mice.

INTRODUCTION: Spinal cord injury (SCI) induces not only local spinal cord pathology but also distal brain dysfunction, causing multi-dimensional impairments in motor, emotional, and cognitive functions. Human umbilical cord mesenchymal stem cells (hUC-MSCs) transplantation has emerged as a promising therapeutic strategy for SCI, but whether it concurrently improves pathological alterations in both the spinal cord and brain remains unclear. METHODS: After establishing a mouse model of spinal cord contusion, hUC-MSCs were transplanted intrathecally starting 7 d post-injury and administered week

Systemic pharmacological suppression of neural activity reverses learning impairment in a mouse model of Fragile X syndrome.

The enhancement of associative synaptic plasticity often results in impaired rather than enhanced learning. Previously, we proposed that such learning impairments can result from saturation of the plasticity mechanism (Nguyen-Vu et al., 2017), or, more generally, from a history-dependent change in the threshold for plasticity. This hypothesis was based on experimental results from mice lacking two class I major histocompatibility molecules, MHCI H2-Kb and H2-Db (MHCI KbDb-/-), which have enhanced associative long-term depression at the parallel fiber-Purkinje cell synapses in the cerebellum (P

Nicotinamide Adenine Dinucleotide Phosphate Oxidases Are Everywhere in Brain Disease, but Not in Huntington's Disease?

Huntington's disease (HD) is an inherited neurodegenerative disorder characterized by neuronal loss and tissue atrophy mainly in the striatum and cortex. In the early stages of the disease, impairment of neuronal function, synaptic dysfunction and white matter loss precedes neuronal death itself. Relative to other neurodegenerative diseases such as Alzheimer's and Parkinson's disease and Amyotrophic Lateral Sclerosis, where the effects of either microglia or NADPH oxidases (NOXs) are recognized as important contributors to disease pathogenesis and progression, there is a pronounced lack of inf

Wogonin Mitigates Depression by Inhibiting TNF-α/TNFR1/CXCL1 Signalling-Mediated Astrocyte Activation.

Wogonin exhibits notable anti-inflammatory and neuroprotective effects. Current study aimed to investigate the antidepressant potential of wogonin and its underlying mechanisms through in vitro and in vivo experiments. In vitro, wogonin attenuated lipopolysaccharide (LPS)-induced astrocyte activation. Furthermore, wogonin inhibited the TNF-α/TNFR1/CXCL1 signalling axis, as evidenced by decreased protein levels of TNF-α and CXCL1 and reduced interaction between TNF-α and its receptor TNFR1. The addition of recombinant TNF-α (r-TNF-α) blocked these inhibitory effects of wogonin. In vivo, in a ch

Beyond surgery: Repurposing anesthetics for treatment of central nervous system disorders.

The development of new drugs is a complex, expensive, and time-consuming process, often fraught with a high likelihood of failure. Amid these obstacles, drug repurposing, which identifies new therapeutic applications for already existing medications, offers a more economical and time-saving approach, particularly in the challenging field of neurological and psychiatric disorders. This narrative review explores both preclinical and clinical studies to examine the potential of anesthetics such as ketamine, nitrous oxide, isoflurane, sevoflurane, propofol, dexmedetomidine, and sodium oxybate in t

N-3 PUFA supplementation in adulthood modulates diet-induced depressive-like phenotype in female rats.

Low consumption of omega-3 polyunsaturated fatty acids (n-3 PUFA) during development has been linked to increased risk of developing depressive symptoms. The present study assesses the influence of chronic n-3 PUFA supplementation in a rodent model of depressive-like phenotype induced by long-life depletion of n-3 PUFA in the diet. These behavioural and biological consequences already start to become apparent in adolescence and tend to worsen if the n-3 PUFA deficiency is prolonged. Here, we investigated whether the reintroduction of n-3 PUFA at a later stage of development can reverse these a

Neuroinflammation and Mental Health in Multiple Sclerosis and Autoimmune Encephalitis: Bridging Biological Mechanisms and Psychosocial Factors.

Autoimmune neuroinflammatory diseases, including multiple sclerosis (MS) and autoimmune encephalitis (AE), are characterized by dysregulated immune activity within the central nervous system that leads to both neurological and psychiatric symptoms. While MS is typically a chronic condition indicated by demyelination and progressive neurodegeneration, AE often presents more acutely through antibody-mediated disruption of synaptic function. Despite differences in disease course and mechanisms, both conditions highlight the significant impact of immune-mediated processes on cognition, mood, and b

Object Motion Detection Enabled by Reconfigurable Neuromorphic Vision Sensor under Ferroelectric Modulation.

Increasing the demand for object motion detection (OMD) requires shifts of reducing redundancy, heightened power efficiency, and precise programming capabilities to ensure consistency and accuracy. Drawing inspiration from object motion-sensitive ganglion cells, we propose an OMD vision sensor with a simple device structure of a WSe2 homojunction modulated by a ferroelectric copolymer. Under optical mode and intermediate ferroelectric modulation, the vision sensor can generate progressive and bidirectional photocurrents with discrete multistates under zero power consumption. This design enable

Pathophysiology of Cerebellar Degeneration in Mitochondrial Disorders: Insights from the Harlequin Mouse.

By means of a proteomic approach, we assessed the pathways involved in cerebellar neurodegeneration in a mouse model (Harlequin, Hq) of mitochondrial disorder. A differential proteomic profile study (iTRAQ) was performed in cerebellum homogenates of male Hq and wild-type (WT) mice 8 weeks after the onset of clear symptoms of ataxia in the Hq mice (aged 5.2 ± 0.2 and 5.3 ± 0.1 months for WT and Hq, respectively), followed by a biochemical validation of the most relevant changes. Additional groups of 2-, 3- and 6-month-old WT and Hq mice were analyzed to assess the disease progression on the pro

Interferon therapy and its association with depressive disorders - A review.

Interferons (IFNs) are important in controlling the innate immune response to viral infections. Besides that, studies have found that IFNs also have antimicrobial, antiproliferative/antitumor and immunomodulatory effects. IFNs are divided into Type I, II and III. Type I IFNs, in particular IFN-α, is an approved treatment for hepatitis C. However, patients developed neuropsychological disorders during treatment. IFN-α induces proinflammatory cytokines, indoleamine 2,3-dioxygenase (IDO), oxidative and nitrative stress that intensifies the body's inflammatory response in the treatment of chronic

MicroRNAs: Novel Mechanism Involved in the Pathogenesis of Microwave Exposure on Rats' Hippocampus.

Microwave-induced adverse health outcomes have been gaining much attention in recent years. The hippocampus is sensitive and vulnerable to microwave exposure. Studies from our group and others showed that microwave-induced structural and functional injury of hippocampus, accompanied with alteration of gene and protein expression. It has been demonstrated that microRNAs (miRNAs) were involved in the physiological and pathological processes of brain. In this study, the miRNAs expression profiles of microwave-exposed hippocampus were detected by microarray analysis and verified by real-time polym

Estrogen and hippocampal synaptic plasticity.

During the past several years, there has been increasing interest in the effects of estrogen on neural function. This enthusiasm is driven, in part, by the results of early clinical studies suggesting that estrogen therapy given after menopause may prevent, or at least delay, the onset of Alzheimer's disease in older women. However, later clinical trials of women with probable Alzheimer's disease had contrary results. Much of the current research related to estrogen and brain function is focused in two directions. One involves clinical studies that examine the potential of estrogen in protecti

Caspase-3 in the central nervous system: beyond apoptosis.

Caspase-3 has been identified as a key mediator of neuronal programmed cell death. This protease plays a central role in the developing nervous system and its activation is observed early in neural tube formation and persists during postnatal differentiation of the neural network. Caspase-3 activation, a crucial event of neuronal cell death program, is also a feature of many chronic neurodegenerative diseases. This traditional apoptotic function of caspase-3 is challenged by recent studies that reveal new cell death-independent roles for mitochondrial-activated caspase-3 in neurite pruning and

Acute aerobic exercise is associated with a biphasic, temporally ordered shift in serum extracellular vesicle microRNA expression in major depressive disorder.

INTRODUCTION: Acute aerobic exercise (AAE) is known to reduce symptoms of Major Depressive Disorder (MDD), yet the underlying biological mechanisms remain poorly understood. Here, we profiled microRNA (miRNA) expression within the serum extracellular vesicle (EV) enriched fraction (hereafter 'EV-enriched miRNA') of individuals diagnosed with MDD at multiple timepoints: before, during (mid), immediately after, and 75 min following a single 30-min session of moderate aerobic exercise. By characterizing the temporal dynamics of EV-enriched miRNA responses to AAE, our objective was to uncover mole

Differential induction of LTP and LTD is not determined solely by instantaneous calcium concentration: an essential involvement of a temporal factor.

Two opposite types of synaptic plasticity in the CA1 hippocampus, long-term potentiation (LTP) and long-term depression (LTD), require postsynaptic Ca2+ elevation. To explain these apparently contradictory phenomena, the current view assumes that a moderate postsynaptic increase in Ca2+ leads to LTD, whereas a large increase leads to LTP. No detailed study has so far been attempted to investigate whether the instantaneous Ca2+ elevation level differentially induces LTP or LTD. We therefore used low-frequency (1 Hz) stimulation of Schaffer collateral/commissural fibers in rat hippocampal slices

SSRIs and SNRIs (SRI) in Pregnancy: Effects on the Course of Pregnancy and the Offspring: How Far Are We from Having All the Answers?

Serotonin has important roles in the development of the brain and other organs. Manipulations of synaptic serotonin by drugs such as serotonin reuptake inhibitors (SRI) or serotonin norepinephrine reuptake inhibitors (SNRI) might alter their development and function. Of interest, most studies on the outcome of prenatal exposure to SRI in human have not found significant embryonic or fetal damage, except for a possible, slight increase in cardiac malformations. In up to a third of newborns exposed to SRI, exposure may induce transient neonatal behavioral changes (poor neonatal adaptation) and i
