[Value of serum galactomannan assay in the diagnosis of pediatric invasive pulmonary aspergillosis].

OBJECTIVE: Galactomannan (GM) is a major aspergilli cell-wall constituent released into circulation during the early stage of invasive disease, and can be detected. Many studies suggest that serum galactomannan assay has an excellent sensitivity and specificity for the early diagnosis of adult invasive aspergillosis (IA). However, there have been few studies on serum galactomannan assay in pediatric patients. Therefore, we evaluate the value of serum galactomannan assay in the diagnosis of pediatric invasive pulmonary aspergillosis in this study. METHOD: Blood samples were obtained from 88 ch

Rheological properties and sugar composition of locust bean gum from different carob varieties (Ceratonia siliqua L.).

The seeds of the main Italian carob varieties, Latinissima and Tantillo, and those of two selected accessions of Latinissima were evaluated in terms of yield, rheological properties, and sugar composition of the endosperm (LBG). The separation of the seed components in Latinissima and its seedlings yielded meanly 52.2% gum, 17.4% germ, and 30.5% tegument, whereas Tantillo furnished a lower gum yield (38.5%) and a higher yield of tegument (45.8%). The viscosity of 1% LBG aqueous solutions was measured at different shear rates (3-60 rpm), pH values (3.0-6.0), and temperatures (10-60 degrees C).

Bulking agents in the treatment of obesity.

The effect of two bulk-forming appetite depressants, methylcellulose and gum guar, on food intake has been studied in 11 (4 male, 7 female) healthy volunteers--3 of whom were overweight. 10 g of active ingredient were given in two equal doses daily for periods of 1 week of each agent. Individually weighed dietary intakes were obtained over four consecutive weeks; the first and third weeks were without medication. Both gum guar and methylcellulose were equally effective in reducing appetite by 10%.

Redox-stratified macromolecule degradation supports microbial survival in the oligotrophic Kermadec Trench sediments.

Kermadec Trench is a hadal ecosystem in the South Pacific with the water depth 10,047 m. The trench bottom harbors a highly active and populated microbial community, despite the surface sediment is characterized as extremely pressurized, oligotrophic and with low oxygen concentration. It is intriguing, as well as technically challenging, to investigate the microbial adaptation strategies therein. Here we performed the in situ RNA fixation on sediment samples with the assistant of Fendouzhe manned submersible, to approach natural status of microbial metabolisms on both genomic and transcriptomi

Comprehensive structural analysis and immunostimulant activity of complex polysaccharides from dietary fibers of biribá (Annona mucosa) pulp, a tropical fruit from Amazon region.

Tropical fruits are an important and yet underexplored source of dietary fibers, and biribá (Annona mucosa), a fruit native to the Amazon region, remains poorly characterized in this regard. In this study, soluble (SDF) and insoluble dietary fiber (IDF) fractions from biribá fruit pulp were extracted, fractionated, and structurally characterized using a combination of enzymatic-gravimetric methods, chromatographic techniques, monosaccharide composition, linkage analysis, and NMR spectroscopy. Six distinct polysaccharides were identified. The SDF fraction yielded a highly methyl-esterified (DE

Use of narsoplimab for eculizumab-refractory adult transplant-associated thrombotic microangiopathy (TA-TMA).

Transplant-associated thrombotic microangiopathy (TA-TMA) is a fatal complication associated with hematopoietic stem cell transplant (HSCT). Endothelial dysfunction and complement activation cause consumptive thrombocytopenia with intravascular hemolysis, resulting in end-organ damage, especially to the kidneys and lungs. There are no U.S. Food and Drug Administration (FDA)-approved agents for TA-TMA, although Eculizumab is the most commonly used agent to treat TA-TMA. Patients who do not respond to Eculizumab have a dismal prognosis, with reported mortality up to 80%. Narsoplimab, a mannan-bi

Insights on molecular drivers of phenotypic plasticity in Candidozyma auris.

Candidozyma auris (or Candida auris) is the first fungal species declared as a global health threat by the U.S. Center for Disease Control and Prevention (CDC). Its high environmental persistence, immune evasion, phenotypic heterogeneity, and multidrug resistance are major contributors to its high mortality rate. Under diverse environmental conditions, C. auris exhibits marked phenotypic plasticity, switching from non-aggregative to aggregative form and, in some contexts, pseudohyphal-like morphotypes. However, the significance of this heterogeneity in pathogenesis and drug tolerance remains p

Effects of Dietary Sterol-Enriched Yeast Hydrolysate on Ovarian Development, Reproductive Endocrine Status, Immune-Inflammatory Responses, Antioxidant Indices, and Egg Quality in Late-Laying Yellow-Feathered Broiler Breeder Hens.

This study evaluated graded dietary supplementation with sterol-enriched yeast hydrolysate (SYH) in late-laying yellow-feathered broiler breeder hens. A total of 720 55-week-old hens received 0, 0.2, 0.5, or 0.8 g/kg of SYH for six weeks after a one-week adaptation period. SYH is a composite product containing sterols, mannan, nucleotides, and amino nitrogen; therefore, the observed effects cannot be attributed to sterols alone. Eggshell strength increased at 0.5 and 0.8 g/kg, whereas fertility and hatchability were unaffected. The 0.5 g/kg group showed the highest ovarian index values and the

Polysaccharide hydrogels as immunologically active vaccine depots for cancer immunotherapy.

Cancer vaccines aim to elicit durable, antigen-specific antitumour immunity, yet clinical translation remains constrained by inefficient antigen retention, weak innate immune activation, and poor spatiotemporal coordination of immune priming. Biomaterial-based vaccine depots, particularly hydrogels incorporating natural polysaccharides, can address these barriers by creating immune-instructive microenvironments that support localised and sustained antigen delivery. Polysaccharide-based hydrogels combine structural support, tuneable release kinetics, and intrinsic immunomodulatory properties th

The First Report on the Structure of Polysaccharide Surface Antigens of the Clinical Klebsiella oxytoca 0.062 Strain and the Contribution in the Serological Cross-Reactions.

Klebsiella oxytoca bacilli co-form the human intestinal microbiota, but in favorable conditions, they may also affect immunocompromised individuals, causing urinary tract infections, bacteremia, or antibiotic-associated hemorrhagic colitis. The growing numbers of clinical outbreaks of K. oxytoca infections make these bacteria an emerging pathogen, which is still masked by the predominant K. pneumoniae isolates. Thus, it is very important to advance knowledge on K. oxytoca pathogenicity. This work aims to characterize a urine isolate, K. oxytoca 0.062, from central Poland, which appears to pres

Structural characteristics of two polysaccharides from L. fumosum (Pers.) P.D. Orton. and their immunomodulatory effects on RAW264.7 macrophages.

Two polysaccharides, LFP-I and LFP-II, were isolated from L. Fumosum mushroom, and their structures and immunostimulatory effects were investigated. Structural analysis revealed that LFP-I had a molecular weight of 13,288 Da and consisted of fucose (13.12 %), mannose (15.75 %), glucose (46.46 %) and galactose (24.67 %). LFP-II, with a molecular weight of 13,288 Da, was primarily composed of glucose. Bond structure analysis and nuclear magnetic resonance (NMR) characterization confirmed that LFP-I possessed an α-D-(1 → 6)-linked galactan backbone with double substitutions at the O-2 and O-3 pos

From ASCA breakthrough in Crohn's disease and Candida albicans research to thirty years of investigations about their meaning in human health.

Anti-Saccharomyces cerevisiae antibodies (ASCA) are human antibodies that can be detected using an enzyme-linked immunosorbent assay involving a mannose polymer (mannan) extracted from the cell wall of the yeast S. cerevisiae. The ASCA test was developed in 1993 with the aim of differentiating the serological response in two forms of inflammatory bowel disease (IBD), Crohn's disease and ulcerative colitis. The test, which is based on the detection of anti-oligomannosidic antibodies, has been extensively performed worldwide and there have been hundreds of publications on ASCA. The earlier studi

Proteomics Insights into Medullary Sponge Kidney Disease: Review of the Recent Results of an Italian Research Collaborative Network.

BACKGROUND: Medullary sponge kidney (MSK) disease is a rare and neglected congenital condition typically associated with nephrocalcinosis/nephrolithiasis, urinary concentration defects, and cystic anomalies in the precalyceal ducts that, although sporadic in the general population, is relatively frequent in renal stone formers. The physiopathologic mechanism associated with this disease is not fully understood, and omics technologies may help address this gap. SUMMARY: The aim of this review was to provide an overview of the current state of the application of proteomics in the study of this

TSC1 Suppresses Macrophage Necroptosis for the Control of Infection by Fungal Pathogen Candida albicans.

Candida albicans is the most common, opportunistic human fungal pathogen whose complex interplay with the host innate immune system remains incompletely understood. In this study, we revealed that infection macrophages with C. albicans triggers prominent cell death, which is largely attributed to the RIPK3/MLKL-mediated necroptosis. Our results further demonstrated that the TSC1-mTOR pathway plays a pivotal role in the control of macrophage necroptosis upon engaging the Dectin-1/2 and TLR-2/4 pathways through fungal components β-glucan/α-mannan or Sel1, respectively. Notably, the rapamycin-sen

Dengue and the Lectin Pathway of the Complement System.

Dengue is a mosquito-borne viral disease causing significant health and economic burdens globally. The dengue virus (DENV) comprises four serotypes (DENV1-4). Usually, the primary infection is asymptomatic or causes mild dengue fever (DF), while secondary infections with a different serotype increase the risk of severe dengue disease (dengue hemorrhagic fever, DHF). Complement system activation induces inflammation and tissue injury, contributing to disease pathogenesis. However, in asymptomatic or primary infections, protective immunity largely results from the complement system's lectin path

Inflammatory Bowel Disease Types Differ in Markers of Inflammation, Gut Barrier and in Specific Anti-Bacterial Response.

Crohn's disease (CD), ulcerative colitis (UC) and inflammatory bowel disease (IBD) associated with primary sclerosing cholangitis (PSC-IBD), share three major pathogenetic mechanisms of inflammatory bowel disease (IBD)-gut dysbiosis, gut barrier failure and immune system dysregulation. While clinical differences among them are well known, the underlying mechanisms are less explored. To gain an insight into the IBD pathogenesis and to find a specific biomarker pattern for each of them, we used protein array, ELISA and flow cytometry to analyze serum biomarkers and specific anti-microbial B and

Physiologically Relevant Alternative Carbon Sources Modulate Biofilm Formation, Cell Wall Architecture, and the Stress and Antifungal Resistance of Candida glabrata.

Flexibility in carbon metabolism is pivotal for the survival and propagation of many human fungal pathogens within host niches. Indeed, flexible carbon assimilation enhances pathogenicity and affects the immunogenicity of Candida albicans. Over the last decade, Candida glabrata has emerged as one of the most common and problematic causes of invasive candidiasis. Despite this, the links between carbon metabolism, fitness, and pathogenicity in C. glabrata are largely unexplored. Therefore, this study has investigated the impact of alternative carbon metabolism on the fitness and pathogenic attri

Effect of Nicotinamide Against Candida albicans.

Nicotinamide (NAM) has a long history in clinical applications and can be safely used for treating various diseases. In recent years, NAM was found to exhibit antimicrobial activities, inhibiting the growth of Plasmodium falciparum, Mycobacterium tuberculosis, and human immunodeficiency virus (HIV). Here we investigated the activity of NAM against Candida albicans, one of the most prevalent human fungal pathogens. Our results showed that NAM exhibited significant antifungal activity against C. albicans, including fluconazole-resistant isolates. NAM could also effectively suppress biofilm forma

Genetic variations of the bitter taste receptor TAS2R38 are associated with obesity and impact on single immune traits.

SCOPE: Changes in genetic variations affecting the taste receptor, type 2, member 38 (TAS2R38) may identify the interacting mechanism leading to obesity and potential associations with proteins partaking in innate immunity, such as surfactant protein D (SPD) and mannan-binding lectin (MBL). METHODS AND RESULTS: We evaluated haplotypes of the bitter-taste receptor TAS2R38 in an identification sample of 210 women in different weight conditions, including anorexia nervosa and obesity. The association with SPD and MBL was tested in an independent sample picturing general population (n = 534). The

Modeling eye movements in visual agnosia with a saliency map approach: bottom-up guidance or top-down strategy?

Two recent papers (Foulsham, Barton, Kingstone, Dewhurst, & Underwood, 2009; Mannan, Kennard, & Husain, 2009) report that neuropsychological patients with a profound object recognition problem (visual agnosic subjects) show differences from healthy observers in the way their eye movements are controlled when looking at images. The interpretation of these papers is that eye movements can be modeled as the selection of points on a saliency map, and that agnosic subjects show an increased reliance on visual saliency, i.e., brightness and contrast in low-level stimulus features. Here we review thi
