The development of rhythmic 6-sulfatoxymelatonin excretion in urine was studied in healthy full-term and premature infants during the first 12 months of life. There was little evidence of rhythmic 6-sulfatoxymelatonin excretion before 9 to 12 weeks of age in full-term infants. Over this period, excretion increased five to six times compared to the excretion at 6 weeks (08 +/- 103 vs. 2973 +/- 438 pmol/24 h) with the major proportion of the hormone metabolite being excreted between 0200-1000 h. At 24 weeks of age, total 6-sulfatoxymelatonin excretion was 25% of adult levels. Premature infants (…

Melatonin, a hormone from the pineal gland, was tested for its anticonvulsant effects in male gerbils. Daily administration of melatonin (25 micrograms injection-1, s.c.) for ten weeks reduced the number and severity of seizures (total convulsion score: 7.67 +/- 1.83 in controls vs 2.47 +/- 0.90 in melatonin-injected animals, p < 0.05) associated with the injection of the convulsant, pentylenetetrazol (PTZ, 60 mg kg-1, s.c.). However, neither 12 weeks of short photoperiod exposure (LD 10:14) nor biweekly administration of melatonin pellets altered PTZ-induced convulsions. Overall, melatonin-in…

The effect of prolonged, oral melatonin treatment on spontaneous mammary tumor development in female C3H/Jax mice was studied. Melatonin was administered at a dose of 25 micrograms/mouse/day from 21 to 44 days of age and 50 micrograms/mouse/day from day 45 onwards. By the age of 12 months, 62.5% of control animals developed tumors as opposed to 23.1% in the melatonin treated group (P less than 0.02). In both control and treated mice, the thoracic pairs of mammary glands were obviously more susceptible to spontaneous mammary tumor development, as at least 50% of the total tumors developed in th…

The results presented here show clearly that the rapid nocturnal increase in circulating melatonin in sheep is associated with an equally rapid increase in melatonin production in the pineal gland. Pineal NAT activity and NAS concentration also increased under this condition, indicating that NAT activity is an important factor in this process. According to the current model for the regulation of pineal melatonin production in the rat, the large nocturnal increase in NAT activity is the major factor responsible for the daily rhythm in melatonin, as well as for the opposite rhythm in serotonin a…

The author describes how phototherapy may treat seasonal affective disorder (SAD) by stimulating the production of retinal dopamine and suppressing the production of retinal melatonin. This hypothesis offers a framework in which winter-induced retinal dopamine deficiency or retinal melatonin overactivity may cause SAD and by which light reverses this syndrome. This hypothesis is consistent with recent data indicating that phototherapy in SAD acts specifically through the eyes.

The pineal gland has captured man's attention as early in recorded history as the Greeks when philosophers considered it the "seat of the so … he 'fine tuning' and integrating of various neural and endocrine functions. The secretion of pineal melatonin has been shown to decline progressively with age. Recent hypotheses of aging have suggested that cumulative neuronal insults associated with free radical production may be associated with the process of aging. There is evidence to suggest that melat …

The influence of a single dose of melatonin administered to pinealectomized and sham operated rats at different times of day on serum T3, T4 concentrations was investigated. It was found that such melatonin influence depends upon a time of day. Both pinealectomy and exogenous melatonin did not influence the thyroid activity in the morning. The most remarkable pineal effect on the thyroid appeared during the night.

A daily rhythm in melatonin concentration in Harderian glands (HG) of female golden hamsters is described and is characterized by significant reductions following lights-on. Concentrations in HG of intact or pinealectomized males are low and consistent over a 24-hour period. Castration of males is accompanied by increased levels of melatonin in the HG to approach those of females. When castrated males are exposed to short photoperiods, however, melatonin levels are typically low. Levels of serotonin (5-HT), 5-hydroxytryptophan (5-HTP), and 5-hydroxyindoleacetic acid (5-HIAA) are extremely vari…

The annual cycle of reproductive activity in the Syrian hamster, Mesocricetus auratus, is the result of interaction between seasonal changes in daylength (photoperiodism) and seasonal changes in responsiveness to daylength (seasonality). The present experiment was designed to investigate the role of the pineal gland and its hormone, melatonin, in the alternation of seasonality (scotosensitivity and scotorefractoriness). Male hamsters were maintained on short daylengths (10L:14D) to establish scotorefractoriness, and then they were transferred to long daylengths (14L:10D) for conversion to scot…

Plasma melatonin circadian profiles were investigated in a group of 4 patients with anorexia nervosa and 4 healthy regularly cycling women. There were no differences in the mean age of both groups, whereas the anorexia nervosa patients had lower mean body weight (37.8 +/- 2.0 vs 57.0 +/- 4.9 kg) and body mass index (13.9 +/- 1.1 vs 20.8 +/- 2.0). Samples were collected every 2 h and plasma melatonin was measured by using a RIA with an iodinated tracer. Anorexia nervosa patients exhibited higher diurnal (60.7 +/- 1.8 vs 25.4 +/- 1.72 pmol/l, P less than 0.02) and nocturnal (419.2 +/- 37.4 vs 10…

The authors investigated the melatonin response to 5-methoxypsoralen in 39 healthy adult and elderly subjects and in 13 demented, 13 depressed, and 13 schizophrenic inpatients. Subjects' plasma melatonin levels were evaluated before they took single, oral 40-mg doses of 5-methoxypsoralen and 3 hours afterward. The preliminary results indicate that the melatonin response of the pineal gland to 5-methoxypsoralen is reduced in demented patients compared to that in healthy elderly subjects. Depressed and schizophrenic patients had a normal melatonin response to the drug. This finding suggests that…

The central role of the pineal gland and its hormone melatonin (MEL) in mammalian photoperiodic responses is discussed in terms of: 1) evidence for the involvement of MEL in photoperiodism, 2) which feature of the MEL secretion profile might be most important for regulating photoperiodic responses, 3) evidence for the modulation of responses to changes in daylength based on previous photoperiod exposure (i.e., photoperiodic history) and 4) how the MEL signal might be processed at its target sites to elicit physiological responses.

The direction of change in daylength provides the seasonal time cue for the timing of puberty in many mammalian species. The pattern of melatonin secretion from the pineal gland transduces the environmental light-dark cycle into a signal influencing the neuroendocrine control of sexual maturation. The change in duration of nocturnal melatonin secretion is probably the key feature of the melatonin signal which conveys daylength information. This information may also be used by neuroendocrine axes controlling seasonal changes in pelage colour, growth and metabolism. The mechanism of action of me…

Melatonin was measured by radioimmunoassay in several genetic strains of Japanese quail. Plasma melatonin (PM), measured at the nighttime peak, was highest in wild type quail reared on a diurnal lighting schedule; this PM peak was suppressed in continuous light. Albino quail had low melatonin levels, whether reared under diurnal conditions or in continuous light. Ocular melatonin was also suppressed in albinos and in dilute mutants. At midday sampling, melatonin was only half as high in albinos as in wild types. Intraocular pressure (IOP, daytime readings) was uniformly low in diurnal birds an…

The stimulatory effect of serotonin and the inhibitory effect of melatonin on iodine incorporation by thyroid follicular cells are both inhibited by the serotonin antagonists methysergid and cyphroheptadine. Serotonin and melatonin can mutually prevent each other's action. A collaboration of melatonin and serotonin in the regulation of thyroid function is implied from the experimental observations.

Biomarkers of urinary melatonin represent an attractive non-invasive method for enhancing breast cancer diagnosis and follow-up, as they reflect combined circadian physiology in a readily accessible and repeatable urine test. There has been a growing interest in this field, driven by the understanding that melatonin plays a role in regulating the endocrine system, influencing sleep patterns, managing circadian rhythms, affecting exposure to night-time light, and impacting the body's response to cancer and its treatment. The urinary analyte 6-sulfatoxymelatonin has become the most viable surrog…

PURPOSE: Anticancer drugs like cyclophosphamide (CTX) cause severe ovarian damage, often leading to infertility, while current fertility preservation methods are invasive and costly. This study investigates whether melatonin (MEL) can protect ovarian function in a CTX-induced premature ovarian failure (POF) mouse model, offering a potential noninvasive and cost-effective fertoprotective approach.
METHODS: A CTX-induced POF mouse model was used to assess MEL's protective effects. Ovarian function was evaluated by analyzing follicle counts, ovarian weight, serum hormone levels, apoptosis marker…
