Voluntary alcohol consumption and plasma beta-endorphin levels in alcohol-preferring rats chronically treated with naltrexone.

It is well known that alcohol consumption leads to an increase in the levels of beta-endorphin (an endogenous opioid), which can contribute to the reinforcing effect of alcohol. Our previous studies have shown that repeated treatment with naltrexone, a nonselective opioid antagonist, results in increased plasma beta-endorphin levels. Ample studies in animals and humans have shown that naltrexone diminishes ethanol consumption. The aim of the present study in alcohol-preferring rats (Warsaw High Preferring; WHP) was to investigate the effect of 10 days of naltrexone treatment (2 mg/kg i.p.) on …

Long-term naltrexone treatment normalizes the pituitary response to gonadotropin-releasing hormone in polycystic ovarian syndrome.

OBJECTIVE: To investigate the influence of opioid system on the exaggerated LH response to GnRH test in the polycystic ovarian syndrome (PCOS). DESIGN: Pituitary stimulation (GnRH 100 micrograms) in a group of PCOS patients under basal condition and after 4 weeks of treatment with naltrexone. RESULTS: In the PCOS group, the naltrexone treatment determines a significant reduction of the LH response (calculated as the area under curve) to GnRH test, with a similar significant decrease of the LH:FSH. CONCLUSION: Naltrexone normalizes in the PCOS group the pituitary response to GnRH test, aboli…

Mitragynine and naltrexone alone and in combination reduce alcohol self-administration in female Sprague Dawley rats.

BACKGROUND: Mitragynine (MG), an alkaloid found in leaves of Mitragyna speciosa (kratom), is used for pain and opioid withdrawal. MG's effects are thought to be mediated through opioid receptors. The mu opioid antagonist naltrexone (NTX), indicated for the treatment of alcohol use disorder (AUD), reduces alcohol self-administration in animals. MG also alters alcohol consumption and alleviates alcohol withdrawal. Therefore, we tested MG, NTX, and both combined on alcohol self-administration in rats. METHODS: MG (0.3-3.0 mg/kg, IP) and NTX (0.3-3.0 mg/kg, IP) alone and the combinations of equal…

A Membrane-Centric Plasma Lipidomic Signature of Response to Long-Acting Naltrexone in Alcohol Use Disorder.

Long-acting naltrexone is a first-line pharmacotherapy for alcohol use disorder (AUD), but clinical response is heterogeneous and the underlying biology remains incompletely understood. In a randomised, double-blind, placebo-controlled trial of naltrexone implants (n = 70), the prespecified primary endpoint-percentage of heavy-drinking days (PHDD) over 24 weeks-favoured naltrexone (median 1.49% vs. 13.39%; p = 0.042). In a prespecified Week-12 mechanistic lipidomics substudy, untargeted liquid chromatography-mass spectrometry (LC-MS) profiling was performed in a responder-enriched subset compr…

A comparative study evaluating the efficacy of naltrexone versus acamprosate as anticraving agents in alcohol use disorder.

BACKGROUND: Alcohol use disorders (AUDs) are characterized by excessive alcohol consumption, negatively impacting health, social life, and economic status. Globally, AUD contributes to significant disease burden, causing millions deaths annually. Despite the availability of treatments, relapse rates remain high. Naltrexone and acamprosate, two FDA-approved anticraving agents, are widely used to prevent relapse. However, comparative studies in Indian populations are limited, prompting this study to assess the efficacy of these drugs in treating AUD. AIM: This study aimed to compare the effecti…

Effect of naltrexone pretreatment on ketamine-induced glutamatergic activity and symptoms of depression: a randomized crossover study.

We investigated the potential role of the opioid system in modulating glutamatergic effects of ketamine administration in major depressive disorder. Twenty-six adults with major depressive disorder participated in a double-blind crossover study, receiving oral placebo or 50 mg naltrexone before an intravenous infusion of 0.5 mg per kg ketamine. Brain glutamatergic activity in the anterior cingulate cortex was measured using functional magnetic resonance spectroscopy and depressive symptoms were assessed with the Montgomery-Åsberg Depression Rating Scale. Naltrexone attenuated the increase in g…

Low-dose naltrexone for the treatment of fibromyalgia: protocol for a double-blind, randomized, placebo-controlled trial.

BACKGROUND: Low-dose naltrexone (LDN) is used widely as an off-label treatment for pain despite limited evidence for its effectiveness. A few small trials with a high risk of bias have investigated the effect of LDN on pain associated with fibromyalgia in women, but larger and more methodologically robust studies are needed. The primary aim of this randomized controlled trial is to investigate if 12 weeks of LDN treatment is superior to placebo in reducing the average pain intensity during the last 7 days in women with fibromyalgia. METHODS: A single-center, permuted block randomized, double-…

Evaluating the Impact of Naltrexone on the Rat Gambling Task to Test Its Predictive Validity for Gambling Disorder.

Gambling Disorder has serious consequences and no medications are currently approved for the treatment of this disorder. One factor that may make medication development difficult is the lack of animal models of gambling that would allow for the pre-clinical screening of efficacy. Despite this, there is evidence from clinical trials that opiate antagonists, in particular naltrexone, may be useful in treating gambling disorder. To-date, the effects of naltrexone on pre-clinical models of gambling have not been evaluated. The purpose of the present study was to evaluate the effects of naltrexone …
