In vivo transcutaneous penetration of nicotinates and sensitive skin.

Proclivity to develop irritant reactions and transcutaneous penetration of nicotinates has been investigated in 20 subjects of both sexes, divided into reactors and nonreactors on the basis of the responses to irritant stimuli. 1% sodium lauryl sulphate (patch application for 24 h) and 5% lactic acid in aqueous solutions were used to detect chemical and sensory (subjective) irritation. The vasodilatation induced was measured using a chromameter for 1 h after topical application of the chemical. The area-under-the-curve response and the peak response was used to assess the in vivo penetration o

Effects of cigarette smoke and nicotine on feeding and energy.

Much evidence has accumulated indicating that cigarette smokers weigh less than non-smokers and that smokers gain weight when they cease smoking. In the present study we evaluated the effects of cigarette smoke and nicotine on food intake, weight gain, resting energy output, brown fat mass and opiate binding (opiates initiate feeding in sated rats) in rats. Chronic smoke exposure slightly suppressed growth rate and food intake after 14 days of smoke exposure. Blood glucose levels and intrascapular brown adipose mase were increased as a result of smoke exposure. Hamsters chronically exposed to …

Nicotinic-like effects and tissue disposition of isopropylamine.

Previous studies in our laboratory have demonstrated that the acute sympathomimetic effects of isopropylamine are due to stimulation of autonomic ganglia. The present studies were designed to determine whether isopropylamine produced ganglionic blockade after its initial stimulation of autonomic ganglia. Infusion of isopropylamine (2.5 mg/kg/min) produced an initial increase in arterial pressure and heart rate which was followed by a prolonged hypotension and bradycardia; lower doses produced only a hypotensive response. After infusion of isopropylamine, the positive chronotropic responses to

Alteration of the physicochemical properties of triphosphoinositide by nicotinic ligands.

The concentrations of nicotinic drugs required to effect a 50% transfer of triphosphol[3H]isositide from an aqueous phase to a nonaqueous phase closely approximated their concentrations for both in vivo neuromuscular blocking activity and binding to purified nicotinic receptors, and correlated well (correlation coefficient = 0.95)) with their biological activities measured by other workers in an Electrophorus electroplax preparation. The triphospho[3H]inositide transfer induced by nicotinic ligands was dependent on the lipid concentration and was potentiated by Ca2+. The affinity constants of

Psychotic pica, nicotinism, and complicated myocardial infarction.

The authors report a case of a schizophrenic patient who repeatedly consumed a wide variety of inedible materials, including significant qua … medico-anthropologic point of view. The toxic organic brain syndrome caused by chronic ingestion of nicotine-containing products has also been neglected in psychiatric publications. Descriptions of the neuropsychiatric complications of subacute and chronic nicotinism have been restricted to textbooks of toxicology, where greater emphasis has been given to …

Dissociable roles of dopamine D1 and nicotinic receptors in nicotine-motivated responding and impulsive action in a Go/No-Go self-administration task.

Cigarette smoking remains one of the most important preventable causes of premature death worldwide. However, the mechanisms sustaining nicotine dependence and relapse are incompletely understood, particularly the role of impulsive action in persistent tobacco use. Dopamine D1 and nicotinic acetylcholine receptors both regulate nicotine-related behavior, but whether they contribute differently to nicotine-motivated responding and nicotine-induced impulsive action is unclear. The present study examined the effects of D1 receptor blockade and stimulation in male and female rats trained to self-a…

SR9883 is a novel small-molecule enhancer of α4β2* nicotinic acetylcholine receptor signaling that decreases intravenous nicotine self-administration in rats.

BACKGROUND: Most smokers attempting to quit will quickly relapse to tobacco use even when treated with the most efficacious smoking cessatio … nterface of α4/α4 and α4/α5 subunits. RESULTS: The NS9283 derivative SR9883 enhanced the effect of nicotine on α4β2* nAChR stoichiometries containing low-affinity agonist binding sites, with EC50 values from 0.2-0.4 μM. SR9883 had no effect on α3β2* or α3β4* nAChRs. SR9883 was bioavailable after intravenous (1 mg kg-1) and oral (10-20 mg kg-1) administrat …

Nicotine Induced Neurocognitive Protection and Anti-inflammation Effect by Activating α 4β 2 Nicotinic Acetylcholine Receptors in Ischemic Rats.

INTRODUCTION: The main objective of this study was to explore the mechanism of nicotine improving cognitive impairments in ischemic rats. METHODS: Twenty adult male Sprague-Dawley (SD) rats underwent ischemic model surgery by injecting endothelin-1 into the left thalamus, which were classified into four different groups with different intervention: nicotine (1.5 mg/kg/d), dihydro-β-erythroidine (DHβE; 3 mg/kg/d), nicotine (1.5 mg/kg/d) + DHβE (3 mg/kg/d), or saline, after ischemic model surgery. Another five male SD rats also underwent same surgery, while not injecting endothelin-1 but saline…

Nicotine deteriorates the osteogenic differentiation of periodontal ligament stem cells through α7 nicotinic acetylcholine receptor regulating Wnt pathway.

AIMS: Cigarette smoking is one of the high risk factors of adult chronic periodontitis and nicotine is the well established toxic substance in cigarette. However, the mechanism of nicotine induced periodontitis is still unknown. Here we studied whether nicotine impaired the osteogenic differentiation of human periodontal ligament stem cells (hPDLSCs) through activating α7 nicotinic acetylcholine receptor (α7 nAChR). METHODS: hPDLSCs with multi differentiation potential and surface makers for mesenchymal stem cells were harvested by limiting dilution technique. The level of mineralized nodule …

The SARS-CoV-2 Virus and the Cholinergic System: Spike Protein Interaction with Human Nicotinic Acetylcholine Receptors and the Nicotinic Agonist Varenicline.

Severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) is responsible for the worldwide coronavirus disease 2019 (COVID-19) pandemic. Although the pathophysiology of SARS-CoV-2 infection is still being elucidated, the nicotinic cholinergic system may play a role. To evaluate the interaction of the SARS-CoV-2 virus with human nicotinic acetylcholine receptors (nAChRs), we assessed the in vitro interaction of the spike protein of the SARS-CoV-2 virus with various subunits of nAChRs. Electrophysiology recordings were conducted at α4β2, α3β4, α3α5β4, α4α6β2, and α7 neuronal nAChRs expressed
