Diverse endophytic nitrogen-fixing bacteria isolated from wild rice Oryza rufipogon and description of Phytobacter diazotrophicus gen. nov. sp. nov.

Twenty-three nitrogen-fixing bacteria were isolated from surface-sterilized stems and roots of wild rice Oryza rufipogon. Four clusters were defined among these bacteria by SDS-PAGE protein patterns and further confirmed by IS-PCR finger-printing analysis. Phylogenetic analysis of 16S rRNA gene sequences showed that the representative strains LS 8 and LS 18 of cluster II formed a monophyletic group sharing 94.0-97.3% similarities with defined enterobacterial species within the genera Salmonella, Citrobacter, Pantoea, Klebsiella, and Enterobacter. DNA-DNA hybridization, physiological, biochemic

Multi-site CO2 fixation in triazolates: cooperative O,N binding enhanced by solvation and counter-ion effects.

The cooperative interaction of nitrogen and oxygen centers in aromatic heterocycles provides an effective pathway for charge-assisted CO2 capture. Building on the pioneering work of Luo et al. on hydroxy-pyridine systems and our recent PCCP study on hydroxy-substituted N-heterocycles, this work extends the O,N-cooperative binding concept to triazolate frameworks. Density functional theory (DFT) and molecular electrostatic potential (MESP) analyses were performed on neutral, monoanionic, and dianionic 1,2,3- and 1,2,4-triazoles to elucidate how charge, counter-ion and solvation govern CO2 adsor

Soybean α-Amylase Gene Family: Structure and Expression in Response to Abiotic Stresses.

α-Amylases are involved in starch breakdown, thereby influencing plant development. Information on the α-amylase genes in soybean is limited. Here, we identified five soybean α-amylase genes from subfamilies AtAMY1 (GmaAMY5), AtAMY2 (GmaAMY4), AtAMY3 (GmaAMY1, GmaAMY3), and AMY6 (GmaAMY2). In silico analysis indicated that all five genes were actively expressed in leaves, flowers, and pods but weakly in roots. GmaAMY1-GmaAMY5 mRNAs were predicted to be targets of miRNAs associated with stress response, organ development, and nitrogen fixation. Putative GmaAMY1-GmaAMY5 proteins contained α-amyl…

Featuring the Role of Coordination Geometry in Effective CO2 Fixation in Lower-Dimensional Hybrid Copper Phosphonate Frameworks.

The rising atmospheric CO2 concentrations and their role in climate change necessitate the formulation of effective techniques for CO2 capture and utilization. The catalytic conversion of CO2 into valuable compounds offers a possible method for reducing its environmentally harmful effects. The present study explores the investigation of two amine-templated one-dimensional copper phosphonate frameworks: [C3N6H6][Cu2(H2hedp)]·H2O (I) and [C4N2H12][Cu3(hedp)2]·4H2O (II) via a hydrothermal synthetic strategy using H4hedp (1-hydroxyethane-1,1-diphosphonic acid) as the primary ligand and two differe

Persulfidation of host NADPH oxidase RbohB by rhizobial 3-mercaptopyruvate sulfurtransferase maintains redox homeostasis and promotes symbiotic nodulation in soybean.

Reactive oxygen species play a crucial role in various stages of the legume-rhizobia symbiosis, from initial nodulation signaling to nodule senescence. However, how rhizobial redox-related proteins regulate symbiotic nodulation in legumes remains largely unknown. By combining transcriptomics, proteomics, and biochemical and molecular genetics, we investigated the role of the Sinorhizobium fredii Q8 enzyme 3-mercaptopyruvate sulfurtransferase (3MST). Although 3MST was not the primary enzyme responsible for hydrogen sulfide (H2S) production under our conditions, its absence significantly impaire

Microspatial gene expression patterns in the Amazon River Plume.

We investigated expression of genes mediating elemental cycling at the microspatial scale in the ocean's largest river plume using, to our knowledge, the first fully quantitative inventory of genes and transcripts. The bacterial and archaeal communities associated with a phytoplankton bloom in Amazon River Plume waters at the outer continental shelf in June 2010 harbored ∼ 1.0 × 10(13) genes and 4.7 × 10(11) transcripts per liter that mapped to several thousand microbial genomes. Genomes from free-living cells were more abundant than those from particle-associated cells, and they generated mor

Diversity and Structural Composition of Bacterial Communities in Mineral Springs of the Andean-Amazonian Transition Zone in Northern Peru.

Mineral springs in the Andean-Amazonian transition represent poorly explored aquatic ecosystems whose hydrogeochemical heterogeneity may favor the establishment of specialized microbial communities. In this study, bacterial communities in sediments from 19 mineral springs in northern Peru were characterized using 16S rRNA gene sequencing, and their relationships with hydrogeochemical conditions and predicted functional potential were evaluated. The springs exhibited temperatures ranging from 15.1 to 38.2 °C and total dissolved solids (TDS) concentrations ranging from 0.022 to 162.5 mg L-1, ref

Differential effects of sulfide-induced transformation of biodegradable and conventional microplastics on sedimentary CO2 and CH4 emissions: Underlying microbiome-mediated mechanisms.

The accumulation of microplastics (MPs) in sediments presents serious ecological risks. Although sulfidation is a key aging process in anoxi … nriched microbiome (e.g., Acidobacteriota, ester-hydrolyzing Myxococcota) adapted to acidic stress, nitrogen fixation, and pathogenicity, likely diverted carbon flows. In contrast, sulfidation-aged PE (PE-S) exhibited surface oxidation, which led to a 36.7% increase in CH4 emissions, along with higher dissolved organic carbon (DOC) and microbial metabolic quotient …

Synthesis and application of a cobalt(II)-incorporated hydrazone-linked covalent organic framework for the synthesis of cyclic carbonates and oxazolidinones via CO2 fixation under solvent-free conditions.

Herein, a hydrazone-linked covalent organic framework (TFPB-OD) was constructed using 1,3,5-tris(4-formylphenyl)benzene (TFPB) and oxalyldihydrazide (OD) monomeric units. Co(II) was embedded via post-synthetic modification, yielding a well-coordinated, nitrogen- and oxygen-rich porous framework, Co(II)-TFPB-OD, for the synthesis of an active Lewis acidic center-containing heterogeneous catalyst. Both metal-containing and metal-free hydrazone-linked COFs were thoroughly characterised using different characterization techniques, such as FT-IR spectroscopy, PXRD, BET surface area analysis, XPS, T

Engineering Chlorella-based consortia for sustainable food wastewater treatment: nutrient recovery, pollutant degradation and advanced harvesting.

Microalgae-based bioremediation of wastewater enables simultaneous nutrient removal and production of value-added biomass. This study systematically evaluated three geographically distinct Chlorella sp. strains for semi-continuous treatment of food wastewater, addressing critical challenges that hinder practical application. All strains demonstrated substantial nutrient removal (NO3--N > 73%, TP > 78%), with film-forming Chlorella sp. CQ uniquely sustaining complete nitrate elimination. This strain simultaneously produced biomass in which essential amino acids accounted for 40% of total amino

Sequential UV mutagenesis and CO2 acclimation reprogram metabolic carbon allocation in Euglena gracilis.

Euglena gracilis is promising for CO2 bioutilization, but its metabolic response to elevated CO2 is poorly characterized. Here, the LE-ZW mutant, obtained via UV mutagenesis and CO2 acclimation, was evaluated under 5% and 10% CO2. LE-ZW exhibited 7% higher cell density and a 61% increase in photosynthetic carbon fixation at 10% CO2 compared with 5% CO2. Carbon allocation shifted with CO2 concentration: lipid accumulation peaked at 20.57% under 5% CO2, while paramylon content reached 37.37% under 10% CO2. Protein levels increased by 4% under high CO2 (vs 5% CO2 in LE-ZW). Transcriptomic and met

Metatranscriptional Response of Chemoautotrophic Ifremeria nautilei Endosymbionts to Differing Sulfur Regimes.

Endosymbioses between animals and chemoautotrophic bacteria are ubiquitous at hydrothermal vents. These environments are distinguished by high physico-chemical variability, yet we know little about how these symbioses respond to environmental fluctuations. We therefore examined how the γ-proteobacterial symbionts of the vent snail Ifremeria nautilei respond to changes in sulfur geochemistry. Via shipboard high-pressure incubations, we subjected snails to 105 μM hydrogen sulfide (LS), 350 μM hydrogen sulfide (HS), 300 μM thiosulfate (TS) and seawater without any added inorganic electron donor (

The microbe-mediated mechanisms affecting topsoil carbon stock in Tibetan grasslands.

Warming has been shown to cause soil carbon (C) loss in northern grasslands owing to accelerated microbial decomposition that offsets increased grass productivity. Yet, a multi-decadal survey indicated that the surface soil C stock in Tibetan alpine grasslands remained relatively stable. To investigate this inconsistency, we analyzed the feedback responses of soil microbial communities to simulated warming by soil transplant in Tibetan grasslands. Whereas microbial functional diversity decreased in response to warming, microbial community structure did not correlate with changes in temperature
