Quercetin suppress microglial neuroinflammatory response and induce antidepressent-like effect in olfactory bulbectomized rats.

In rodents, olfactory bulbectomy (OBX) results in several behavioral and biochemical changes, useful as a screening model for antidepressants. Recent evidences suggest that quercetin; a bioflavonoid exhibits a variety of behavioral effects including anxiolytic, antidepressant, etc. Since microglia are commonly implicated in the neuroinflammation cascade of depression, we hypothesized that quercetin might involve microglial inhibition pathway in its antidepressant-like effects. To support such a possibility, we investigated the interaction of quercetin with a known microglial inhibitor (minocyc

Regional sympathetic blockade attenuates activation of intestinal macrophages and reduces gut barrier failure.

BACKGROUND: Endotoxin-induced activation of monocytes may lead to extravasation of cells, excessive production of nitric oxide, and subsequent epithelial injury in the gut. Regional sympathetic blockade by means of thoracic epidural anesthesia has been implicated to protect the epithelial barrier. This study tested the hypothesis that thoracic epidural anesthesia decreases epithelial permeability by attenuating monocytic production of nitric oxide and nitrosative stress. METHODS: Rats were anesthetized, hemodynamically monitored, and mechanically ventilated. Endotoxemia was induced by an intr

Apoptosis-inducing high (.)NO concentrations are not sustained either in nascent or in developed cancers.

Nitric oxide ((.)NO) induces apoptosis at high concentrations by S-nitrosating proteins such as glyceraldehyde-3-phosphate dehydrogenase. Th … NO concentration is further depleted; (.)NO is rapidly consumed by red blood cells (RBCs) through S-nitrosation of RBC glutathione and hemoglobin, and by oxidation to nitrate by RBC oxyhemoglobin. Angiogenesis-inhibiting antibodies are currently used to treat cancers; their mode of action is not, as previously thought, reduction of the tumor O2 or nutrient su …

Pyruvate protects glucose-deprived Müller cells from nitric oxide-induced oxidative stress by radical scavenging.

The cellular defense of Müller cells against oxidative and nitrosative stress was examined after the addition of the nitric oxide donor papanonoate. Glucose concentrations of > or = 550 microM efficiently protected the Müller cells from cell death by maintaining high ATP and glutathione and allowing only a moderate increase of free radicals. Fluorescence-activated cell sorting (FACS) analysis showed that 22% of the cells underwent apoptosis whereas necrosis was strongly suppressed. Under glucose deprivation, the intracellular concentration of ATP declined to 15% after 1 h; glutathione dropped

Inflammatory properties of IgG modified by oxygen radicals and peroxynitrite.

In inflammatory arthritis, there is evidence indicating that the affected tissues produce large amounts of oxygen-free radicals and NO. Herein, we examine the biologic effects of exposure of IgG to hypochlorous acid (HOCl) and peroxynitrite (ONOO). The concentrations of IgG modified by chlorination and nitrosation were measured in synovial fluids from inflammatory and noninflammatory arthritis. Human IgG was exposed to increasing concentrations of HOCl and ONOO, and the resulting products were tested for complement component binding; binding to FcgammaRI; activation of polymorphonuclear neutro…

Effects of curcumin on mitochondria in neurodegenerative diseases.

Neurodegenerative diseases (NDs) result from progressive deterioration of selectively susceptible neuron populations in different central nervous system (CNS) regions. NDs are classified in accordance with the primary clinical manifestations (e.g., parkinsonism, dementia, or motor neuron disease), the anatomic basis of neurodegeneration (e.g., frontotemporal degenerations, extrapyramidal disorders, or spinocerebellar degenerations), and fundamental molecular abnormalities (e.g., mutations, mitochondrial dysfunction, and its related molecular alterations). NDs include the Alzheimer disease and

Regulation of S-Nitrosylation in Aging and Senescence.

Nitric oxide signals through several distinct mechanisms, including interaction with the heme group of guanylyl cyclase enzymes resulting in modulation of cGMP levels in the vascular endothelium. Alternatively, reactive nitrogen oxide species can bind cysteine residues in target proteins forming S-nitrosothiols. S-nitrosylation is recognized as an important post-translational modification of dozens of proteins, which plays a key role in cellular homeostasis, metabolism, and various disease states. By denitrosylating target proteins, S-nitrosoglutathione reductase (GSNOR/ADH5) plays a pivotal r

Oxidative/nitrative stress in the pathogenesis of systemic sclerosis: are antioxidants beneficial?

Systemic sclerosis (SSc) is a multisystem autoimmune disease: characterised from the clinical side by progressive vasculopathy and fibrosis of the skin and different organs and from the biochemical side by fibroblast deregulation with excessive production of collagen and increased expression of nicotinamide adenine dinucleotide phosphate oxidase 4 (NOX4). The latter contributes to an overproduction of reactive oxygen species that through an autocrine loop maintains NOX4 in a state of activation. Reactive oxygen and nitrogen species are implicated in the origin and perpetuation of several clini

Direct covalent modification as a strategy to inhibit nuclear factor-kappa B.

Nuclear Factor-KkappaB (NF-kappaB) is a transcription factor whose inappropriate activation may result in the development of a number of diseases including cancer, inflammation, neurodegeneration and AIDS. Recent studies on NF-kappaB mediated pathologies, made therapeutic interventions leading to its inhibition an emerging theme in pharmaceutical research. NF-kappaB resides in the cytoplasm and is activated by several time-dependent factors, leading to proteasome-dependent degradation of its inhibitory protein (IkappaB), resulting in free NF-kappaB (p50 and p65 subunits, involved in disease st

A SNO storm in skeletal muscle.

Dysregulated S-nitrosylation of proteins characterizes a broad array of human disorders, but its role in disease etiology is not well understood. Two new studies (Durham et al., 2008; Bellinger et al., 2008) now show that hyper-S-nitrosylation of the ryanodine receptor calcium release channel (RyR1) in skeletal muscle disrupts calcium ion flux. This disruption underlies the impaired contractility and cellular damage of skeletal muscle during strenuous exercise and in a spectrum of congenital muscle disorders including malignant hyperthermia.

Metallotoxicity-driven oxidative perturbations in spinal cord injury: Unraveling the pathophysiological nexus between metal-induced redox imbalance and disease severity: A correlational and multivariate analysis.

BACKGROUND: Secondary injury processes, especially oxidative stress and inflammation, are crucial in aggravating neuronal damage and restricting recovery in spinal cord injury (SCI), a crippling neurological disorder. Disruption of metal homeostasis may further amplify redox imbalance by impairing antioxidant defenses and promoting inflammatory pathways; however, integrated clinical evidence linking metal dyshomeostasis with oxidative, nitrosative, and inflammatory biomarkers in human SCI remains limited. OBJECTIVE: To investigate the interrelationships among trace and toxic metal levels, oxi

TSPO safeguards porphyrin-iron balance under anoxic conditions in Bacillus cereus.

The physiological role of the bacterial translocator protein (TSPO) under anoxic conditions remains poorly defined. Here, we show that deletion of tspO in Bacillus cereus growing under fermentative anoxic conditions leads to increased porphyrin accumulation, redox imbalance, and impaired growth. Cellular proteome analyses reveal extensive remodeling of central metabolism, including increased abundance of proteins in the siroheme branch of tetrapyrrole biosynthesis, increased abundance of nitrate assimilation and respiratory nitrate reduction components, and broad alterations in iron, metal ion

S-nitrosylation contributes to ER stress and aggresome formation in secosterol-B-mediated endothelial dysfunction.

The involvement and the in vivo relevance of endoplasmic reticulum (ER) stress in the atherosclerotic process are well established, but the mechanisms have been only partly elucidated. Emerging evidence indicates that ER protein folding pathways are sensitive to nitric oxide (NO) fluctuations and therefore heavily vulnerable under conditions of nitrosative stress. Recent research indicates that protein S-nitrosylation (S-NO), a key redox-mediated modification involved in several disorders, affects neuronal function by altering ER stress sensor proteins. However, the mechanisms by which ER prot

Bergenin attenuates neuropathic pain by modulation of TRPV1/CGRP signalling and neuro-inflammatory mechanisms.

BACKGROUND AND PURPOSE: Neuropathic pain is a chronic and disabling condition characterized by persistent sensory hypersensitivity and inadequate treatment options. Neuroinflammation and TRPV1/CGRP-driven nociceptive pathways are critical contributors to its pathogenesis. Bergenin, a naturally occurring C-glucoside with established anti-inflammatory and antioxidant activities, may offer a promising therapeutic approach. EXPERIMENTAL APPROACH: Neuropathic pain was induced in rats using the chronic constriction injury (CCI) model. Bergenin (25, 50, and 100 mg/kg, i.p.) and gabapentin (30 mg/kg,

Liposomal Piceatannol Mitigates Methotrexate-Induced Oxidative Renal Injury via Modulation of Nrf2/HO-1, TLR4/NF-κB, MAPK, and Apoptotic Pathways in Rats.

Methotrexate (MTX) is a widely used chemotherapeutic and immunosuppressive agent, but its clinical utility is limited by oxidative stress-mediated renal toxicity. This study evaluated the nephroprotective potential of the dietary polyphenolic bioactive molecule piceatannol (PIC) in its crude and liposomal nanoparticle (PIC-LNP) forms against MTX-induced kidney injury in rats. Sixty rats were allocated into six groups and received vehicle, PIC, PIC-LNPs, MTX, or combinations of MTX with PIC or PIC-LNPs. MTX administration induced marked renal dysfunction and oxidative/nitrosative stress, reflec

Molecular Magnetic Resonance Imaging Visualizes Nitric Oxide Dynamics in a Mouse Model of Acute Myocardial Inflammation.

Nitric oxide (NO) is an essential signaling molecule that appears early in inflammation and persists in chronic diseases, making it a promising biomarker for early detection and treatment monitoring. Although transient NO production supports immune defense, sustained elevations lead to nitrosative stress, tissue injury, and accelerated disease progression. However, NO's short half-life and deep-tissue localization make noninvasive detection challenging. Current diagnostic methods lack combined sensitivity, specificity, and spatial resolution needed to effectively image NO in vivo. To address t

Post-myocardial infarction heart failure and long-term high-fat diet: Cardiac endoplasmic reticulum stress and unfolded protein response in Sprague Dawley rat model.

BACKGROUND: Myocardial infarction (MI) significantly contributes to the global mortality rate, often leading to heart failure (HF) due to left ventricular remodeling. Key factors in the pathomechanism of HF include nitrosative/oxidative stress, inflammation, and endoplasmic reticulum (ER) stress. Furthermore, while a high-fat diet (HFD) is known to exacerbate post-MI cardiac remodeling, its impact on these critical factors in the context of HF is not as well understood. AIMS: This study aimed to assess the impact of post-MI HF and HFD on inflammation, nitro-oxidative stress, ER stress, and un
