Relative contribution of brain and peripheral connections to postnatal growth and cell accretion in the rat olfactory bulb.

Total weight and DNA (cell number) determination in whole olfactory bulbs (OB) revealed that unilateral transection of the olfactory peduncle in newborn rats causes 2 X more (40%) deficit in cell number in the operated OB than section of the olfactory nerve. The deficit due to combined operations ('isolation' of OB) were additive. Similar effects were observed on weight gain. It is inferred that both olfactory nerve and sources in the brain exert marked influences on postnatal cell accretion and growth in the OB and that the greater share of the brain is due to its direct contribution of new m…

An electrophysiological study of the accessory olfactory bulb in the rabbit--I. Analysis of electrically evoked potential fields.

Following electrical stimulation of the vomeronasal nerves, the primary olfactory nerves, the lateral olfactory tract and the corticomedial amygdala, we have made a study of evoked potentials in the rabbit accessory olfactory bulb. Vomeronasal nerve stimulation evoked a complex field potential consisting of a compound action potential followed by 4 negative waves (N1, N2, N3, N4). In contrast to the field potential elicited in the main olfactory bulb following primary olfactory nerve stimulation, there was either no evoked wave or only a weak positive component of the field in the accessory bu…

Localization of synaptic responses in the in vitro turtle olfactory bulb using the [14C]2-deoxyglucose method.

Studies were carried out on the in vitro turtle olfactory bulb preparation with a modification of the [14C]-2-deoxyglucose (2-DG) technique. Electrical stimulation of either the dorsal or ventral division of the olfactory nerve produced broad bands of activity, spreading across laminae, in the corresponding half of the olfactory bulb. Pretreatment of the isolated preparation in a low calcium bath eliminated focal 2-DG uptake induced by electrical stimulation of the nerve. In contrast, pretreatment with the GABAergic antagonist, bicuculline, produced a notable increase in 2-DG accumulation. The…

Field potentials in the olfactory bulb of the rainbow trout (Salmo gairdneri): evidence for a dendrodendritic inhibitory pathway.

Field potentials have been recorded from the olfactory bulb of the teleost fish, Salmo gairdneri. Stimulation of the olfactory nerve generates a complex wave pattern, which when recorded near the surface in the centre of the bulb consists of one positive and three negative components, P, N1, N2 and N3. The N1 has been attributed to depolarization of mitral cell dendrites, N2 to depolarization of the superficial dendrites of granules cells, and P to subsequent hyperpolarization of mitral cells. Possible origins of the N3 wave are discussed. Spikes from single units recorded in the mitral cell l…

Gestational methyl donor deficiency alters key proteins involved in neurosteroidogenesis in the olfactory bulbs of newborn female rats and is associated with impaired olfactory performance.

Gestational methyl donor deficiency (MDD) leads to growth retardation as well as to cognitive and motor disorders in 21-d-old rat pups. These disorders are related to impaired neurogenesis in the cerebral neurogenic areas. Olfactory bulbs (OB), the main target of neuronal progenitors originating from the subventricular zone, play a critical role during the postnatal period by allowing the pups to identify maternal odour. We hypothesised that growth retardation could result from impaired suckling due to impaired olfactory discrimination through imbalanced apoptosis/neurogenesis in the OB. Since…

Activation of the cyclic AMP response element-binding protein signaling pathway in the olfactory bulb is required for the acquisition of olfactory aversive learning in young rats.

Long-term memory formation requires both gene expression and protein synthesis. Phosphorylation of the transcription factor cyclic AMP response element-binding protein (CREB) is thought to be important in processes underlying long-term memory. To clarify the role of CREB in olfactory aversive learning in young rats, we carried out behavioral pharmacology and Western blot analyses. On postnatal day 11, oligodeoxynucleotides were infused directly into the bilateral olfactory bulbs through cannulae implanted prior to training in a classical conditioning paradigm with citral odor and foot shock. O…

Electrical stimulation of primary olfactory nerve induces two types of variations in the extracellular potassium activity within the glomerulus of the rat olfactory bulb in vivo.

Primary olfactory nerve stimulation induces a two-step increase in extracellular potassium, as recorded by K+-sensitive microelectrodes inserted into single glomeruli of the rat olfactory bulb. The second rise in K+ (K-peak) occurs in a graded, activity-dependent manner at the basal pole of the glomerular neuropile. This phenomenon appears to be directly related to the rate of extracellular K+ accumulation and is accompanied by a substantial depression of the mitral cell firing rate.

Possible Volumetric Changes in the Hippocampus and Olfactory Bulbs After COVID-19: A Stereological MRI Study.

COVID-19 affects the olfactory system and the hippocampus, leading to severe olfactory and cognitive impairments. This study aimed to quantify volumetric changes in the olfactory bulbs and hippocampus in patients with COVID-19, and to investigate these differences in relation to loss of smell and taste. This stereological MRI study included 60 COVID-19 (+) participants and 54 non-COVID-19 participants. Using OsiriX software, the volumes of the olfactory bulb and hippocampus were manually measured from 3T MRI images. Compared with controls, participants with loss of smell and taste had smaller …

In Vivo Study of Dynamics and Stability of Dendritic Spines on Olfactory Bulb Interneurons in Xenopus laevis Tadpoles.

Dendritic spines undergo continuous remodeling during development of the nervous system. Their stability is essential for maintaining a functional neuronal circuit. Spine dynamics and stability of cortical excitatory pyramidal neurons have been explored extensively in mammalian animal models. However, little is known about spiny interneurons in non-mammalian vertebrate models. In the present study, neuronal morphology was visualized by single-cell electroporation. Spiny neurons were surveyed in the Xenopus tadpole brain and observed to be widely distributed in the olfactory bulb and telencepha…

A morphological study of the vomeronasal organ and the accessory olfactory bulb in the Korean roe deer, Capreolus pygargus.

The vomeronasal organ (VNO) and accessory olfactory bulb (AOB) of the Korean roe deer (Capreolus pygargus) were studied histologically to evaluate their morphological characteristics. Grossly, the VNO, encased by cartilage, has a paired tubular structure with a caudal blind end and a rostral connection through incisive ducts on the hard palate. In the VNO, the vomeronasal sensory epithelium (VSE) consists of galectin-3-positive supporting cells, protein gene product (PGP) 9.5-positive receptor cells, and basal cells. The vomeronasal respiratory epithelium (VRE) consists of a pseudostratified e…

The periglomerular cell of the olfactory bulb and its role in controlling mitral cell spiking: a computational model.

Interneurons in the olfactory bulb are key elements of odor processing but their roles have not yet being fully understood. Two types of inhibitory interneurons, periglomerular and granule cells, act at two different levels within the olfactory bulb and may have different roles in coordinating the spiking of mitral cells, which are the principal output neurons of the olfactory bulb. In this work we introduce a reduced compartmental model of the periglomerular cell and use it to investigate its role on mitral cell spiking in a model of an elementary cell triad composed of these two cell types p…

Distribution of the A3 subunit of the cyclic nucleotide-gated ion channels in the main olfactory bulb of the rat.

Previous data suggest that cyclic GMP (cGMP) signaling can play key roles in the circuitry of the olfactory bulb (OB). Therefore, the expression of cGMP-selective subunits of the cyclic nucleotide-gated ion channels (CNGs) can be expected in this brain region. In the present study, we demonstrate a widespread expression of the cGMP-selective A3 subunit of the cyclic nucleotide-gated ion channels (CNGA3) in the rat OB. CNGA3 appears in principal cells, including mitral cells and internal, medium and external tufted cells. Moreover, it appears in two populations of interneurons, including a subs…

Differentiation of mitral cell dendrites in the developing main olfactory bulbs of normal and naris-occluded rats.

The morphological differentiation of mitral cell dendrites during embryonic and early postnatal development was examined in the main olfactory bulb of rats to determine a possible role of afferent activity in the development of the dendrites. Mitral cells and olfactory nerve fibers were labeled with 1,1'-dioctadecyl-3,3,3', 3'-tetramethylindocarbocyanine perchlorate (DiI) and fluorescein-conjugated lectin (Ulex europeus agglutinin-I), respectively. Morphogenesis of mitral cell dendrites proceeded as previously described (Malun and Brunjes [1996] J. Comp. Neurol. 368:1-16); that is, undifferent…

Serotonergic neurons are present and innervate blood vessels in the olfactory bulb of the laboratory shrew, Suncus murinus.

The distribution and characteristics of serotonin-immunoreactivity in the olfactory bulb of the laboratory shrew (Suncus murinus, insectivore) was studied immunohistochemically. Serotonergic neurons were found only in the subependymal layer of the main olfactory bulb. These neurons were 8-12 microm in size and bipolar in shape. These serotonergic neurons had smooth nerve fibers which innervate blood vessels located mainly in the subependymal layer of the main olfactory bulb. On the other hand, other serotonergic nerve fibers with varicosities, which must be extrinsic, were detected in most olf…

Dynamic mapping at the laminar level of odor-elicited responses in rat olfactory bulb by functional MRI.

We have applied functional MRI (fMRI) based on blood oxygenation level-dependent (BOLD) image-contrast to map odor-elicited olfactory responses at the laminar level in the rat olfactory bulb (OB) elicited by iso-amyl acetate (10(-2) dilution of saturated vapor) with spatial and temporal resolutions of 220x220x1,000 micro(m) and 36 s. The laminar structure of the OB was clearly depicted by high-resolution in vivo anatomical MRI with spatial resolution of 110x110x1,000 micro(m). In repeated BOLD fMRI measurements, highly significant (P < 0.001) foci were located in the outer layers of both OBs. …

Distribution of astroglia in glomeruli of the rat main olfactory bulb: exclusion from the sensory subcompartment of neuropil.

During an entire lifetime, sensory axons of regenerating olfactory receptor neurons can enter glomeruli in the olfactory bulb and establish synaptic junctions with central neurons. The role played by astrocytes in this unique permissiveness is still unclear. Glomerular astrocytes have been identified by immunocytochemistry for glial fibrillary acidic protein and S100 proteins at the light and electron microscopic levels. The latter labeling included submicroscopic lamellar and filopodial extensions of astroglial processes. Cell bodies and processes accumulate along the border between juxtaglom…
