A comprehensive model has yet to emerge, but it seems likely that numerous mechanisms contribute to the specificity of olfactory sensory neuron (OSN) axon innervation of the olfactory bulb. Elsewhere in the nervous system the Wnt/Fz family has been implicated in patterning of anterior-posterior axes, cell type specification, cell proliferation, and axon guidance. Because of our work describing cadherin-catenin family member expression in the primary olfactory pathway, and because mechanisms of Wnt-Fz interactions can depend in part on catenins, we were encouraged to explore Wnt-Fz expression a

1. Intracellular recordings were made from the output neurons (mitral and tufted cells) of the rat olfactory bulb during electrical orthodromic stimulation of the olfactory nerve layer (ONL) and antidromic stimulation of the lateral olfactory tract and posterior piriform cortex (pPC) to test for physiological differences among the neuron types. Many of these neurons were identified by intracellular injections of biocytin, and others were identified by their pattern of antidromic activation. 2. Both marked and unmarked mitral cells showed large inhibitory postsynaptic potentials (IPSPs) in resp

Because of their anatomical situation, the olfactory nerves constitute a natural obstacle to exploring the anterior cranial fossa, making them vulnerable to traumatic, tumor, or iatrogenic lesions. Consequently, accurate knowledge of their microsurgical anatomy is of particular importance for surgeons to ensure the correct execution of surgical procedures with minimal sequelae, the least functional deterioration, and better therapeutic results. This review describes the functional and microsurgical anatomy of the olfactory nerves, illustrated with pictures of simulations based on cadaveric dis

Odorant stimulation of receptor cells results in a calcium influx that activates the transduction pathway. Ca2+ acceptors, such as calmodulin, may mediate between the change in intracellular calcium and the conductance mechanism underlying the initial electrical event. Ca2+ acceptors also may participate in subsequent processing of olfactory information. The identification and characterization of these molecules, therefore, should provide important information about the complex signal transduction pathway involving calcium in olfaction as well as other sensory systems. The present study descri

We report on responses of olfactory receptor neurons (ORNs) upon application of amino acids and forskolin using a novel slice preparation of the olfactory epithelium of Xenopus laevis tadpoles. Responses were measured using the patch-clamp technique. Both amino acids and forskolin proved to be potent stimuli. Interestingly, a number of ORNs that responded to amino acids did not respond to forskolin. This suggests that some amino acids activate transduction pathways other than the well-known cAMP-mediated one. The differential processing of cAMP-mediated stimuli on the one hand and amino acid s

Living in an aquatic environment, often devoid of light but rich in dissolved compounds, fish have highly developed chemosensory and chemical signalling systems. The olfactory and gustatory systems comprise the major chemosensory pathways. Despite considerable variations in structural organization of the peripheral olfactory organ throughout fish species, ultrastructural organization of the olfactory sensory epithelium is extremely consistent. The olfactory receptor cell is a bipolar neurone which is directly exposed to the external environment and sends information to the brain by its own axo

The ability to make the world of odorants intelligible is a key capability of the Drosophila olfactory system that we shall call olfactory intelligence. Characterizing the functional logic of the Drosophila early olfactory system that makes the natural world of odorants intelligible is a major challenge in neuroscience. Starting by modeling the space of odorants using constructs of both semantic and syntactic information, we establish that the Antenna Lobe and Mushroom Body Calyx first decompose the confounding representation of the Antenna into a concentration independent odorant semantics in

Prion diseases, or transmissible spongiform encephalopathies (TSEs), are a class of fatal neurodegenerative diseases caused by the entry and spread of infectious prion proteins (PrPSc) in the central nervous system (CNS). These diseases are endemic to certain mammalian animal species that use their sense of smell for a variety of purposes and therefore expose their nasal cavity (NC) to PrPSc in the environment. Prion diseases that affect humans are either inherited due to a mutation of the gene that encodes the prion protein, acquired by exposure to contaminated tissues or medical devices, or

The nasal cavity of Mantidactylus betsileanus, a frog of the Madagascar-Comoroan endemic family Mantellidae, is characterized by a unique internal architecture. Unlike the state commonly observed in anurans, the two discernible olfactory subsystems of M. betsileanus (the main olfactory organ and the vomeronasal organ) are anatomically separated from each other, suggesting an enhanced functional differentiation. Here we evaluate the ontogenetic formation of this extraordinary anatomical state based on a histological study of a developmental series of M. betsileanus. The olfactory system of prem

Paradoxically, although humans have a superb sense of smell, they don't trust their nose. Furthermore, although human odorant detection thresholds are very low, only unusually high odorant concentrations spontaneously shift our attention to olfaction. Here we suggest that this lack of olfactory awareness reflects the nature of olfactory attention that is shaped by the spatial and temporal envelopes of olfaction. Regarding the spatial envelope, selective attention is allocated in space. Humans direct an attentional spotlight within spatial coordinates in both vision and audition. Human olfactor

Destruction of the vascularisation of the olfactory structures during fronto-orbital surgical approaches to the sellar region may result in anosmia as a complication. The goal of this study was to describe the sources of blood supply to the proximal olfactory tract and the macroscopic distribution of these vessels. 20 human brains fixed in formalin with arteries injected with ink-coloured gelatine were studied using a surgical microscope and the micro-dissection technique. The vessels running along the olfactory tract posteriorly and anteriorly on its inferior and superior surface were observe

Over the past 3 decades, research exploring potential neuronal replacement therapies have focused on replacing lost neurons by transplanting cells or grafting tissue into diseased regions of the brain [Nat. Neurosci. 3 (2000) 67-78]. Over most of the past century of modern neuroscience, it was thought that the adult brain was completely incapable of generating new neurons. However, in the last decade, the development of new techniques has resulted in an explosion of new research showing that neurogenesis, the birth of new neurons, normally occurs in two limited and specific regions of the adul

In the current study, we addressed two questions: First, is the olfactory placode necessary for the development of the olfactory bulb and the entire telencephalon? Second, does the olfactory placode contribute cells to the olfactory bulb? We addressed these questions by unilaterally ablating the olfactory placode in chick embryos before an olfactory nerve was produced and, in a second series of experiments, by replacing the ablated chick olfactory placode with a quail olfactory placode. Our results indicate that the olfactory placode is critical for olfactory bulb development, but is not neces

A preparation has been developed in the dog which allows recording the electrical activity from an olfactory nerve twig containing the axons of a small group of olfactory receptor cells. The dog's response to n-pentyl acetate is vigorous and stable, like that of other air-breathing animals. The dog's response magnitude dependence on the nasal flow rate was noticeable for n-pentyl acetates, but not so great as for n-butyric acid. The response to n-butyric acid strongly depends on the nasal flow. The start of the nasal air flow caused an increase of neural activity, which is called flow response

1. Single unimodal (olfactory) or multimodal (olfactory and mechanosensory) neurons in the antennal lobe of the deutocerebrum of the American cockroach were characterized functionally by microelectrode recording, and their morphological types and positions in the brain were established by dye injection. Thus individual, physiologically identified neurons of known shape could be mapped in reference to the areas of soma groups, glomeruli, tracts and their projection regions in the brain. 2. All of these neurons send processes to deutocerebral glomeruli, i.e., the regions in which the axons of an

OBJECTIVE: With the trend of population ageing, cognitive frailty in older adults is an increasingly pressing public health issue. Identifying early-stage risk factors has become a key priority for researchers with the goal of preventing disability. This study aimed to test a conceptual model and elucidate the pathways leading to cognitive frailty using Structural Equation Modelling (SEM).
METHODS: We conducted a cross-sectional design with data from outpatient departments within a tertiary medical centre in southern Taiwan. Participants (n = 208) were aged 65 years or older and able to commu

Contrary to all other sensory systems, olfactory information is processed predominantly ipsilaterally. Furthermore, odor localization, based on inter-nostril differences, is usually not possible under controlled conditions. These two observations suggest information exchange between both cerebral hemispheres in the olfactory system, although the exact anatomical substrate remains unknown. This study aimed to identify the anatomical structures necessary for odor localization, with a particular focus on the role of interhemispheric communication. We assessed the ability to localize pure olfactor

OBJECTIVE: Our study aimed to determine whether the short latency afferent inhibition (SAI) response could be associated with the severity of olfactory dysfunction in PD patients.
METHODS: A total of 71 PD patients and 20 controls were enrolled. All PD patients were classified into 3 groups by the severity of the olfactory deficit. Single-pulse transmagnetic stimulation (TMS) parameters and SAI were assessed.
RESULTS: The integrated SAI in the PD with anosmia and PD with hyposomia groups was significantly less inhibited than that in the PD with normosmia and control groups [64.79 {Interquart

BACKGROUND: Olfactory schwannomas of the anterior cranial base (ACB) are rare tumors, and their association with hereditary hemorrhagic telangiectasia (HHT) has not previously been described. The majority of ACB schwannomas arise from the sinonasal tracts and may demonstrate intracranial extension. We report a case of an olfactory schwannoma-dense adherence to the basal frontal lobe. Complete tumor resection was performed through a unilateral extended endonasal endoscopic approach with preservation of the contralateral olfactory bulb. Anterior cranial base repair was achieved with the use of a
