New alternative ingredients and genetic selection are the next game changers in rainbow trout nutrition: a metabolomics appraisal.

The formulation of sustainable fish feeds based on plant ingredients supplemented by alternative ingredients to plant (insect, micro-algae, yeast) and genetic selection of fish for plant-based diets were tested on rainbow trout in two separate experiments. Plant-based diets and corresponding diets supplemented with an ingredient mix: insect, micro-algae and yeast in Experiment A, and insect and yeast in Experiment B were compared to commercial-like diets. In experiment A, the mix-supplemented diet was successful in compensating the altered growth performance of fish fed their respective plant-

Goat semen cryopreservation with rainbow trout seminal plasma supplemented lecithin-based extenders.

The objective of this study was to determine the optimum concentrations of rainbow trout seminal plasma (RTS) supplemented extenders for goat semen quality at post-thaw and after incubation. Five sexually mature Saanen goat (Capra aegagrus hircus) were used for semen collection. Pooled semen was diluted with soybean lecithin-based extender without RTS (control) or supplemented with different concentrations of RTS (1%, 2%, 4% or 8%), at a final concentration of 150 × 106 spermatozoon/ml. Sperm motility, plasma membrane functional integrity (HOST), damaged acrosome (PSA-FITC), mitochondrial acti

Biochemical reference intervals and pathophysiological changes in Flavobacterium psychrophilum-resistant and -susceptible rainbow trout lines.

Host genetic resistance against disease-causing pathogens can be enhanced through family-based selective breeding. At present, there is an incomplete understanding of how artificial selection of fish alters host physiology and response following pathogen exposure. We previously reported the generation of selectively-bred rainbow trout Oncorhynchus mykiss lines with either increased resistance (ARS-Fp-R) or susceptibility (ARS-Fp-S) to bacterial cold water disease (BCWD). This study (1) determined baseline reference-range intervals for packed cell volume (PCV) and 18 plasma biochemistry analyte…

Acute mortality, bacterial load, and pathology of select lines of adult rainbow trout challenged with Weissella sp. NC36.

A challenge for improving disease resistance in fish through genetics is to understand specificity of resistance and whether selection for one pathogen alters the response to unrelated pathogenic microorganisms. Adult Rainbow Trout Oncorhynchus mykiss that had been bred for differential susceptibility to Flavobacterium psychrophilum, the causative agent of bacterial cold water disease (BCWD) and designated ARS-Fp-R (resistant), ARS-Fp-S (susceptible), and ARS-Fp-C (control line), as well as a pool of commercial-stock Rainbow Trout, were intraperitoneally challenged with Weissella sp. NC36. Cli…

Pinealectomy does not affect the entrainment to light nor the generation of the circadian demand-feeding rhythms of rainbow trout.

The pineal organ and its secretory product melatonin are regarded as synchronizers of daily rhythms to the external light/dark (LD) cycle. In fish, the pineal organ acts as a direct photoreceptor, transducing light information into neural and humoral (melatonin) signals. In the present study, we investigate a possible role for the pineal organ and melatonin in the regulation of feeding rhythms of rainbow trout, Oncorhynchus mykiss. We used individual rainbow trout placed in an insulated room at constant temperature (14 degrees C). Fish were self-fed ad lib by means of self-feeders coupled to a…

Detection of triploidy in fish using the cytokinesis-blocked method for erythrocyte and hepatic cells.

The use of two cytochalasin-blocked methods with erythrocyte and hepatic cells was developed to identify spontaneous and/or induced triploid fish by observing the presence of trinucleated cells. Spontaneous triploids were found in two specimens out of fifteen commercial rainbow trout from a hatchery at 2 years of age. Some rare spontaneous triploid (3n) trinucleated cells were also found in the other thirteen diploid (2n) trout specimens under investigation. The use of this novel technique in a study of the triploidy in vitro, allows evaluation of the polyploid specimens under field and labora

Freshwater browning as a hidden threat - Transcriptomic responses in fish gills exposed to fulvic acid.

Human activities and climate change have significantly increased humic substances in freshwater ecosystems over the last few decades. This increase is particularly concerning during seasonal changes or after heavy rainfall, when concentrations can easily increase up to tenfold. This phenomenon, known as "browning," has unknown consequences for aquatic organisms. This study is the first to determine the effects of increasing humic substance concentrations on the transcriptomic and structural responses in the gills of rainbow trout (Oncorhynchus mykiss). Overall, 27 genes mainly involved in xeno…

Tapeworm infection incidence in rural Japan points to a common environmental source of infection.

Dibothriocephalus nihonkaiensis is a zoonotic tapeworm transmitted to humans through consumption of raw or undercooked fish or wild meat. Between 2022 and 2023, Yamagata Prefecture reported an increase in cases compared with 2017-2021, when none were observed. We conducted a clinical and environmental investigation to clarify infection sources. Four confirmed and one suspected patient were identified, all presenting with gastrointestinal symptoms. Exposures were linked to raw cherry salmon (Oncorhynchus masou) in three cases and undercooked bear meat in one case. Praziquantel treatment (10-20 …

Effects of fishing stress on fatty acid and amino acid composition and glycolipid metabolism in triploid rainbow trout.

Triploid Oncorhynchus mykiss is an important economic fish worldwide. Fishing stress can affect its growth and meat quality. This study first explored the effects of fishing stress on fatty acid and amino acid in triploid O. mykiss. Results showed fishing stress significantly reduced the content of docosadienoic acid, Gly, Arg, and DAA (P < 0.05). Targeted lipidomics analysis furthered suggested that some lipid molecules belonging to TG, DG, PC, Cer, ChE, and So were significantly up-regulated; while some lipid molecules belonging to Cer, LPE, LPC, PS, PC, and SM were significantly down-regula…

Detection, sequencing, and tissue distribution of piscine orthoreovirus 2-like virus in diseased coho salmon in Alaska.

We performed a diagnostic disease investigation on a cohort of coho salmon (Oncorhynchus kisutch) fingerlings in Alaska exhibiting anorexia, gaping mouths, anemia, and increased mortality. Histologic examination revealed mild-to-severe myocardial degeneration and lymphohistiocytic and neutrophilic myocarditis, moderate splenic histiocytosis, and mild renal histiocytosis. Piscine orthoreoviruses 1 and 3 were not detected by molecular methods, and no other viruses could be cultured on 3 common diagnostic fish cell lines. De novo assembly produced a viral genome of 10 linear segments with >80% ho…

Detection of traces of tetracyclines from fish with a bioluminescent sensor strain incorporating bacterial luciferase reporter genes.

Bioluminescent Escherichia coli K-12 strain for the specific detection of the tetracycline family of antimicrobial agents was optimized to work with fish samples. The biosensing strain contains a plasmid incorporating the bacterial luciferase operon of Photorhabdus luminescens under the control of the tetracycline responsive element from transposon Tn10 (Korpela et al. Anal. Chem. 1998, 70, 4457-4462). The extraction procedure of oxytetracycline from rainbow trout (Oncorhynchus mykiss) tissue was optimized. There was neither need for centrifugation of homogenized tissue nor use of organic solv…

Signal transmission in the photosensitive pineal organ of the rainbow trout: modulation of ganglion cell activity by intrinsic dopamine.

The photosensitive pineal organ of the rainbow trout (Oncorhynchus mykiss) transduces photic information into nycthemeral neuronal signals. To investigate origin, cellular localization, and functional significance of pineal catecholamines, we performed HPLC-analysis of catecholamines and tyrosine hydroxylase (TH) activity, as well as immunocytochemical and electrophysiological studies. In biochemical and immunocytochemical investigations, pineal cells were found to contain endogenous TH. Using HPLC-analysis, the presence of a catecholamine precursor (L-dopa), catecholamines (dopamine, norepine…

Gyrodactylus salmonis infection impairs the olfactory system of rainbow trout.

Monogenean worms are ectoparasites that are known to be infectious to a wide variety of fish. Few species of monogenean parasites have been reported in the olfactory chamber of fish in current peer-reviewed literature. However, the impacts of these parasites on the olfactory system are not well understood. In this study, the effects of Gyrodactylus salmonis on the olfactory system structure and performance were investigated in rainbow trout (Oncorhynchus mykiss). The olfactory performance of the infected fish was examined using an electro-olfactography (EOG) technique, while the ultrastructure…

Population-level impacts of reducing artificial light at night across an entire migratory corridor.

Artificial light at night (ALAN) is a widespread anthropogenic pollution that poses challenges for wildlife in nearly every ecosystem on planet Earth. Although there has been substantial progress on understanding the effects of ALAN on animal physiology and behaviour, few studies have estimated its population-level impacts in the wild, leaving the potential conservation benefits of ALAN reduction unresolved. Here, we evaluated the population-level effect of ALAN reduction through its impact on juvenile Chinook salmon (Oncorhynchus tshawytscha) vulnerability to predation during oceanward migrat…

Discovery of an unrecognized nidovirus associated with granulomatous hepatitis in rainbow trout.

Rainbow trout (Oncorhynchus mykiss) is the principal species of inland-farmed fish in the Western hemisphere. Recently, we diagnosed in farmed rainbow trout a disease in which the hallmark is granulomatous-like hepatitis. No biotic agents could be isolated from lesions. Still, unbiased high-throughput sequencing and bioinformatics analyses revealed the presence of a novel piscine nidovirus that we named "Trout Granulomatous Virus" (TGV). TGV genome (28,767 nucleotides long) is predicted to encode non-structural (1a and 1 ab) and structural (S, M, and N) proteins that resemble proteins of other…

Impact of Vaccination and Pathogen Exposure Dosage on Shedding Kinetics of Infectious Hematopoietic Necrosis Virus (IHNV) in Rainbow Trout.

Vaccine efficacy in preventing clinical disease has been well characterized. However, vaccine impacts on transmission under diverse field conditions, such as variable pathogen exposure dosages, are not fully understood. We evaluated the impacts of vaccination on disease-induced host mortality and shedding of infectious hematopoietic necrosis virus (IHNV) in Rainbow Trout Oncorhynchus mykiss. Fish, in up to three different genetic lines, were exposed to different dosages of IHNV to simulate field variability. Mortality and viral shedding of each individual fish were quantified over the course o…

Verapamil does not modify catalytic activity of CYP450 in rainbow trout after long-term exposure.

Little is known about the effects of the cardiovascular drug verapamil (VRP) on metabolic processes in fish. Most calcium channel blockers including VRP are metabolized by cytochrome P450 (CYP450) enzymes. In this study we investigated the in vivo effect of VRP on some CYP450-mediated reactions in juvenile rainbow trout (Oncorhynchus mykiss). Fish were exposed to sublethal concentrations of VRP (0.5, 27 and 270 μg l(-1)) for 0, 21, and 42 day. The following CYP450-mediated reactions were studied in hepatic microsomes: O-dealkylation of ethoxyresorufin, methoxyresorufin, and pentoxyresorufin, h…

Mitochondrial DNA of Anisakis simplex s.s. as a potential tool for differentiating populations.

Anisakis simplex s.s. specimens from Gadus morhua, Gadus ogac, Reinhardtius hippoglossoides, Clupea harengus, Micromesistius poutassou, Pleurogrammus monopterygius, and Oncorhynchus nerka, caught in the north Atlantic and Pacific, were examined. Identity of the parasite species was confirmed using a molecular key. All specimens were used for amplifying a part of the non-coding sequence of the AT-rich region, flanked by the nad6-nad4L-trnW-trnE-rrnS-trnS2, and trnN-trnY-nad1 genes. This part of the mtDNA was chosen as a promising fragment, which could yield information regarding population diff…
