Parabacteroides distasonis regulates PPARɑ to alleviate colchicine-induced colonic injury through p-cresol sulfate.

Colchicine has been extensively utilized for treating gouty arthritis over many years, yet its dose-limiting gastrointestinal toxicity remains a significant clinical challenge. While diarrhoea and nausea are well recognised, the extent of colchicine-induced occult colonic injury-including its impact on the gut microbiota and mucosal homeostasis-is poorly defined. This study investigates colchicine-induced colonic injury (e.g., oxidative stress, mitochondrial dysfunction) in vivo and in vitro and also the imbalance of gut microbiota composition in mice. It revealed a significant decreased conte

Fenofibrate differentially activates PPARα-mediated lipid metabolism in rat kidney and liver.

Fenofibrate, a peroxisome proliferator-activated receptor α (PPARα) agonist, is widely prescribed to treat hyperlipidemia and has therapeutic potential in liver and kidney diseases. However, fenofibrate is also associated with adverse effects, including elevated creatinine and liver and kidney toxicity, although the underlying mechanisms remain unclear. In addition, how fenofibrate regulates lipid metabolism differently in the liver and kidney is not well understood. Therefore, in this study, we investigated the dose-dependent effects of fenofibrate on liver and kidney metabolism in rats, with

Melatonin alleviates cadmium-induced nonalcoholic fatty liver disease in ducks by alleviating autophagic flow arrest via PPAR-α and reducing oxidative stress.

Cadmium (Cd) is an important environmental pollutant that causes liver damage and induces nonalcoholic fatty liver disease (NAFLD). NAFLD is a fat accumulation disease and has significant effects on the body. Melatonin (Mel) is an endogenous protective molecule with antioxidant, anti-inflammatory, antiobesity, and antiaging effects. However, whether Mel can alleviate Cd-induced NAFLD and its mechanism remains unclear. First, in vivo, we found that Mel maintained mitochondrial structure and function, inhibited oxidative stress, and reduced Cd-induced liver injury. In addition, Mel alleviated li

Efficacy and Safety of K-877 (Pemafibrate), a Selective PPARα Modulator, in European Patients on Statin Therapy.

OBJECTIVE: High plasma triglyceride (TG) is an independent risk factor for cardiovascular disease. Fibrates lower TG levels through peroxisome proliferator-activated receptor α (PPARα) agonism. Currently available fibrates, however, have relatively low selectivity for PPARα. The aim of this trial was to assess the safety, tolerability, and efficacy of K-877 (pemafibrate), a selective PPARα modulator, in statin-treated European patients with hypertriglyceridemia. RESEARCH DESIGN AND METHODS: A total of 408 statin-treated adults were recruited from 68 European sites for this phase 2, randomized

Selective PPARδ agonist seladelpar suppresses bile acid synthesis by reducing hepatocyte CYP7A1 via the fibroblast growth factor 21 signaling pathway.

Peroxisome proliferator-activated receptor delta (PPARδ) agonists have been shown to exert beneficial effects in liver disease and reduce total bile acid levels. The mechanism(s) whereby PPARδ agonism reduces bile acid levels are, however, unknown, and therefore the aim of the present study was to investigate the molecular pathways responsible for reducing bile acid synthesis in hepatocytes, following treatment with the selective PPARδ agonist, seladelpar. We show that administration of seladelpar to WT mice repressed the liver expression of cholesterol 7 alpha-hydroxylase (Cyp7a1), the rate-l

Sulfated glucuronomannan hexasaccharide G6S1 enhanced lipolysis and lipophagy via PPARα pathway.

Nonalcoholic fatty liver disease (NAFLD) is considered as the hepatic manifestation of metabolic syndrome, ranging from benign steatosis to severe non-alcoholic steatohepatitis. Recently, it has been found that lipophagy plays a pivotal role in lipid turnover, which can alleviate NAFLD in hepatocytes. In this study, we found that a highly sulfated glucuronomannan hexamer G6S1 has the ability to enhance lipophagy. When treated with G6S1, the number and the size of lipid droplet (LD) decreased significantly on hepatocytes AML12 cells. Western blot results showed that the expressions of the lipol

Aspirin binds to PPARα to stimulate hippocampal plasticity and protect memory.

Despite its long history, until now, no receptor has been identified for aspirin, one of the most widely used medicines worldwide. Here we report that peroxisome proliferator-activated receptor alpha (PPARα), a nuclear hormone receptor involved in fatty acid metabolism, serves as a receptor of aspirin. Detailed proteomic analyses including cheminformatics, thermal shift assays, and TR-FRET revealed that aspirin, but not other structural homologs, acts as a PPARα ligand through direct binding at the Tyr314 residue of the PPARα ligand-binding domain. On binding to PPARα, aspirin stimulated hippo

Protective role of peroxisome proliferator-activated receptor α agonists in skin barrier and inflammation.

Peroxisome proliferator-activated receptor α (PPARα) is one of the three isoforms of PPARs, which are ligand-activated nuclear transcription factors. PPARα is highly expressed in liver and its agonists are widely used to treat hyperlipidemia. Epidermal keratinocytes express all three isoforms (α, β/δ, and γ) of PPARs and PPARα is particularly important for regulating the epidermal barrier and inflammation. Agonistic ligation of PPARα protects the epidermal barrier function and inhibits the inflammatory response in dermatitis. In this review, we summarize recent topics on the role of PPARα in s

PPARα agonist fenofibrate enhances fatty acid β-oxidation and attenuates polycystic kidney and liver disease in mice.

Peroxisome proliferator-activated receptor α (PPARα) is a nuclear hormone receptor that promotes fatty acid β-oxidation (FAO) and oxidative phosphorylation (OXPHOS). We and others have recently shown that PPARα and its target genes are downregulated, and FAO and OXPHOS are impaired in autosomal dominant polycystic kidney disease (ADPKD). However, whether PPARα and FAO/OXPHOS are causally linked to ADPKD progression is not entirely clear. We report that expression of PPARα and FAO/OXPHOS genes is downregulated, and in vivo β-oxidation rate of 3H-labeled triolein is reduced in Pkd1RC/RC mice, a

Population-based meta-analysis and gene-set enrichment identifies FXR/RXR pathway as common to fatty liver disease and serum lipids.

Nonalcoholic fatty liver disease (NAFLD) is prevalent worldwide. NAFLD is associated with elevated serum triglycerides (TG), low-density lipoprotein cholesterol (LDL), and reduced high-density lipoprotein cholesterol (HDL). Both NAFLD and blood lipid levels are genetically influenced and may share a common genetic etiology. We used genome-wide association studies (GWAS)-ranked genes and gene-set enrichment analysis to identify pathways that affect serum lipids and NAFLD. We identified credible genes in these pathways and characterized missense variants in these for effects on serum traits. We

Tocotrienol-rich fraction of vitamin E in diabetes and cardiovascular disease: From molecular mechanisms to clinical application.

BACKGROUND AND RATIONALE: Tocotrienol-rich fraction (TRF), a naturally occurring mixture of four tocotrienol isoforms (α-, β-, γ-, and δ-tocotrienol) derived principally from palm oil, constitutes a structurally and functionally distinct sub-family of vitamin E with demonstrably superior biological activity compared with α-tocopherol. Diabetes mellitus (DM) and its principal macrovascular complication, cardiovascular disease (CVD), remain the leading drivers of global morbidity and mortality, with an estimated 529 million adults currently affected and projections exceeding 1.3 billion by 2050.

Proximal Tubule-Specific Genetic Deficiency of PPARα Worsens Systemic Lipid and Glucose Metabolism During Fasting.

The proximal tubule (PT) of the kidney is a highly metabolic organ that regulates systemic homeostasis through ketogenesis and gluconeogenesis. Peroxisome proliferator-activated receptor alpha (PPARα), a nuclear receptor controlling fatty acid oxidation (FAO) and homeostasis, is expressed in the PT and is activated by fasting. Although the activation of systemic PPARα is essential for renal and systemic energy metabolism, the role of PPARα in PT has not been established. In this study, kidney PT-specific PPARα knockout mice (Ppara∆KPT) were generated, and the metabolic changes caused by 48 h o

Oleoylethanolamide enhances regulatory T Cell function to accelerate plaque regression in atherosclerosis via PPARα activation.

Regulatory T cells (Tregs) are essential for maintaining immune balance and limiting inflammatory damage within atherosclerotic plaques. Although the endogenous lipid mediator oleoylethanolamide (OEA) has reported anti-inflammatory and metabolic benefits, its effects on Treg differentiation and function during atherosclerosis are incompletely defined. Here, we tested OEA using in vitro naive CD4+ T-cell polarization assays and in vivo atherosclerosis models. OEA increased CD25+Foxp3+ Treg differentiation in polarization cultures and shifted the Treg compartment in atherosclerotic mice toward a
