Epidermal growth factor and basic fibroblast growth factor are induced in guinea-pig tympanic membrane following traumatic perforation.

Epidermal growth factor (EGF) and basic fibroblast growth factor (bFGF or FGF-2) have been shown to enhance the healing of traumatic tympanic membrane (TM) perforations. The action of EGF and bFGF in the TM repair process remains unknown. This study was designed to determine the expression of EGF and bFGF in normal and injured TM by immunohistochemistry. EGF was detected in normal TM mainly in the annulus tympani area. After a perforation in the TM posterior/superior quadrant, EGF was detected around the perforated area in polynuclear cells, in pericytes and in basal epithelial cells. EGF was …

Platelet-derived growth factor receptor β F7 mutations result in and exacerbate the severity of vascular dysplasia in the brain arteriovenous malformation through enhancing angiogenesis.

Brain arteriovenous malformations (bAVMs) are tangles of abnormal vessels that shunt blood directly from arteries to veins. The reduction of pericytes is linked to hemorrhage in bAVMs. The Pdgfb/Pdgfrβ signaling pathway is crucial for regulating pericyte recruitment during angiogenesis. Here, we show that mice with Pdgfrβ F7 mutations developed cerebrovascular dysplasia in the brain’s angiogenic region, associated with increased pro-angiogenic and inflammatory signaling. Interestingly, the expression of activin receptor-like kinase 1 (Alk1) is decreased in the brain’s angiogenic region of Pdgf…

The generation of stable microvessels in ischemia is mediated by endothelial cell derived TRAIL.

Reversal of ischemia is mediated by neo-angiogenesis requiring endothelial cell (EC) and pericyte interactions to form stable microvascular networks. We describe an unrecognized role for tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) in potentiating neo-angiogenesis and vessel stabilization. We show that the endothelium is a major source of TRAIL in the healthy circulation compromised in peripheral artery disease (PAD). EC deletion of TRAIL in vivo or in vitro inhibited neo-angiogenesis, pericyte recruitment, and vessel stabilization, resulting in reduced lower-limb blood perf

Antigen-antibody reactions in allergic human tissues. II. Study by fluorescence technique of the localization of reagins in human skin and their relation to globulins.

The skin of atopic patients contains specific reaginic antibodies in all epidermal cells, in unchallenged as well as antigen-challenged areas. The reagins are also present in the epithelial cells of sebaceous glands, sweat glands, hair follicles and also in macrophages and pericytes. Judged by staining characteristics, their immunologic reaction in tissues with antigen is identical with that of rabbit antibodies. Comparison of tissues stained with conjugated reagins and conjugated anti-human globulin antibodies demonstrates the close relationship of reagins and globulins. In antigen-challenged…

Alpha-Smooth Muscle Actin-Positive Perivascular Cells in Diabetic Retina and Choroid.

Structural alterations of pericytes in microvessels are important features of diabetic retinopathy. Although capillary pericytes had been known not to have α-smooth muscle actin (αSMA), a recent study revealed that a specific fixation method enabled the visualization of αSMA along retinal capillaries. In this study, we applied snap-fixation in wild type and streptozotocin-induced diabetic mice to evaluate the differences in vascular smooth muscle cells of the retina and the choroid. Mice eyeballs were fixed in ice-cold methanol to prevent the depolymerization of filamentous actin. Snap-fixated…

A Novel Transwell Blood Brain Barrier Model Using Primary Human Cells.

Structural alterations and breakdown of the blood brain barrier (BBB) is often a primary or secondary consequence of disease, resulting in brain oedema and the transport of unwanted substances into the brain. It is critical that effective in vitro models are developed to model the in vivo environment to aid in clinically relevant research, especially regarding drug screening and permeability studies. Our novel model uses only primary human cells and includes four of the key cells of the BBB: astrocytes, pericytes, brain microvascular endothelial cells (HBMEC) and neurons. We show that using a …

Laminin and collagen modulate expression of the small leucine-rich proteoglycan fibromodulin in rat anterior pituitary gland.

The anterior pituitary is a complex organ consisting of five types of hormone-producing cells, non–hormone-producing cells such as folliculostellate (FS) cells and vascular cells (endothelial cells and pericytes). We have previously shown that FS cells and pericytes produce fibromodulin, a small leucine-rich proteoglycan (SLRP). SLRPs are major proteoglycans of the extracellular matrix (ECM) and are important in regulating cell signaling pathways and ECM assembly. However, the mechanism regulating fibromodulin expression in the anterior pituitary has not been elucidated. Here, we investigate w…

Aging of the Blood-Brain Barrier and Altered Permeability to Peripheral Immune Cells: Implications for Central Nervous System Disorders.

The blood-brain barrier (BBB), built by endothelial tight junctions, transporters, and the endothelial-pericyte-astrocyte unit, maintains a tightly regulated the central nervous system (CNS) milieu and permits selective immune cell entry. With age, tight junction downregulation, transport and metabolic imbalance, pericyte loss, basement membrane and glycocalyx remodeling, and reactive astrocytosis erode BBB integrity. These shifts reset leukocyte-trafficking thresholds and routes, producing either excessive admission or inadequate entry of peripheral immune cells. The resulting dysregulated im

Loss of Vascular TMEM16A Impairs Cerebral Autoregulation and Exacerbates Ischemia-reperfusion Injury.

TMEM16A forms a Ca²⁺-activated Cl⁻ channel in vascular mural cells (smooth muscle cells and pericytes) that generates depolarizing Cl⁻ efflux upon intracellular Ca²⁺ elevation, thereby amplifying agonist-induced vasoconstriction. TMEM16A has been implicated in excessive capillary pericyte constriction following cerebral ischemia, suggesting that its inhibition may improve post-stroke recovery. However, the impact of systemic vascular TMEM16A inhibition on focal reperfusion efficiency and cerebrovascular autoregulation remains unknown. To address this question, mice with inducible mural cell-sp…

Deletion of RGS5 attenuates high-fat diet-induced cerebrovascular pathology and associated spatial memory deficits.

Diet-induced metabolic dysregulation is associated with cerebral microvascular pathology contributing to cognitive decline and vascular dementia. These alterations include blood-brain barrier (BBB) leakage, pericyte dysfunction, aberrant angiogenesis and perivascular neuroinflammation. Restoration of BBB integrity has been achieved in other conditions by targeting pericytes, for example through knockout of Regulator of G-protein Signaling 5 (RGS5), a sensor protein for hypoxia and oxidative stress highly enriched in these cells. It remains unclear whether deletion of RGS5 can provide vascular …

Cerebellar capillary hemangioblastoma: its histogenesis studied by organ culture and electron microscopy.

A capillary hemangioblastoma from the vermis of a 21-year-old man with von Hippel-Lindau's disease was successfully maintained in organ culture systems on Millipore filter platforms and gelatin sponge foam matrices up to 48 days. Three cell types--endothelial cells, pericytes, and stromal cells--identified by their fine structural features and their architectural relationships to vascular lumens and to the extracellular space were recognized in the original tumor. By light microscopy the explants showed, in the late stages, increased lipid droplets in the stromal cells and perivascular hyaline…

Embryonic development of the CNS microvasculature in the mouse: new insights into the structural mechanisms of early angiogenesis.

Vascularization and expression of blood-brain barrier (bbb) associated morphological characteristics were studied by ultrastructural analysis during the embryonic development of the mouse CNS. At day 9 (E9) capillaries were only found in the perineural mesenchymal tissue exhibiting fenestrations and loosely attached pericytes. At E10 endothelial cells (EC) together with pericytes and other mesenchymal cells invaded the intraneural domain. The immigrating EC showed numerous protrusions and lost their fenestrations as soon as the new intraneural capillaries were formed. Intraneural EC showed pro…

Isolation, Culture, and Characterization of Vascular Smooth Muscle Cells.

Smooth muscle cells (SMC) are the predominant cell type involved in the pathogenesis of atherosclerosis, vascular calcification and restenosis after angioplasty; however, they are also important in the de novo formation of blood vessels through differentiation of mesenchymal cells under the influence of mediators secreted by endothelial cells. In angiogenesis, vascular SMC are formed by proliferation of existing SMC or maturation and differntiation of pericytes. Experimental findings have demonstrated a potential role of putative smooth muscle progenitor cells in the circulation or within adul…

Increased vascular endothelial growth factor expression in patients with bladder pain syndrome/interstitial cystitis: its association with pain severity and glomerulations.

OBJECTIVE: To determine the angiogenic profiles in the bladder of patients with bladder pain syndrome (BPS)/interstitial cystitis (IC), and to evaluate the relationship between these profiles and associated clinical features including pelvic pain and glomerulations. PATIENTS AND METHODS: Angiogenesis and angiogenic components are important in chronic inflammatory disease. High levels of vascular endothelial growth factor (VEGF) have been shown to induce immature angiogenesis, where microvessels have insufficient coverage of pericytes, resulting in haemorrhagic vessels. Biopsy specimens from 3…

[Alveolar Soft Part Sarcoma-The Angiogenic Mechanism Regulated by a Fusion Gene Product].

Alveolar soft part sarcoma(ASPS)is a rare malignant tumor whose origin is unidentified, arising from deep soft tissue and affecting adolescents and young adults. ASPS is characterized by its abundant vascular network forming alveolar structures, and demonstrates frequent hematogenous metastasis. An ASPSCR1-TFE3 fusion gene derived from t(X;17)chromosome translocation is detected as a disease gene in all cases, and the ASPSCR1-TFE3 protein causes abnormal transcriptional regulation. We generated a mouse model for ASPS by introducing ASPSCR1-TFE3 into mouse embryonic mesenchymal cells. In the mo

Angiotropic malignant melanoma and extravascular migratory metastasis: description of 36 cases with emphasis on a new mechanism of tumour spread.

AIMS: We have identified in malignant melanoma an angio-tumoural complex in which tumour cells occupy a pericytic location along the endothelium of microvessels without evidence of intravasation. This pericytic angiotropism of melanoma cells, without any sign of intravasation, suggests that melanoma cells may migrate along the external surface of vessels, a mechanism we have termed 'extravascular migratory metastasis' (EVMM), as distinct from intravascular dissemination. METHODS: The present study describes, for the first time, a series of 36 invasive melanoma cases ascertained for the presen

The phosphorylation of moesin impairs the integrity of vascular basement membrane in pathological angiogenesis.

Our previous studies demonstrated that advanced glycation end products (AGEs) promote angiogenesis in human umbilical vein endothelial cells (HUVECs) and mouse retina through moesin phosphorylation. AGEs-induced angiogenesis is characterized by abnormal VE-cadherin distribution at adherens junctions and decreased pericytes coverage. In this study, we further investigated the alterations in collagen IV (Col-IV) distribution within the basement membrane (BM) of neovesssles using a HUVECs-retinal microvascular pericytes (RMPs) co-culture system and an AGEs-treated mouse model. The role of moesin …

"One stone and two birds" strategy to treat neovascular age-related macular degeneration by a novel retinoid drug, EYE-101.

Choroidal neovascularization (CNV) is a typical pathological feature of neovascular age-related macular degeneration and has become a major … s endothelial cells. EYE-101 exerted its anti-angiogenic effects by targeting endothelial cells and pericytes via antagonizing Wnt/β-catenin signaling and PDGF signaling. Thus, EYE-101 administration may offer an"one stone and two birds" strategy for the prevention and treatment of ocular neovascular disorders.
