BACKGROUND: Dofetilide is an antiarrhythmic drug that blocks the cardiac repolarizing current IKr ((IKr, rapid component of the delayed rectifying potassium current). BACKGROUND: The discovery of calcium receptors and calcimimetics created the possibility of "pharmacologic parathyroidectomy" (phPTX), which decreased secretion of parathormone (PTH). Parathyroid glands of spontaneously hypertensive rats (SHR) and of patients with primary hyperparathyroidism and hypertension secrete parathyroid hypertensive factor (PHF). Parathyroidectomy decreases blood pressure in these rats and in patients. The present study determined whether phPTX induced by calcimimetics decreases mean arterial blood pressure (MAP) in hypertensive rats.

Fourteen dry eye volunteers placed one to two drops of 0.15% AF2975 (N,N-dimethyl-2-phenylethylamine HCl) in one eye and the vehicle in their other eye four times a day for 21 days. The effects of three psychomotor stimulants (mazindol, beta-phenylethylamine and D-phenmetrazine) on electrically evoked neostriatal dopamine release were studied by in vivo voltammetry. Mazindol (10 mg/kg) enhanced release and this effect persisted after dopamine synthesis inhibition by alpha-methyl-p-tyrosine. beta-Phenylethylamine (100 mg/kg) caused a large decrease in stimulated dopamine release and exerted no effect after dopamine synthesis inhibition.
