Soil salinity severely threatens plant growth and crop performance. Hydrogen sulfide (H2S), a plant signal molecule, has been implicated in the regulation of plant responses to salinity stress. However, it is unclear how the transcriptional network regulates H2S biosynthesis during salt stress response. In this study, we identify a rice NAC (NAM, ATAF and CUC) transcription factor, OsNAC35-like (OsNACL35), from a salt-tolerant cultivar 'Sea Rice 86' (SR86) and further show that it may have improved salt tolerance via enhanced H2S production. The expression of OsNACL35 was significantly upregul

The ubiquitin-proteasome system (UPS) constitutes a highly conserved regulatory hub governing protein turnover and signal transduction in eukaryotes, which precisely determines the fate of substrate proteins via dynamic and reversible ubiquitination. During long-term coevolution between plants and viruses, the UPS has evolved into a critical battlefield for host-virus arms races. Plants exploit the substrate recognition specificity and proteolytic activity of the UPS to selectively eliminate essential viral proteins required for infection, thereby establishing multilayered antiviral immune bar

BACKGROUND: Morus mongolica holds significant value in medicinal applications, livestock fodder production, and ecological restoration. Although polyploidization is well-documented in plants for enhancing stress tolerance and altering nutrient composition, the specific beneficial traits conferred by genome duplication in M. mongolica have not yet been fully characterized.
RESULTS: Our results demonstrated that a 72 h preculture followed by 72 h of colchicine treatment (30 mg·L-¹), yielded a tetraploid rate of 25%. Significant differences were observed between diploid and tetraploid plants in

Soybean (Glycine max (L.) Merr.) yields have increased with advances in plant breeding and management practices, including earlier planting. Early planting in temperate climates maximizes yield potential but may increase vulnerability to certain pests and environmental stressors. Neonicotinoid seed treatments, particularly thiamethoxam, are widely marketed to protect soybean seedlings, yet their effectiveness under typical early-season field conditions in Indiana remains unexplored. This 2-year (2023 to 2024) study was conducted at 3 sites in Indiana. Soybeans were planted during 4 discrete pe…

Malnutrition has emerged as one of the major health problems worldwide. Traditional yellow maize has low provitamin-A (proA) content and its genetic base in proA biofortification breeding program of subtropics is extremely narrow. To diversify the proA rich germplasm, 10 elite low proA inbreds were crossed with a proA rich donor (HP702-22) having mutant crtRB1 gene. The F2 populations derived from these crosses were genotyped using InDel marker specific to crtRB1. Severe marker segregation distortion was observed. Seventeen crtRB1 inbreds developed through marker-assisted pedigree breeding and

Interactions between host nutrition and feeding behaviour are central to understanding the pathophysiological consequences of infections of the digestive tract with parasitic nematodes. The manipulation of host nutrition provides useful options to control gastrointestinal nematodes as a component of an integrated strategy. Focussed mainly on the Haemonchus contortus infection model in small ruminants, this chapter (1) illustrates the relationship between quantitative (macro- and micro-nutrients) and qualitative (plant secondary metabolites) aspects of host nutrition and nematode infection, and

Copper (Cu) is an essential micronutrient for plants that functioning as a cofactor in numerous enzymes. However, it becomes toxic in excess, necessitating homeostatic mechanisms. In this review, we synthesize current knowledge of the Cu uptake, transport, and homeostasis in plants, and examine the Cu-mediated regulation of plant secondary metabolism. This review outlines the forms of Cu present in soils and plant systems and describes how roots acquire Cu (predominantly as Cu²⁺, which is then reduced to Cu⁺ at the root surface) via high-affinity transporters. Within the plant, the Cu uptake a

High temperature is a primary abiotic stress that severely constrains crop productivity. Deciphering the regulatory pathways underlying heat responses is essential for breeding heat-tolerant crops with stable yields. Although both epigenetic and epitranscriptomic regulations are involved in plant heat adaptation, their mechanistic interplay remains unclear. Here, integrated epigenomic (H3K9Ac/H3K14Ac) and transcriptomic profiling under heat stress identifies the mRNA m⁵C methyltransferase TaNSUN2 as a key regulator of thermotolerance in wheat. We demonstrate that TaNSUN2 is transcriptionally a

Grapevine (Vitis vinifera) is one of the most widely cultivated plant species of agricultural interest, and is extensively appreciated for its fruits and the wines made from its fruits. Considering the high socio-economic impact of the wine sector all over the world, in recent years, there has been an increase in work aiming to investigate the biodiversity of grapevine germplasm available for breeding programs. Various studies have shed light on the genetic diversity characterizing the germplasm from the cradle of V. vinifera domestication in Georgia (South Caucasus). Georgian germplasm is pla

Wheat (Triticum aestivum L.) remains one of the most important staple crops worldwide and serves as a primary source of calories and protein for billions of people. Global wheat production, however, continues to be threatened by rust diseases caused by Puccinia triticina, Puccinia striiformis f. sp. tritici and Puccinia graminis f. sp. tritici. These pathogens are highly adaptive and frequently give rise to new virulent races, causing resistance in wheat cultivars to breakdown. Under favorable environmental conditions, severe epidemics can develop and substantial yield losses may occur. In res

In arid-region wheat breeding, environmental fluctuations are large, and screening based on single traits or single-year data is often inaccurate. To address these challenges, this study constructed a multi-model collaborative evaluation system to accurately screen advanced lines for high yield potential and inter-annual consistency. Based on 10 yield traits evaluated in 52 advanced wheat lines over two growing seasons, we utilized systematic clustering to classify lines and stepwise regression to pinpoint core contributing traits. Subsequently, principal component analysis (PCA), membership f

Screening global wheat germplasm with a diverse collection of pathogen races expands the catalog of novel Yr loci and identifies new sources of broad-spectrum resistance against evolving Pst populations. Newly emerging highly virulent races of Puccinia striiformis f. sp. tritici (Pst) often defeat deployed resistance genes (Yr), highlighting the need for novel sources of durable resistance. A global diversity panel of 377 spring wheat (Triticum aestivum L.) lines was screened for all-stage resistance (ASR) against a panel of diverse 20 Pst isolates at the seedling stage and for adult-plant-sta

The dynamic flow of heavy metals in ecosystems is disturbing the sustainability of environmental quality and the flow of resources in ecosystems. Detection of genotypes with metal-tolerance ability is a prerequisite for mitigating this burning problem, through restoration of degraded ecosystems, progressive crop productivity, and improvement in efficiency of phytoremediation programs. The application of simple sequence repeat (SSR) markers for tracing metal tolerance in plants is gaining importance among researchers. SSRs, as molecular markers, are highly informative and preferable compared to

Understanding the frequency and distribution of meiotic crossovers (COs) is critical for both fundamental studies on meiosis and for practical applications in plant breeding, where controlling recombination can accelerate crop improvement. Determining CO rates at specific genomic loci has traditionally relied on labor-intensive methods that require the production and genotyping of large progenies. Here, we present a high-throughput protocol for site-specific quantification of meiotic COs in maize using droplet digital PCR (ddPCR). The method is based on genotyping individual pollen nuclei from…

Tea plants (Camellia sinensis) suffer growth limitations under cold stress. Jasmonic acid (JA) and long non-coding RNAs (lncRNAs) are involved in stress responses, yet how lncRNAs regulate JA-mediated cold tolerance remains unclear. Here, we identified an lncRNA, lnc015013, whose silencing compromised cold tolerance in tea plants, a phenotype rescued by exogenous methyl jasmonate (MeJA). Silencing lnc015013 down-regulated CsMYB30 and CsJAZ4/6, while its overexpression had opposite effects. Heterologous expression in Arabidopsis thaliana showed that CsMYB30 enhanced cold resistance, whereas CsJ

Carotenoids are indispensable for human health, required as precursors of vitamin A and efficient antioxidants. However, these plant pigments that play a vital role in photosynthesis are represented at insufficient levels in edible parts of several crops, which creates a need for increasing their content or optimizing their composition through biofortification. In particular, vitamin A deficiency, a severe health problem affecting the lives of millions in developing countries, has triggered the development of a series of high-provitamin A crops, including Golden Rice as the best-known example.

One of the most useful techniques for studying genetic variations in plants is single-nucleotide polymorphisms (SNPs), which are useful in developing molecular markers for genetic research. In this chapter, the development and validation of SNP markers using targeted resequencing and in silico analysis of genomic resources for plant genomic research and breeding are included. Key steps include genomic DNA isolation, whole-genome resequencing, raw read processing, reference-based alignment, SNP detection, and stringent filtering to identify high-confidence variants. The importance of cross-plat

Global food security is one of the utmost essential challenges in the 21st century in providing enough food for the growing population while coping with the already stressed environment. High temperature (HT) is one of the main factors affecting plant growth, development and reproduction and causes male sterility in plants. In male reproductive tissues, metabolic changes induced by HT involve carbohydrates, lipids, hormones, epigenetics and reactive oxygen species, leading to male sterility and ultimately reducing yield. Understanding the mechanism and genes involved in these pathways during t

Wheat grown in Mn-deficient soil has been widely observed to produce much reduced yields. Breeding for Mn-efficient wheat genotypes adapted to Mn-deficient soils would represent a long-term solution for wheat agronomy. To characterize the physiological basis of Mn efficiency in wheat genotypes would facilitate the breeding programs for producing Mn-efficient wheat. Using a solution culture and a soil culture system in the present study, a Mn-efficient UK wheat genotype Maris Butler and a Mn-inefficient UK wheat genotype Paragon have been compared with a Mn-efficient Australian wheat genotype C

Drought inhibits grass development and survival. However, molecular-based studies on drought tolerance mechanisms in bermudagrass (Cynodon dactylon) remain scarce. Therefore, a drought-resistant bermudagrass (Tianshui) and a drought-sensitive (Zhengzhou) genotype were selected and subjected to 28 days of 50% drought stress. Leaves were sampled for RNA sequencing. Under drought stress, 2410 differentially expressed genes (DEGs) were discovered in which 1214 were upregulated (tolerant vs. sensitive) and 1196 downregulated. Kyoto Encyclopedia of Genes and Genomes (KEGG) indicated that these speci
