Nitric oxide: A key player in plant defense against biotic stress.

Nitric oxide (NO), a small yet highly reactive gaseous molecule, has emerged as a crucial signaling component in plant biology, mainly in mediating its defense responses against biotic stress. Apart from mammalian role of NO, it also actively involved in the broad physiological, biochemical, and immunological processes in plants. NO is synthesized through a set of multiple enzymatic and non-enzymatic pathways, whereupon it rapidly accumulates upon pathogen recognition. NO is implicated in plant immunity by working in synergy with reactive oxygen species (ROS) in triggering the hypersensitive r…

Exl1 from Pectobacterium activates a jasmonate-dependent immune network in Arabidopsis thaliana.

Bacterial expansins contribute to the virulence of diverse plant pathogens, yet their biological activity and molecular roles during host interactions remain to be elucidated. Expansin-like protein, Exl1, from phytopathogens from the genus Pectobacterium, activates defense responses in Arabidopsis thaliana involving production of reactive oxygen species (ROS), and the jasmonic acid (JA), ethylene (ET), and salicylic acid (SA) pathways, ultimately reducing disease symptoms upon subsequent infections with P. brasiliense and the fungus Botrytis cinerea. However, the target of Exl1 within the plan

Knowledge on plants used traditionally in the treatment of tuberculosis in Uganda.

ETHNOPHARMACOLOGICAL RELEVANCE: Tuberculosis (TB) is one of the leading causes of morbidity and mortality globally. The disease is especially important because of increasing drug resistant strains and co infection with human immune virus (HIV) and acquired immune disease syndrome (AIDS). Because of this there is need to identify new leads that can be developed into new drugs. The objectives of this study were to (1) document plant species commonly used by traditional medicine practitioners (TMPs) to treat TB, methods of preparation and administration of drugs (2) document disease recognition b

Rheumatoid Arthritis and Secondary Plant Metabolites: An Analysis.

Chronic rheumatoid arthritis (RA) is a systemic inflammatory disease characterized by persistent joint inflammation, progressive joint destruction, and chronic pain. Although modern therapies like disease-modifying antirheumatic medications (DMARDs) can alleviate symptoms, they may also produce side effects. Because of their anti-inflammatory, antioxidant, and immunomodulatory qualities, plant secondary metabolites such as flavonoids, terpenoids, alkaloids, and phenolic acids have attracted attention as prospective RA treatment agents. This review discusses the pathogenesis of RA and provides

Common Ragweed Allergy Under Global Change Linking Invasion-Driven Aeroallergen Exposure with Molecular Sensitization and Allergic Airway Disease.

The case of common ragweed (Ambrosia artemisiifolia) stands as a prime example of an invasive plant that links global change, biological invasion, aeroallergen exposure, and allergic airway disease. However, evidence remains fragmented across invasion biology, aerobiology, molecular allergology, and respiratory medicine. By separating plant occurrence, pollen abundance, molecular allergen dose, sensitisation, and airway disease, this review develops an integrated invasion-exposure-disease logic for interpreting ragweed-related health risk. We examine how climate change, land-use disturbance, r

Plant-produced Recombinant Influenza A Vaccines Based on the M2e Peptide.

BACKGROUND: Influenza is a widely distributed infection that almost annually causes seasonal epidemics. The current egg-based platforms for influenza vaccine production are facing a number of challenges and are failing to satisfy the global demand in the case of pandemics due to the long production time. Recombinant vaccines are an alternative that can be quickly produced in high quantities in standard expression systems. METHODS: Plants may become a promising biofactory for the large-scale production of recombinant proteins due to low cost, scalability, and safety. Plant-based expression sys

The DELLA Protein SLR1 Integrates and Amplifies Salicylic Acid- and Jasmonic Acid-Dependent Innate Immunity in Rice.

Gibberellins are a class of tetracyclic plant hormones that are well known to promote plant growth by inducing the degradation of a class of nuclear growth-repressing proteins, called DELLAs. In recent years, GA and DELLAs are also increasingly implicated in plant responses to pathogen attack, although our understanding of the underlying mechanisms is still limited, especially in monocotyledonous crop plants. Aiming to further decipher the molecular underpinnings of GA- and DELLA-modulated plant immunity, we studied the dynamics and impact of GA and DELLA during infection of the model crop ric…

Cysteine Signalling in Plant Pathogen Response.

The amino acid cysteine is the precursor for a wide range of sulfur-containing functional molecules in plants, including enzyme cofactors and defence compounds. Due to its redox active thiol group cysteine is highly reactive. Synthesis and degradation pathways are present in several subcellular compartments to adjust the intracellular cysteine concentration. However, stress conditions can lead to a transient increase in local cysteine levels. Here we investigate links between cysteine homeostasis and metabolic signalling in Arabidopsis thaliana. The systemic proteome response to cysteine feedi

Choosing the Right Extracellular Vesicle: Cross-Kingdom Immunological Functions Linking Molecular Mechanisms to Therapeutic Applications.

Extracellular vesicles (EVs) are key mediators of intercellular communication across biological kingdoms, with central roles in immune regulation and disease processes. Despite shared structural features, EVs derived from bacteria, plants, and mammalian cells differ substantially in their biogenesis, molecular composition, and immunological functions. EV formation pathways generate vesicles with distinct cargo profiles, including pathogen-associated molecular patterns (PAMPs) in bacterial EVs, regulatory small RNAs in plant-derived vesicles, and cytokines, microRNAs, and antigen-presenting com

Volatile Compounds Emitted by Plant Growth-Promoting Fungus Tolypocladium inflatum GT22 Alleviate Copper and Pathogen Stress.

Previous studies on the intricate interactions between plants and microorganisms have revealed that fungal volatile compounds (VCs) can affect plant growth and development. However, the precise mechanisms underlying these actions remain to be delineated. In this study, we discovered that VCs from the soilborne fungus Tolypocladium inflatum GT22 enhance the growth of Arabidopsis. Remarkably, priming Arabidopsis with GT22 VCs caused the plant to display an enhanced immune response and mitigated the detrimental effects of both pathogenic infections and copper stress. In Vitro regeneration of bio-immunized banana cv. Grand Naine using a novel double-decker temporary immersion bioreactor.

Considering the havoc caused by Fusarium oxysporumf. sp. cubense (TR4) at the global level on banana cultivation, especially in India, there is a high demand for an economical bio-immunized tissue culture technique for the sustainable production of disease-resistant plantlets, wherein bio-immunization is achieved through treatment of tissue culture-derived plants with antifungal secondary metabolites to mitigate the impact of Fusarium wilt. In the current investigation, a novel, user-friendly, and cost-effective Double Decker Temporary Immersion Bioreactor (DDTIB) was designed and optimized fo

The Mitogen-Activated Protein Kinase GhNTF3 Modulates the Arbuscular Mycorrhizal Symbiosis-Immunity Trade-Off in Cotton by Regulating Salicylic Acid Biosynthesis.

Plant immunity is essential for survival against pathogen invasion. However, enhanced immune activation can restrict arbuscular mycorrhizal (AM) fungal colonization. Therefore, plants must finely balance immunity and symbiosis to optimize fitness, but the key regulators underlying this trade-off remain unclear. Here, we identify a novel mitogen-activated protein kinase, GhNTF3, as a central regulator of the AM symbiosis-immunity balance. Knockdown of GhNTF3 promotes AM symbiosis but decreases Verticillium wilt resistance in cotton. GhNTF3 overexpression suppresses AM symbiosis. GhNTF3 interact…

Short-term dietary changes can result in mucosal and systemic immune depression.

Omnivorous animals, including mice and humans, tend to prefer energy-dense nutrients rich in fat over plant-based diets, especially for short periods of time, but the health consequences of this short-term consumption of energy-dense nutrients are unclear. Here, we show that short-term reiterative switching to 'feast diets', mimicking our social eating behavior, breaches the potential buffering effect of the intestinal microbiota and reorganizes the immunological architecture of mucosa-associated lymphoid tissues. The first dietary switch was sufficient to induce transient mucosal immune depre

Thienopyrimidine-type compounds protect Arabidopsis plants against the hemibiotrophic fungal pathogen Colletotrichum higginsianum and bacterial pathogen Pseudomonas syringae pv. maculicola.

Plant activators activate systemic acquired resistance-like defense responses or induced systemic resistance, and thus protect plants from pathogens. We screened a chemical library composed of structurally diverse small molecules. We isolated six plant immune-inducing thienopyrimidine-type compounds and their analogous compounds. It was observed that the core structure of thienopyrimidine plays a role in induced resistance in plants. Furthermore, we highlight the protective effect of thienopyrimidine-type compounds against both hemibiotrophic fungal pathogen, Colletotrichum higginsianum, and b

Advances in alfalfa mosaic virus-mediated expression of anthrax antigen in planta.

Plant viruses show great potential for production of pharmaceuticals in plants. Such viruses can harbor a small antigenic peptide(s) as a part of their coat proteins (CP) and elicit an antigen-specific immune response. Here, we report the high yield and consistency in production of recombinant alfalfa mosaic virus (AlMV) particles for specific presentation of the small loop 15 amino acid epitope from domain-4 of the Bacillus anthracis protective antigen (PA-D4s). The epitope was inserted immediately after the first 25 N-terminal amino acids of AlMV CP to retain genome activation and binding of

Reduced interleukin-2 production and increased CREMα protein expression in vegetarians and vegans due to zinc deficiency.

Nutrition is a key determinant of health and may be regarded as a form of preventive medicine, as an adequate supply of vitamins, fats, proteins, and trace elements is essential for proper immune function. In recent decades, vegetarian and vegan diets have become increasingly popular but may increase the risk of trace element deficiencies if not carefully planned. Zinc deficiency can impair immune responses and reduce resistance to infections. While previous research has mainly focused on animal models or elderly populations, little is known about younger adults. Thus, we investigated the impa

Deciphering the genomic and functional basis of virulence and immune modulation in Pantoea stewartii.

Pantoea stewartii subsp. indologenes is an emerging cereal phytopathogen with unclear virulence mechanisms. We sequenced a rice seed-borne strain, HR3-48, and classified it as P. stewartii subsp. indologenes (Pnsi) through phylogeny of 92 housekeeping genes, ANI (> 99%), and dDDH (> 96%) comparisons. HR3-48 harbors a conserved plant-pathogenic PSI-1 type III secretion system (T3SS) and a single flagellar locus (flag-1) with multiple fliC genes. The tandem fliC456 cassette is functionally redundant for motility and flagellum assembly but contributes significantly to eliciting plant immune respo
