Targeting the MNK1-MYH9 axis blocks YAP1 recruitment to prevent thrombosis and platelet activation-induced NETosis.

Platelet activation and neutrophil extracellular trap (NET) formation are critical drivers of immunothrombosis, yet the molecular crosstalk governing these processes remains incompletely understood. In this study, we unveil a distinct regulatory mechanism whereby Yes-associated protein 1 (YAP1) binds to non-muscle myosin heavy chain 9 (MYH9) to drive platelet activation and thrombosis. Interestingly, we identified MAPK-interacting kinase 1 (MNK1) as a natural competitive "structural shield" in this process. MNK1 physically interacts with MYH9 in resting platelets, thereby blocking the YAP1-bin…

Interferon receptor gene dosage differentially regulates hypoxia-induced platelet activation and pulmonary hypertension in down syndrome.

Trisomy 21 (T21) results in Down syndrome (DS), a condition associated with a high prevalence of pulmonary complications. Pulmonary hypertension (PH) is a significant comorbidity in individuals with T21. Inflammation is a well-established driver of PH, and growing evidence implicates platelets as active contributors to inflammatory-mediated pulmonary vascular disease and PH. In T21, increased interferon (IFN) signaling resulting from an additional IFN receptor gene locus contributes to immune dysregulation, and interactions between platelet activation and IFN signaling may promote thromboinfla…

Ferulenol, a Prenylated Coumarin, Suppresses Collagen-Induced Platelet Activation via PLCγ2-Mediated cPLA2 Signaling in Humans.

BACKGROUND: Cardiovascular diseases (CVDs) are the leading cause of morbidity and mortality worldwide and are strongly associated with atherothrombotic events. Thrombus formation results from the interplay between platelet activation and the coagulation cascade. While anticoagulant systems are essential for maintaining hemostasis, these systems can also increase the risk of bleeding complications. Coumarins led to the development of clinically important oral anticoagulants such as warfarin, which inhibits vitamin K epoxide reductase complex subunit 1 (VKORC1), a key enzyme in vitamin K recycli…

Ursodeoxycholic acid inhibits platelet activation and thrombosis via TREM2: Evidence from mouse models and human studies.

BACKGROUND AND PURPOSE: Current antiplatelet therapies effectively prevent thrombosis but are associated with an increased risk of bleeding, highlighting the need for safer alternatives. Ursodeoxycholic acid (UDCA) is a bile acid derivative with an established clinical safety profile, but its effects on platelet function and thrombosis remain poorly defined. EXPERIMENTAL APPROACH: The effects of UDCA on platelet activation were assessed in human and mouse platelets in vitro and ex vivo. Antithrombotic efficacy and bleeding risk were evaluated in murine models of mesenteric arteriole injury an…

IL-18 potentiates platelet activation and thrombosis through IL-18Rα-dependent MAPKs and PI3K/Akt signaling.

Arterial thrombosis is a leading cause of global cardiovascular mortality. Clinical evidence indicates that elevated IL-18 levels independently predict cardiovascular events, yet its direct role in thrombosis is poorly understood. Hence, targeting IL-18 could be a novel therapeutic strategy. This study investigated the mechanism of IL-18 in platelet activation and evaluated the antithrombotic efficacy of interleukin-18 binding protein (IL-18BP). Through in vitro and in vivo models, including Il18r1-/- mice, we demonstrated that IL-18 enhances platelet activation and thrombus formation via its …

Dioscin from smilax china rhizomes inhibits platelet activation and thrombus formation via up-regulating cyclic nucleotides.

Cardiovascular disease, the leading cause of mortality in the United States, is caused by abnormal platelet accumulation and coagulation. Dioscin has been reported to suppress the growth of tumor-associated cells and trigger apoptosis. However, its mechanism in inhibiting platelet activation has not been confirmed. This study investigates whether dioscin from Smilax china rhizomes exerts antithrombotic effects by regulating the activation of human platelets and explains its mechanism of action. Dioscin increased the production of cyclic adenosine monophosphate (cAMP) and cyclic guanosine monop…

SHIP1 agonist rosiptor inhibits platelet activation and thrombosis by modulating cAMP/cGMP and PI3K/PKC pathway.

Normal platelet activation is critical for proper hemostasis, while abnormal activation can lead to thrombotic diseases. Therefore, searching for substances that can regulate platelet activation will be helpful in preventing and treating thrombotic diseases. Studies have pointed out that SHIP1 can regulate platelet activation, affecting clot retraction and integrin αIIbβ3 activation. Nevertheless, the role of the SHIP1 agonist rosiptor in regulating platelet functions has not been investigated. This study aimed to investigate the antiplatelet and antithrombotic effects of rosiptor. Human plate…

Targeting Rap1b signaling cascades with CDNF: Mitigating platelet activation, plasma oxylipins and reperfusion injury in stroke.

Cerebral reperfusion injury in stroke, stemming from interconnected thrombotic and inflammatory signatures, often involves platelet activation, aggregation and its interaction with various immune cells, contributing to microvascular dysfunction. However, the regulatory mechanisms behind this platelet activation and the resulting inflammation are not well understood, complicating the development of effective stroke therapies. Utilizing animal models and platelets from hemorrhagic stroke patients, our research demonstrates that human cerebral dopamine neurotrophic factor (CDNF) acts as an endoge…

Molecular Insights into the Relationship Between Platelet Activation and Endothelial Dysfunction: Molecular Approaches and Clinical Practice.

Platelets are one of the coagulation cells. When platelet activation occurs, many mediators are released and affect endothelial cells (ECs) and lead to endothelial dysfunction (ED). ED plays an important role in the pathogenesis of many diseases, including cardiovascular disease (CVD). Platelet are of important factors in ED. The release of mediators by platelets causes the stimulation of inflammatory pathways, oxidative stress, and apoptosis, which ultimately result in ED.On the other hand, platelet activation in CVD patients can be associated with a bad prognosis. Platelet activation can inc…

Platelet Activation-Induced In Situ Trapping Metastatic Tumor Cells Strategy for Post-Surgery Tumor Recurrence Immunochemotherapy.

The reasons for the high recurrence and metastasis after triple-negative breast cancer (TNBC) surgery are the potential presence of micrometastases and disseminated tumor cells in the circulation, as well as the immunosuppressive microenvironment caused by the surgery. To address these issues, this work proposed a platelets (PLTs) based hydrogel delivery system for immuno-chemotherapy, (IL-12+NPD-PLT)@dAlg. To utilize the targeting ability of PLTs toward tumor cells and maintain the activity of PLTs, doxorubicin-loaded nanoparticles (NPD) were attached on the surface of PTLs (NPD-PLT). Under t

Is autologous platelet activation the key step in ovarian therapy for fertility recovery and menopause reversal?

Platelets are a uniquely mammalian physiologic feature. As the only non-marine vertebrates to experience menopause, humans have a substantia … and its role in 'ovarian rejuvenation' remains unsettled. One critical aspect in this procedure is platelet activation, a commonly overlooked step in the cytokine release cascade considered essential for successful transition of undifferentiated ovarian stem cells to an oocyte lineage. Poor activation of platelets thus becomes an unforced error, potentially diminish …
