The CO2 and humidity senses of insects in a changing world.

The ability to detect carbon dioxide and water vapor is essential for insect survival. Insects possess specialized receptors and anatomical structures that confer remarkable sensitivity to these environmental factors. As both CO2 and humidity are prominent greenhouse gases, understanding how insects sense and respond to these variables is crucial given global fluctuations in climate. This Review consolidates current insights into the molecular and neural mechanisms underlying CO2 and humidity detection in insects, with a focus on their roles in respiration, water balance and osmoregulation. It

Computational model of flower pattern evolution predicts spontaneous emergence of boundary cell types across the petal epidermis.

Petal patterns play an important role in the reproductive success of flowering plants by attracting pollinators and protecting against environmental factors. Some transcription factors (TFs) involved in petal epidermal cell differentiation have been identified, but little is known about the upstream processes that pre-pattern the petal surface to establish their expression domains. Here, we have developed a computational model of petal pattern evolution to investigate this pre-patterning phase, selecting for gene regulatory network (GRNs) that divide the petal into proximal and distal domains

Targeted Gene Editing in Honey Bees Using Liposome-Based CRISPR-Cas9.

Genome editing technologies have revolutionized molecular biology, enabling precise manipulation of gene functions across diverse organisms. In this study, we introduce a novel liposome-mediated delivery system for CRISPR-Cas9 components targeting the Juvenile Hormone Acid Methyltransferase (JHAMT) gene in honey bees (Apis mellifera anatoliaca). This approach leverages drone sperm cells as vectors for CRISPR-Cas9 transfection, overcoming the technical challenges of embryo microinjection in honey bees, such as low survival rates and labor-intensive procedures. The study involved artificial inse

Extraction and Quantification of Total Anthocyanins, Determination of Anthocyanidin Core Structures, and Characterization of Specific Anthocyanins from Maize.

Anthocyanins are flavonoid pigments that accumulate in fruits and flowers that serve as attractants for pollinators and seed-dispersing organisms. Anthocyanins exhibit diverse chemical structures, characterized both by different anthocyanidin core structures and numerous chemical modifications of the anthocyanidin core. Here, we describe a protocol for the extraction and quantification of total anthocyanins, as well as for the characterization of anthocyanidin core structures and specific anthocyanins, using a spectrophotometer, high-performance liquid chromatography (HPLC), and ultra-high-per

Sex Allocation and Reproductive Success in a Perennial Hermaphroditic Herb: Both Size and Timing Matter.

AbstractIn simultaneous hermaphrodites, resource availability and the temporal distribution of mates determine male and female fitness and optimal sex allocation. In insect-pollinated plants, we expect individuals to allocate more to their female function when they are large and more to their male function when other individuals have many ovules available to be fertilized. Here, we studied the dependence of sex allocation and male and female components of reproductive success on both the size and the timing of reproduction in the plant Pulsatilla alpina (Ranunculaceae), accounting for inbreedi

A wild bumble bee shows intraspecific differences in sensitivity to multiple pesticides.

Wild pollinator declines are increasingly linked to pesticide exposure, yet it is unclear how intraspecific differences contribute to observed variation in sensitivity, and the role gut microbes play in the sensitivity of wild bees is largely unexplored. Here, we investigate site-level differences in survival and microbiome structure of a wild bumble bee exposed to multiple pesticides, both individually and in combination. We collected wild Bombus vosnesenskii foragers (N = 175) from an alpine meadow, a valley lake shoreline and a suburban park and maintained them on a diet containing a herbic

Trends of Total Applied Pesticide Toxicity in German Agriculture.

Expressing temporal changes in the use of pesticides, based not only on amounts (masses) but also on their toxicity for different species groups, was proposed as a sensible approach for evaluating potential environmental risks. Here, we calculated the total applied toxicity (TAT) between 1995 and 2019 for Germany, mapped it, and compared it to the US TAT and other risk indicators. Results show that the German TAT for terrestrial vertebrates decreased over time by about 20%. The TAT increased by a factor of three for fishes, largely due to insecticides, by a factor of two for soil organisms, la

Detection and Concentration of Neonicotinoids and Other Pesticides in Honey from Honey Bee Colonies Located in Regions That Differ in Agricultural Practices: Implications for Human and Bee Health.

This is a preliminary study conducted to analyze the presence and concentration of pesticides in honey obtained from honey bee colonies located in two regions with managed ecosystems that differ in the intensity and technification of agricultural practices. Fourteen pesticides at variable concentrations were detected in 63% of the samples analyzed. The pesticides most frequently found at higher concentrations were insecticides (neonicotinoids, followed by organophosphates), herbicides, and fungicides. The number, frequency, and concentration of pesticides were higher in samples collected from

Liquid Chromatographic Determination of Trace Bioavailable Neonicotinoids in Soil with Dispersive Liquid-Liquid Microextraction and Its Application for Experimental Monitoring.

Dispersive liquid-liquid microextraction (DLLME) was applied to extract bioavailable neonicotinoids involved in the uptake from soil through roots to plants. To quantitatively extract bioavailable neonicotinoids with the proposed DLLME, 3.5 mL of dichloromethane (extractant)/acetonitrile (dispersive solvent) (6:1, v/v) was injected into 5 mL of aqueous soil extracts in which 1 g of sodium chloride was previously dissolved. The separated dichloromethane phase after sonication and centrifugation was evaporated, reconstituted with a mobile phase, and determined with high-performance liquid chroma

Arabidopsis leucine-rich repeat malectin receptor-like kinases regulate pollen-stigma interactions.

Flowering plants contain tightly controlled pollen-pistil interactions required for promoting intraspecific fertilization and preventing interspecific hybridizations. In Arabidopsis (Arabidopsis thaliana), several receptor kinases (RKs) are known to regulate the later stages of intraspecific pollen tube growth and ovular reception in the pistil, but less is known about RK regulation of the earlier stages. The Arabidopsis RECEPTOR-LIKE KINASE IN FLOWERS1 (RKF1)/RKF1-LIKE (RKFL) 1-3 cluster of 4 leucine-rich repeat malectin (LRR-MAL) RKs was previously found to function in the stigma to promote

Availability of Using Honeybees as Bioindicators of Pesticide Exposure in the Vicinity of Agricultural Environments in Taiwan.

While pollinating, honeybees are subject to exposure to a variety of pesticides; with their characteristics of certain foraging distances, they could serve as bioindicators of pesticide exposure in a neighborhood. We conducted a study to assess availability by collecting and analyzing bee samples from 15 apiaries located in East Taiwan and dust samples from the adjacent environment, and by finding relations between both samples. Seventeen pesticides were selected for the analysis using gas or liquid chromatography coupled with mass spectrometry, and eight (three insecticides, two herbicides, a

Behavior and gut bacteria of Partamona helleri under sublethal exposure to a bioinsecticide and a leaf fertilizer.

The exposure of bees to agrochemicals during foraging and feeding has been associated with their population decline. Sublethal exposure to agrochemicals can affect behavior and the microbiota. Gut microbiota is associated with insect nutritional health, immunocompetence, and is essential for neutralizing the damage caused by pathogens and xenobiotics. Research on the effect of the bioinsecticides and fertilizers on the microbiota of bees remains neglected. In this study, we assessed the sublethal effect of both bioinsecticide spinosad and the fertilizer copper sulfate (CuSO4) on the behavior a

QSAR modeling in ecotoxicological risk assessment: application to the prediction of acute contact toxicity of pesticides on bees (Apis mellifera L.).

Despite their indisputable importance around the world, the pesticides can be dangerous for a range of species of ecological importance such as honeybees (Apis mellifera L.). Thus, a particular attention should be paid to their protection, not only for their ecological importance by contributing to the maintenance of wild plant diversity, but also for their economic value as honey producers and crop-pollinating agents. For all these reasons, the environmental protection requires the resort of risk assessment of pesticides. The goal of this work was therefore to develop a validated QSAR model t

An ABC transporter, OsABCG26, is required for anther cuticle and pollen exine formation and pollen-pistil interactions in rice.

Wax, cutin and sporopollenin are essential components for the formation of the anther cuticle and the pollen exine, respectively. Their lipid precursors are synthesized by secretory tapetal cells and transported to the anther and microspore surface for deposition. However, the molecular mechanisms involved in the formation of the anther cuticle and pollen exine are poorly understood in rice. Here, we characterized a rice male sterile mutant osabcg26. Molecular cloning and sequence analysis revealed a point mutation in the gene encoding an ATP binding cassette transporter G26 (OsABCG26). OsABCG

Neonicotinoids target distinct nicotinic acetylcholine receptors and neurons, leading to differential risks to bumblebees.

There is growing concern over the risk to bee populations from neonicotinoid insecticides and the long-term consequences of reduced numbers of insect pollinators to essential ecosystem services and food security. Our knowledge of the risk of neonicotinoids to bees is based on studies of imidacloprid and thiamethoxam and these findings are extrapolated to clothianidin based on its higher potency at nicotinic acetylcholine receptors. This study addresses the specificity and consequences of all three neonicotinoids to determine their relative risk to bumblebees at field-relevant levels (2.5 ppb).

Landscape determinants of fine-scale genetic structure of a small rodent in a heterogeneous landscape (Hluhluwe-iMfolozi Park, South Africa).

Small mammals provide ecosystem services, acting, for example, as pollinators and seed dispersers. In addition, they are also disease reservoirs that can be detrimental to human health and they can also act as crop pests. Knowledge of their dispersal preferences is therefore useful for population management and landscape planning. Genetic data were used alongside landscape data to examine the influence of the landscape on the demographic connectedness of the Natal multimammate mouse (Mastomys natalensis) and to identify landscape characteristics that influence the genetic structure of this spe

Honeybees Produce Millimolar Concentrations of Non-Neuronal Acetylcholine for Breeding: Possible Adverse Effects of Neonicotinoids.

The worldwide use of neonicotinoid pesticides has caused concern on account of their involvement in the decline of bee populations, which are key pollinators in most ecosystems. Here we describe a role of non-neuronal acetylcholine (ACh) for breeding of Apis mellifera carnica and a so far unknown effect of neonicotinoids on non-target insects. Royal jelly or larval food are produced by the hypopharyngeal gland of nursing bees and contain unusually high ACh concentrations (4-8 mM). ACh is extremely well conserved in royal jelly or brood food because of the acidic pH of 4.0. This condition prote

Metatranscriptome Analysis of Fig Flowers Provides Insights into Potential Mechanisms for Mutualism Stability and Gall Induction.

A striking property of the mutualism between figs and their pollinating wasps is that wasps consistently oviposit in the inner flowers of the fig syconium, which develop into galls that house developing larvae. Wasps typically do not use the outer ring of flowers, which develop into seeds. To better understand differences between gall and seed flowers, we used a metatranscriptomic approach to analyze eukaryotic gene expression within fig flowers at the time of oviposition choice and early gall development. Consistent with the unbeatable seed hypothesis, we found significant differences in gene
