Chemical modification of squid axon K+ channel -SH groups with the organic mercurial compound p-hydroxymercuriphenylsulfonic acid (PHMPS).

In internally dialyzed voltage-clamped squid axons, intracellular or extracellular addition of the sulfhydryl group (-SH) specific reagent p-hydroxymercuriphenylsulfonic acid (PHMPS), causes major modifications in the magnitude and kinetic parameters of the delayed rectifier K+ current. PHMPS produces a dramatic slow-down of the macroscopic current activation kinetics with a simultaneous reduction in its amplitude. In addition, it causes a marked increase in the delay of the macroscopic current at various pre-pulse potentials (Cole-Moore shift). The main effect of PHMPS at the single channel l

In vitro virucidal activity of chemical compounds against porcine transmissible gastroenteritis virus and porcine respiratory coronavirus.

Porcine transmissible gastroenteritis virus (TGEV) and porcine respiratory coronavirus (PRCV) are enveloped, single-stranded RNA viruses belonging to the genus Alphacoronavirus in the family Coronaviridae. In the absence of effective treatments, these viruses may lead to significant economic losses in the swine industry. To date, there is no or limited information about the efficiency of disinfectants against TGEV and PRCV. Therefore, in this study, we investigated the virucidal activity of eleven chemical compounds against TGEV and PRCV using a quantitative suspension test method based on the

Fluorescent recognition of potassium and calcium ions using functionalised CdSe / ZnS quantum dots.

Schiff base receptor 1a has been synthesised and attached to the surface of preformed CdSe/ZnS Quantum Dots (QDs) to form QD-conjugate 2a. While 1a was determined to be selective for Mg2+, 2a demonstrated selectivity for both K+ and Ca2+ when tested against a range of physiologically and environmentally relevant cations by changes in the fluorescence spectra. Thus, the nanoparticle surface functions as a scaffold for the organisation of receptors enabling semi-selective binding. The fluorescence response was shown to be linear between 15-50 microM for K+ and 2-35 microM for Ca2+. It was also d

Halogenated N-(1,3,4-oxadiazol-2-yl) benzamides are effective eradicators of methicillin-resistant Staphylococcus aureus biofilms.

Due to the ever-increasing threat of methicillin-resistant Staphylococcus aureus (MRSA), we have embarked on a campaign to discover novel antibacterial agents which are effective at eradicating both MRSA, as well as acting on pre-formed MRSA biofilms. Using the known scaffold of N-(1,3,4-oxadiazol-2-yl)benzamides, we performed a halogenation study which has led to the identification of HSGN-2241. This novel compound was found to effectively kill many multi-drug resistant Gram-positive clinical isolates in a bactericidal manner. The growth inhibition mechanism of HSGN-2241 was found to be via p

Effect of nutrient omissions on bread wheat and tef crops grain yield in Western Amhara, Ethiopia.

The decline in soil nutrients in Ethiopia, particularly in Western Amhara, is causing low crop productivity. Some researchers have argued that the application of K, S, Zn, and B in blended, individual, and complex forms affects crop yield. Identification of the prime yield-limiting nutrient is the key to solvesoil nutrient problems. A field experiment was conducted at Burie-Wemeberema, Debere Elias, Gozamen and Gonji Qolela districts of Western Amhara in the 2022 cropping season. A composite soil sample was taken at a depth of 0-20 cm to determine soil chemical properties. Bread wheat and tef

Co-application of potassium and thiourea for mitigating salinity stress in wheat seedlings.

Salt stress disrupts ionic homeostasis and induces oxidative damage, leading to reduced plant growth and development. Determining the lethal dose of NaCl is essential for developing effective mitigation strategies. Potassium and other antioxidant regulatory compounds often prove insufficient in addressing salt-induced toxicity, especially in wheat. This laboratory experiment aimed to determine NaCl concentration that reduces plant growth by 50% and to evaluate the effectiveness of different mitigation approaches involving potassium and thiourea supplementation at 0 mM, 10 mM, and 15 mM under t

GYY4137 and Sodium Hydrogen Sulfide Relaxations Are Inhibited by L-Cysteine and KV7 Channel Blockers in Rat Small Mesenteric Arteries.

Donors of H2S may be beneficial in treating cardiovascular diseases where the plasma levels of H2S are decreased. Therefore, we investigated the mechanisms involved in relaxation of small arteries induced by GYY4137 [(4-methoxyphenyl)-morpholin-4-yl-sulfanylidene-sulfido-λ5-phosphane;morpholin-4-ium], which is considered a slow-releasing H2S donor. Sulfides were measured by use of 5,5'-dithiobis-(2-nitro benzoic acid), and small rat mesenteric arteries with internal diameters of 200-250 µm were mounted in microvascular myographs for isometric tension recordings. GYY4137 produced similar low le

Photo-amygdalin: light-dependent control over hydrogen cyanide release and cytotoxicity.

Amygdalin is a natural glycosidic compound found in bitter almonds and apricot seeds. After enzymatic hydrolysis, amygdalin forms a cyanohydrin which spontaneously decays to release toxic hydrogen cyanide in a process called cyanogenesis. Due to this capacity to release cyanide, it has a long and controversial history of use as an anticancer therapeutic in alternative medicine. However, the inability to control amygdalin's cyanogenesis hinders its efficient medical use and renders it potentially dangerous. In this study we describe the design and development of 'photo-amygdalin', a compound wh

Comparative study on organic composts in blueberry: insights into soil physicochemical properties and heavy metal control.

Composts of edible fungal substrates (CEFS) and Chinese herbal residues (CCHR) are potential ecological organic fertilizers, which are sustainable for blueberry cultivation. But how the two composts affect the fertility and the migration or transformation of heavy metals (HMs) in blueberry soil is unknown. In this study, we conducted a field trial by comparing CEFS and CCHR to the special fruit organic fertilizer in the market (SFOF) and potassium sulfate compound fertilizers (PSCF) according to a three-year continuous fertilization experiment in blueberry. We aimed to reveal the effect of dif

Construction and application of soil-amending microbial inoculant for straw decomposition.

The current body of research on using composite microbial inoculants in soil to promote straw decomposition, potassium solubility, phosphorus solubility, and crop growth is relatively limited. In this study, we developed a composite microbial inoculant, designated HC-2, through the enrichment of the field with two strains of Bacillus. The agent displays multifunctionality and promotes rot prevention, solubilization of potassium and phosphorus, growth stimulation, and disease suppression. To further investigate its effects on soil fertility and microbial communities, field experiments were cond

Beta-KTx14.3, a scorpion toxin, blocks the human potassium channel KCNQ1.

Potassium channels play a key role in regulating many physiological processes, thus, alterations in their proper functioning can lead to the development of several diseases. Hence, the search for compounds capable of regulating the activity of these channels constitutes an intense field of investigation. Potassium scorpion toxins are grouped into six subfamilies (α, β, γ, κ, δ, and λ). However, experimental structures and functional analyses of the long chain β-KTx subfamily are lacking. In this study, we recombinantly produced the toxins TcoKIK and beta-KTx14.3 present in the venom of Tityus

Ethanol has thyrotropin-like activity in cultured porcine thyroid follicles.

We describe TSH-like activity of ethanol for thyroid hormone formation in the physiological culture system. Porcine thyroid follicles were preincubated with 0-100 mM (0-0.58%) ethanol in the presence of 0-1280 microU/ml bovine TSH for 24 h; these follicles were then incubated with the mixture of Na125I and NaI to measure iodide uptake, iodine organification, and de novo thyroid hormone formation. Ethanol stimulated iodide uptake in a dose-response manner in TSH-free medium. Ethanol augmented the effect of TSH on iodide uptake, iodide organification, and thyroid hormone formation in the presenc

Optimizing Therapies in Heart Failure: The Role of Potassium Binders.

Heart failure (HF) is a worrisome cardiac pandemic with a negative prognostic impact on the overall survival of individuals. International guidelines recommend up-titration of standardized therapies in order to reduce symptoms, hospitalization rates, and cardiac death. Hyperkalemia (HK) has been identified in 3-18% of HF patients from randomized controlled trials and over 25% of HF patients in the "real world" setting. Pharmacological treatments and/or cardio-renal syndrome, as well as chronic kidney disease may be responsible for HK in HF patients. These conditions can prevent the upgrade of

Potassium currents inhibition by gambierol analogs prevents human T lymphocyte activation.

Gambierol is a marine polycyclic ether toxin, produced along with ciguatoxin congeners by the dinoflagellate Gambierdiscus toxicus. We have recently reported that two truncated skeletal analogs of gambierol comprising the EFGH- and BCDEFGH-rings of the parent compound showed similar potency to gambierol on voltage-gated potassium channels (Kv) inhibition in neurons. Gambierol and its truncated analogs share the main crucial elements for biological activity, which are the C28=C29 double bond within the H-ring and the unsaturated side chain. Since Kv channels are critical for the regulation of c

A novel potassium channel blocking toxin from the scorpion Pandinus imperator: A 1H NMR analysis using a nano-NMR probe.

The three-dimensional solution structure of a novel peptide, Pi 1, purified from the venom of the scorpion Pandinus imperator and specific for potassium channels was determined by homonuclear proton NMR methods at 500 MHz from nanomole amounts of compound. P. imperator toxin is a voltage-dependent potassium channel specific peptide capable of blocking the shaker B K+ channels expressed in Sf9 cells in culture (Spodoptera frugiperda cell line no. 9) and displacing labeled noxiustoxin from rat brain synaptosomal membranes. The toxin has only 35 amino acid residues but is stabilized by four disul

Luzindole and ramelteon share a melatonin receptor-independent blocking effect on voltage-gated potassium KV4.2 channels.

Melatonin is a pineal hormone that regulates circadian rhythms primarily through melatonin receptors. We recently demonstrated that melatonin inhibits the voltage-gated potassium KV4.2 channel (IC50 = 479 μM) through a melatonin receptor-independent mechanism. However, it remains unclear whether this inhibitory effect is specific to melatonin or shared by structurally related compounds. In the present study, we examined the effects of melatonin-related compounds on KV4.2 channels using human embryonic kidney 293 cells stably expressing KV4.2 channels. Whole-cell patch-clamp recordings showed t

From Retigabine to Azetukalner: Reviving Voltage-Gated Potassium Channels for Epilepsy Therapy.

Heterotetrameric KV7.2/3 potassium channels were retrospectively identified as the target of the analgesic flupirtine and the antiepileptic retigabine (ezogabine). Clinical utility of these agents ended after decades or 6 years, respectively, before their full scope was explored. Market withdrawals in 2017 and 2018 hampered research in other medical fields and left a gap for researchers using these compounds off-label in experimental indications and for patients with KV7.2/3 malfunctions. Failure of these drugs might be regarded as a reason to abandon this class of compounds, due to the notion
