A highly sensitive and simple assay for the detection of circulating autoantibodies against full-length bullous pemphigoid antigen 180.

Bullous pemphigoid antigen 180 (BP180) is the target of autoantibodies in various subepidermal blistering diseases. The most common one is bullous pemphigoid (BP). The pathological importance of anti-BP180 antibodies has been demonstrated in a passive transfer mouse model. However, sensitive assays for routinely detecting circulating antibodies directed against both intra- and extracellular domains of BP180 are only available in specialized laboratories. In addition, most current assays use prokaryotic recombinant fragments of BP180 that lack conformation-dependent epitopes. A simple and very

In vivo detection of antisense HIV-1 transcripts in untreated and ART-treated individuals.

Natural antisense transcripts (AST) are expressed in eukaryotes, prokaryotes, and viruses and can possess regulatory functions at the transcriptional and/or post-transcriptional levels. In vitro studies have shown that HIV-1 AST promote viral latency through epigenetic silencing of the proviral 5' long terminal repeat. However, expression of AST in vivo has not been convincingly demonstrated. Here, we used single RNA template amplification and sequencing to demonstrate expression of AST in unstimulated PBMC collected from people with HIV-1 (PWH). Our results show that expression levels of AST

Molecular characterisation of Toxoplasma gondii Mre11 reveals unique structural features and potential as a therapeutic target.

The Mre11 enzyme plays a central role in essential cellular processes such as DNA repair and meiosis, where its endonuclease and exonuclease activities are critical for maintaining genomic stability. Previous studies have linked Mre11 to cell cycle regulation and cancer progression; however, knowledge about DNA processing mechanisms in medically relevant protozoan parasites remains limited. Toxoplasma gondii harbours several essential genes, involved in the DNA damage response, including mre11. The aim of this study was to characterize T. gondii Mre11 (TgMre11) at the molecular level as a pote

Carboxysomes: The next frontier in biotechnology and sustainable solutions.

Some bacteria possess microcompartments that function as protein-based organelles. Bacterial microcompartments (BMCs) sequester enzymes to optimize metabolic reactions. Several BMCs have been characterized to date, including carboxysomes and metabolosomes. Genomic analysis has identified novel BMCs and their loci, often including genes for signature enzymes critical to their function, but further characterization is needed to confirm their roles. Among the various BMCs, carboxysomes, which are found in cyanobacteria and some chemoautotrophic bacteria, and are most extensively investigated. The

A secreted fungal subtilase interferes with rice immunity via degradation of SUPPRESSOR OF G2 ALLELE OF skp1.

Serine protease subtilase, found widely in both eukaryotes and prokaryotes, participates in various biological processes. However, how fungal subtilase regulates plant immunity is a major concern. Here, we identified a secreted fungal subtilase, UvPr1a, from the rice false smut (RFS) fungus Ustilaginoidea virens. We characterized UvPr1a as a virulence effector localized to the plant cytoplasm that inhibits plant cell death induced by Bax. Heterologous expression of UvPr1a in rice (Oryza sativa) enhanced plant susceptibility to rice pathogens. UvPr1a interacted with the important rice protein S

The algal selenoproteomes.

BACKGROUND: Selenium is an essential trace element, and selenocysteine (Sec, U) is its predominant form in vivo. Proteins that contain Sec are selenoproteins, whose special structural features include not only the TGA codon encoding Sec but also the SECIS element in mRNA and the conservation of the Sec-flanking region. These unique features have led to the development of a series of bioinformatics methods to predict and research selenoprotein genes. There have been some studies and reports on the evolution and distribution of selenoprotein genes in prokaryotes and multicellular eukaryotes, but

The immunoreactivity of the anti-p21Ras single-chain fragment variant KGH-R1 and its predicted binding sites to p21Ras.

Aim: Previously, we constructed a novel anti-p21Ras single-chain antibody fragment, KGH-R1-single-chain fragment variant (ScFv). However, the immunoreactivity of this antibody toward p21Ras is still unclear. Materials & methods: ELISAs, immunohistochemistry, western blotting and immunofluorescence were used to identify the immunoreactivity of KGH-R1-ScFv toward p21Ras. An in silico approach was used to determine the protein structures of KGH-R1-ScFv and p21Ras and then to predict the site involved in the binding of KGH-R1-ScFv to p21Ras. Results: KGH-R1-ScFv had a specific immune reaction with

A preliminary study on the application of PspA as a carrier for group A meningococcal polysaccharide.

This study aimed to explore the feasibility of pneumococcal surface protein A (PspA) as a carrier protein. Three recombinant pneumococcal surface proteins from three different clades were expressed by the prokaryotic expression system and conjugated to group A meningococcal polysaccharide (GAMP) to generate three polysaccharide-protein conjugates. The conjugates, unconjugated proteins, GAMP, and GAMP-TT vaccine bulk (used as positive control) were immunized into mice, and their immune effects were assessed by the methods of enzyme-linked immunosorbent assay (ELISA), flow cytometry (FCM), and s

It is not all about regeneration: Planarians striking power to stand starvation.

All living forms, prokaryotes as eukaryotes, have some means of adaptation to food scarcity, which extends the survival chances under extreme environmental conditions. Nowadays we know that dietary interventions, including fasting, extends lifespan of many organisms and can also protect against age-related diseases including in humans. Therefore, the capacity of adapting to periods of food scarcity may have evolved billions of years ago not only to allow immediate organismal survival but also to be able to extend organismal lifespan or at least to lead to a healthier remaining lifespan. Planar

The Vibrio Cholerae Type VI Secretion System: Evaluating its Role in the Human Disease Cholera.

Vibrio cholerae, the marine bacterium responsible for the diarrheal disease cholera, utilizes a multitude of virulence factors to cause disease. The importance of two of these factors, the toxin co-regulated pilus (TCP) and cholera toxin (CT), has been well documented for pandemic O1 and epidemic O139 serogroups. In contrast, endemic non-O1 and non-O139 serogroups can cause localized outbreaks of cholera-like illness, often in the absence of TCP and CT. One virulence mechanism used by these strains is the type VI secretion system (T6SS) to export toxins across the cell envelope and confer toxi

Evaluation of the recombinant Heat shock protein B (HspB) of Coxiella burnetii as a potential antigen for immunodiagnostic of Q fever in goats.

Coxiella burnetii is an intracellular bacterium that causes a worldwide zoonosis, the Q fever. Currently, to diagnose the infection in ruminants, whole cell antigens-based ELISAs are used. In this study a heat shock protein, the HspB, was evaluated for its ability to be recognized by the goat immune system and its capacity to sign a stage of infection. The htpB gene of C. burnetii was cloned and sequenced. A high identity (>90%) was observed among the htpB genes of four ruminant strains tested. A recombinant protein was expressed in a prokaryotic expression system. The rHspB protein was used t

Recombinant bivalent vaccine against foot-and-mouth disease virus serotype O/A infection in guinea pig.

In this study, two DNA fragments encoding amino acid (141-160)-(21-140)-(141-160) of the VP1 of FMDV (foot-and-mouth disease virus) serotype O and (138-160)-(21-40)-(138-160) of the serotype A FMDV were chemically synthesized. These two tandem-repeat fragments were ligated and transfected into prokaryotic expression vector pTrcHis A to construct pTH-O-A. The other vector called pTH-O-scIgG-A was constructed similarly only that the two tandem-repeat DNA fragments were linked by the bovine-IgG heavy chain coding sequence. Guinea pigs immunized with the two bivalent vaccines pTH-O-A and pTH-O-scI

Cobalt complexes in biology and medicine: enzymatic functions, pharmacological applications, and health challenges.

Cobalt is an essential trace element in biochemistry that plays a crucial role in the structure and function of several important biomolecules. In this review, vitamin B12 is discussed as one of the best-known examples in this area. Various forms of this vitamin, including methylcobalamin and adenosylcobalamin, play a crucial role in metabolic reactions in mammals and prokaryotes. It also discusses cobalt-containing enzymes that are essential for various biological processes. These enzymes are B12-dependent enzymes, which are well studied, and cobalt-containing enzymes, which are less well kno

Molecular Equilibrist: The Small Heat Shock Protein IbpA from Mycoplasma.

Small heat shock proteins (sHSPs) serve as "first aid" stress-response proteins in both eukaryotes and prokaryotes. Their holdase activity enables binding to partially denatured proteins, maintaining them in a folding-competent state under stress. The sHSP IbpA from the mycoplasma Acholeplasma laidlawii is a unique member of its family, combining the functions of two Escherichia coli sHSPs that typically act in tandem. In this study, we demonstrate for the first time that IbpA forms distinct supramolecular structures under contrasting temperature stresses in crowded environments without any ar

[Expression of recombinant fibronectin FNIII8-10 in Pichia pastoris and fermentation process optimization].

The 8-10 repeat of fibronectin (FN) type III domain (FNIII8-10) contains multiple integrin-binding sites and serves as the core module mediating cell adhesion. Currently, recombinant FNIII8-10 is expressed only in prokaryotic microorganisms. This study aims to establish a Pichia pastoris expression system for the high-level production of recombinant FNIII8-10. Although the recombinant FNIII8-10 was successfully expressed in Pichia pastoris, the secretion efficiency was low. Through rational signal peptides design and fermentation optimization, secretion efficiency was significantly improved. S

Unraveling the role of distinct cytoskeletal motility structures in Mycoplasma pneumoniae relatives.

Mycoplasma represents a unique genus of prokaryotic bacteria distinguished by the absence of a cell wall, a characteristic that sets it apart from other bacteria. Within the Mollicutes class, phylogenetic analysis reveals three distinct categories: Spiroplasma, Mycoplasma and Acholeplasma. Mycoplasmas within Pneumoniae are recognized for their capacity to induce a range of diseases in both humans and animals, frequently impacting respiratory and reproductive health. The representative strains in Pneumoniae group, particularly the M. pneumoniae clusters, have garnered significant attention due

Immunoinformatics design and experimental expression of a multi-epitope vaccine simultaneously targeting AAV2 and HAdV-F41 against acute hepatitis of unknown etiology.

The recent global outbreak of acute hepatitis of unknown etiology (AHUE) in children has raised significant health concerns due to the severity of infections, some of which require liver transplants and can lead to fatalities. Emerging evidence suggests that AHUE is caused by a co-infection involving AAV2 and HAdV-F41. Through immunoinformatics, we identified optimal T-cell and B-cell epitopes from AAV2's VP1 and AAP, as well as from HAdV-F41's long fiber, short fiber, and hexon proteins. To enhance specific immune responses, we incorporated the pan DR-binding epitope (PADRE) and Mycobacterium

Structure and genome editing of type I-B CRISPR-Cas.

Type I CRISPR-Cas systems employ multi-subunit effector Cascade and helicase-nuclease Cas3 to target and degrade foreign nucleic acids, representing the most abundant RNA-guided adaptive immune systems in prokaryotes. Their ability to cause long fragment deletions have led to increasing interests in eukaryotic genome editing. While the Cascade structures of all other six type I systems have been determined, the structure of the most evolutionarily conserved type I-B Cascade is still missing. Here, we present two cryo-EM structures of the Synechocystis sp. PCC 6714 (Syn) type I-B Cascade, revea
