Inhibition of the enzyme purine nucleoside phosphorylase (PNP) reduces refractoriness to transfused platelets in a dog model.

To better define the role of T-cells in alloimmunization, we induced purine nucleoside phosphorylase (PNP) deficiency in a dog platelet transfusion model. Short-term administration of eight different drug schedules using several combinations of the PNP inhibitor 8-aminoguanosine and/or deoxyguanosine did not induce significant toxicity in four treated animals as demonstrated by blood chemistries, cell counts, and autologous platelet recovery and survival measurements. However, continuous long-term daily administration of these agents produced significant renal and/or hepatic toxicity leading t

Effect of zinc or copper deficiency on erythrocyte purine nucleoside phosphorylase and pyruvate kinase activity in the rat.

The effect of deficiencies of zinc or copper in the rat on the activity of purine nucleoside phosphorylase and pyruvate kinase in erythrocytes, liver and muscle was investigated. The data showed no effect of either zinc or copper deficiencies on the activity of these enzymes in the tissues tested. It is concluded that these enzymes are not useful markers for either zinc or copper deficiency.

Lesch-Nyhan syndrome and its pathogenesis: purine concentrations in plasma and urine with metabolite profiles in CSF.

Purine metabolism in the Lesch-Nyhan syndrome has been re-examined in 10 patients. Hypoxanthine and xanthine concentrations in plasma and CSF and urinary excretion have been studied, on and off allopurinol treatment, using high performance liquid chromatographic methods. Accumulation of the substrate, hypoxanthine, of the missing hypoxanthine guanine phosphoribosyltransferase (HPRT) enzyme, is more marked in urine and in CSF than in plasma. The greater increase in CSF is consistent with the most metabolically active tissue, brain, showing the most marked functional changes. The function of HPR

Relation between coronary artery stenosis and myocardial purine metabolism, histology and regional function in humans.

In 54 patients undergoing elective or emergency aortocoronary bypass grafting, angiographic and electrocardiographic changes were studied. Five patients with unstable angina and five patients with evolving myocardial infarction were included. High energy phosphate metabolism and the histologic appearance of the myocardium were analyzed in transmural biopsy specimens acquired at the time of surgery. In patients without anterior infarction on the electrocardiogram, severe stenosis of the left anterior descending coronary artery resulted in a reduction of anterior wall motion that was associated

Excess purine degradation in exercising muscles of patients with glycogen storage disease types V and VII.

To investigate purine catabolism in exercising muscles of patients with muscle glycogen storage disease, we performed ischemic forearm exercise tests and quantitated metabolites appearing in cubital venous blood. Two patients with glycogen storage disease type V and three with glycogen storage disease type VII participated in this study. Basal lactate concentrations lowered in every patient with glycogen storage disease type V or type VII. Two patients with glycogen storage disease type VII, who had markedly elevated concentrations of serum uric acid (14.3 and 11.9 mg/dl, respectively), showed

Augmentation by theophylline of [3H]purine release from vascular adrenergic nerves: evidence for presynaptic autoinhibition.

The effect of theophylline (a P1-purinoceptor blocker) on the tritium release evoked by high KCl (50 mM) and l-epinephrine (3 microM) was evaluated using [3H]adenosine-prelabeled pulmonary artery of the rabbit. It has previously been shown that KCl elicits purine release from adrenergic nerve terminals whereas epinephrine releases it from postsynaptic sites in this artery. The KCl-induced [3H]purine efflux was significantly enhanced by theophylline (10-100 microM), whereas the efflux induced by epinephrine (3 microM) was not enhanced but reduced by this drug at 100 microM. In contrast to theop

In vitro screening of potential anti-cancer chemicals: effect of purine pyrimidine analogues on seed germination.

Germinating Cicer arietinum seeds were used to simulate embryonic tissue for evaluating toxicity of purine and pyrimidine analogues. 6-Mercaptopurine stimulated germination, whereas 8-azaadenine, 8-azaguanine, 2,6-diaminopurine, 6-amino, 2-hydroxypurine and 5-fluorouracil inhibited germination. 6-Methyluracil, 5-aminouracil and methylxanthines, viz. caffeine, theophylline, theobromine and uric acid exhibited little inhibitory effects. RNA synthesis ensued immediately after water imbibition and reached maximum at 12 h of germination. 2,6-Diaminopurine, 8-azaadenine, 8-azaguanine and 5-fluoroura

Urinary excretion of orotic acid, orotidine and other pyrimidines in a patient with purine nucleoside phosphorylase deficiency.

Urinary orotidine and orotic acid have been determined in a patient with purine nucleoside phosphorylase (PNP) deficiency under various dietary therapeutic conditions. For this purpose a new procedure for the analysis of both compounds has been developed, consisting of prefractionation with Dowex 1X8, followed by two HPLC steps on a micro Bondapak NH2 and a micro Bondapak C18 column. With this method normal as well as slightly elevated excretions of orotic acid have been found in our patient. No evidence was obtained for inhibition of OPRT by purine (deoxy)nucleosides as a cause of pyrimidine

Regulation of purine nucleotide synthesis. Effects of inosine on normal and hypoxantine-guanine phosphoribosyltransferase-deficient fibroblasts.

Incubation of normal and hypoxanthine-guanine phosphoribosyltransferase-deficient (mutant) human fibroblasts with inosine results in increased intracellular concentration of 5-phosphoribosyl 1-pyrophosphate (PP-ribose-P). The magnitude of this increase is dependent on the concentration of the nucleoside and results from donation of the ribose moiety of inosine to the ribosyl phosphate moiety of PP-ribose-P through ribose phosphate intermediates. During incubation, rates of purine nucleotide synthesis de novo, estimated by incorporation of (14C) formate into formylglycinamide ribotide, are dimi

Purine and citrate metabolism in urolithiasis and bladder cancer: An integrated genetic, gene-expression, and targeted metabolite analysis.

BACKGROUND: Metabolic reprogramming is a hallmark of bladder cancer (BC), but the contribution of inherited variation in purine and citrate metabolism remains incompletely understood. Because urolithiasis shares selected metabolic and epidemiological features with BC, we compared both disorders to identify shared and disease-specific molecular patterns. METHODS: We analysed polymorphisms in ACLY, XDH, PNP, HPRT1, and GART together with targeted gene expression in peripheral blood-derived RNA and urinary citrate, hypoxanthine, and glycine measured by 1H NMR in a Polish case-control cohort. Cardiac injury and purine metabolic remodeling in Carassius auratus under environmentally relevant carbonate alkalinity exposure.

With the intensification of global warming and human activities, inland freshwaters are increasingly salinized and alkalized, often with carbonate alkalinity predominating. Such conditions disrupt acid-base and ion homeostasis in freshwater fish and may induce systemic energy metabolic reprogramming. However, the effects of carbonate alkalinity stress on the heart, an organ with high energy demands, remain poorly understood. Therefore, Carassius auratus (C. auratus), which exhibits relatively high tolerance to carbonate alkalinity, was selected as the experimental species in the present study.

Next generation smart functionalized nanocomposites for ultrasensitive and real time detection of purine derived metabolic biomarkers.

The monitoring of metabolic biomarkers is currently undergoing a radical change from traditional centralized laboratory standards to decentralized point-of-care testing, particularly in the management of metabolic disorders, such as gout, and renal dysfunction. This review explores next-generation functionalized nanocomposites such as metal-organic frameworks, two-dimensional (2D) transition-metal carbides, nitrides, and carbonitrides (MXenes) and Single-Atom Nanozymes (SANs) which function as sensitive elements in sensing platforms for biomarkers detection and serve as robust, non-enzymatic m

Unveiling crosstalk regulations within the polyamine pathway and between polyamine and purine pathways in Aedes aegypti females.

We previously demonstrated that ornithine decarboxylase (ODC) deficiency critically impairs nitrogen metabolism and survival in Aedes aegypti. To further examine the role of the polyamine pathway in Ae. aegypti nitrogen metabolism, we evaluated the expression of three additional genes encoding proteins involved in the biosynthetic pathway: S-adenosylmethionine decarboxylase, spermidine synthase (SdS), spermine synthase (SmS), and seven genes encoding proteins involved in the catabolic pathway in fat body, midgut and Malpighian tubules by qPCR. Distinct transcriptional profiles were observed in

Temporal metabolic reprogramming in DSS-induced colitis identifies purine metabolism and trigonelline as novel therapeutic targets.

BACKGROUND: Inflammatory bowel disease (IBD) is a multifactorial disorder characterized by aberrant immune activation and metabolic dysregulation. Despite significant advances in understanding immune mechanisms, the temporal dynamics of metabolic alterations during intestinal inflammation and their therapeutic implications remain poorly defined. METHODS: To investigate metabolic reprogramming during colitis progression, we conducted time-resolved metabolomic profiling of the colon, mesenteric lymph nodes (MLNs), and serum in a dextran sulfate sodium (DSS)-induced murine colitis model at days

Exploring the Most Effective Strategy for Purine Metabolite Quantification in Veterinary Medicine Using LC-MS/MS.

Measuring purine metabolites in the serum and urine may help in the early diagnosis of urolith formation, the correction of allopurinol therapeutic dosages, and the evaluation of diet-related alterations. To properly monitor these endogenous compounds and assess their physiological concentration ranges in biological fluids, highly specific and accurate analytical approaches are required. Colorimetric assays are generally used for this purpose, although their cross-reactivity could lead to incorrect determinations. Given the importance of selectivity in detecting endogenous compounds, this stud
