Molecular phylogeny of the filaria genus Onchocerca with special emphasis on Afrotropical human and bovine parasites.

Filarial parasites of the genus Onchocerca are found in a broad spectrum of ungulate hosts. One species, O. volvulus, is a human parasite that can cause severe disease (onchocerciasis or 'river blindness'). The phylogenetic relationships and the bionomics of many of the nearly 30 known species remain dubious. Here, the phylogeny of 11 species representing most major lineages of the genus is investigated by analysing DNA sequences from three mitochondrial genes (ND5, 12S and 16S rRNA) and portions of the intergenic spacer of the nuclear 5s rRNA. Special emphasis is given to a clade containing a

Expansion of CD4 T cells expressing a highly restricted TCR structure specific for a single parasite epitope correlates with high pathology in murine schistosomiasis.

The hepatic immunopathology in schistosomiasis mansoni is mediated by CD4 T cells specific for egg antigens and varies considerably among mouse strains. Previous studies in high pathology C3H mice suggested that a strong T cell response was due to the recognition of an immunodominant epitope within the major egg antigen Sm-p40 (Sm-p40(234-246)). Using a panel of T cell hybridomas, we have now examined the egg antigen-specific TCR repertoire in two high pathology strains, C3H and CBA. We found that nearly half of the hybridomas responded to the Sm-p40(234-246 )epitope and, of these, nearly all

A cathepsin F of adult Clonorchis sinensis and its phylogenetic conservation in trematodes.

A novel 28 kDa cysteine protease (Cs28CF) secreted by the hepatobiliary trematode, Clonorchis sinensis was identified. The protease was purified from the excretory-secretory products (ESP) of the adult worm using DEAE-ion exchange and Arginine-Sepharose 4B chromatography. It showed a high activity between pH 6.5 and 7.5 in a dithiothreitol (DTT)-dependent manner. Inhibitors specific to cysteine proteases down-regulated the activity. Addition of Cs28CF to monkey cholangiocyte cultures resulted in approximately 95% cell death after 7 days. The full-length cDNA (1078 bp) encoded a single peptide

A Tousled-like kinase (TLK) homolog required for male germline stem cell homeostasis in the planarian Schmidtea mediterranea.

Tousled-like kinases (TLKs) are serine/threonine kinases that are conserved across metazoan life and involved in DNA stability and cell division. Mammalian TLKs are most highly expressed in the testes, but their role in spermatogenesis has remained elusive because mutations result in embryonic lethality. Here, we identify two TLK homologs in the planarian Schmidtea mediterranea (Smed-TLK1a and Smed-TLK1b) and reveal a testis-specific requirement for Smed-TLK1a. Preferential expression of Smed-TLK1a was detected in testes of S. mediterranea hermaphrodites by whole-mount in situ hybridization, a

'First record of Hsiungia pekingensis (Nematoda: Strongylidae) in North America: Morphological and molecular identification of a rare equine strongyle.

Equids are infected by a diversity of gastrointestinal nematode parasites, including 64 species of equine strongyle nematodes from19 genera. Despite numerous surveys of horse strongyles worldwide, certain geographic regions and rare species remain understudied. In 1964, a new species of equine strongyle, Cylicocyclus pekingensis, was described from a donkey in China. Subsequently, this species was recorded in horses from Kazakhstan and reclassified as Hsiungia pekingensis (K'ung and Yang, 1964), the only species in this genus. Since then, H. pekingensis has not been reported elsewhere, with li

Scleroductus (Platyhelminthes: Monogenea) infecting Rhamdia spp. (Siluriformes: Heptapteridae) in Mexico, and their phylogenetic position within Gyrodactylidea.

Gyrodactylidae Cobbold, 1864 is a monophyletic family of hyperviviparous species, with 36 genera and approximately 700 species. Here, we focused on one of these genera - namely Scleroductus Jara & Cone, 1989 - characterizing its morphological variation systematically and testing its phylogenetic position in relation to other genera in the family. We collected and describe two new species of monogenean parasites of Scleroductus infecting Neotropical freshwater catfishes in Mexico. Scleroductus veracruzano n. sp. infects Guatemalan chulín, Rhamdia guatemalensis in Veracruz, while Scleroductus oa

Morphological and Molecular Studies of Tetracotyle-Type Metacercariae of the Genus Cotylurus Szidat, 1928 (Trematoda) from the Gravel Snail Lithoglyphus naticoides (Gastropoda) and Host Sex Dependent Differences in Infection Rate.

The Ponto-Caspian invader, the gravel snail Lithoglyphus naticoides (C. Pfeiffer, 1828), is infected with a diverse community of digenetic trematodes in its colonized range and most often serves as first intermediate host. We have performed the parasitological examination of L. naticoides snails sampled in Kaunas water reservoir (Lithuania) and found yet unknown strigeid metacercariae of the tetracotyle type using these snails as second intermediate host. In this study, we report data on morphology and molecular analysis based on two markers, the partial 28S rDNA gene and the ITS2 region of th

Occurrence and host associations of Dollfustrema bagarii (Digenea: Bucephalidae) in channel catfish cage culture in Northern Vietnam.

A survey was conducted across 11 provinces in Northern Vietnam to investigate the distribution of Dollfustrema bagarii metacercariae - a causative agent of significant losses in cultured channel catfish (Ictalurus punctatus) - and to identify its first intermediate host. A total of 337 fish specimens were collected from 103 farms. Metacercariae of D. bagarii were detected in 72 farms (69.9% of farms), and 44.51% of the sampled fish were positive. The prevalence in lowland provinces was 3.14 times higher than that in mountainous regions (p < 0.001), while no significant difference was observed

PHYLOGENETIC POSITION OF NEOGOGATEA (DIPLOSTOMOIDEA; CYATHOCOTYLIDAE).

Neogogatea Chandler and Rausch, 1947, is a small genus of cyathocotylid digeneans parasitic in the intestines of birds of prey. The genus contains only 3 species: 2 from the Nearctic and 1 from the Palearctic. No sequence data from mature, properly identified representatives of Neogogatea were available until now. In the present study, we collected mature adult Neogogatea kentuckiensis (Cable, 1935) Hoffman and Dunbar, 1963, from osprey in Georgia, Florida, and Delaware, and sequenced fragments of the nuclear large ribosomal subunit (28S) gene and cytochrome c oxidase subunit I (COI) mtDNA gen

Paronatrema davidbowiei n. sp. (Trematoda: Syncoeliidae), a new parasite of the pelagic thresher (Alopias pelagicus) and its phylogenetic relationships within the suborder Hemiurata Skrjabin & Guschanskaja, 1954.

A new species of hemiurid trematode found on the gills and in the aorta of the pelagic thresher Alopias pelagicus from the eastern Pacific, off Costa Rica, is described based on an integrative taxonomic approach that includes the use of light and scanning electron microscopy, and 28S rDNA sequencing. Phylogenetic analysis was also performed to explore, for the first time, the relationships of a member of the subfamily Otiotrematinae within the suborder Hemiurata. Paronatrema davidbowiei n. sp. can be distinguished from the congeners by having tegumental spines on the dorsal surface of the fore

New record and phylogenetic assessment of Apopharynx bolodes (Braun, 1902) (Digenea: Psilostomidae), a parasite of Eurasian Coot Fulica atra Linnaeus, 1758 (Aves: Rallidae).

Phylogenetic studies of aberrant species are of considerable scientific interest because their taxonomic rank in traditional systems based on morphological characters is not infrequently overestimated. Apopharynx bolodes (Braun, ) is one of the few psilostomid digeneans devoid of the pharynx. This is considered a sufficient basis for assigning it and similar species to the subfamily Apopharynginae. We found A. bolodes in Fulica atra Linnaeus, 1758 from Belarus, described it morphologically, and genotyped it by the 28S rRNA gene and the ITS2 region. It is the first molecular data on A. bolodes

Ocular Dirofilariasis Case in Thailand Confirmed by Molecular Analysis to Be Caused by Dirofilaria immitis.

A rare ocular dirofilariasis case along with the clinical characteristics, treatment, and outcome is reported. A whitish roundworm (10.6 cm long and 0.5 mm width) emerged from the pterygium, a triangular tissue growth on the cornea of the eye, of a male patient. The worm had a rounded anterior part, mouth without lips, smooth cuticular surface, and short rounded posterior tail with spicules: these features suggested that it was a male Dirofilaria sp. Molecular identification confirmed that the worm belonged to Dirofilaria immitis. This is the first molecular confirmation that D. immitis is a c

Case Report: Periorbital Filariasis Caused by Brugia malayi.

Brugia malayi is a lymphatic nematode that accounts for approximately 10% of lymphatic filariasis cases worldwide. It is endemic in several countries in South and Southeast Asia. In Thailand, B. malayi is endemic in the southern region. The extralymphatic presentation of B. malayi is rare. Here, we report the case of a woman residing in the central region of Thailand who presented with an erythematous periorbital nodule at the left medial canthal area caused by lymphatic filaria. A viable sexually mature filarial adult was removed from the lesion. The nematode species was identified as B. mala

Central European parasitic flatworms of the family Renicolidae Dollfus, 1939 (Trematoda: Plagiorchiida): molecular and comparative morphological analysis rejects the synonymization of Renicola pinguis complex suggested by Odening.

The Renicolidae are digenean parasites of piscivorous and molluscivorous birds. Although they exhibit few morphological autapomorphies and are highly variable, the numerous suggested re-classifications within the family have never been supported by any molecular analyses. We address the possible synonymization of species within the Renicola pinguis complex suggested previously by Odening. We provide and analyse sequences of two nuclear (ITS2, 28S rDNA) and two mitochondrial (CO1, ND1) DNA loci of central European species of the Renicolidae, namely Renicola lari, Renicola pinguis and Renicola s

Steinernema pwaniensis n. sp., a new entomopathogenic nematode (Nematoda: Steinernematidae) from Tanzania.

A new species of entomopathogenic nematode, Steinernema pwaniensis n. sp., belonging to the glaseri group, is described from Tanzania. The infective juveniles of S. pwaniensis n. sp. are characterized by a body length of 978 μm (808-1131), distance from anterior end to excretory pore of 86 μm (80-95) and a tail length of 87 μm (75-95). The ratios a, c and E% of S. pwaniensis n. sp. are substantially lower than those of all other African 'glaseri' group members. The first-generation males of S. pwaniensis n. sp. can be distinguished by a large spicule length of 92 μm (80-97) and by the absence

Tissue Degeneration following Loss of Schistosoma mansoni cbp1 Is Associated with Increased Stem Cell Proliferation and Parasite Death In Vivo.

Schistosomiasis is second only to malaria in terms of the global impact among diseases caused by parasites. A striking feature of schistosomes are their ability to thrive in their hosts for decades. We have previously demonstrated that stem cells, called neoblasts, promote homeostatic tissue maintenance in adult schistosomes and suggested these cells likely contribute to parasite longevity. Whether these schistosome neoblasts have functions independent of homeostatic tissue maintenance, for example in processes such as tissue regeneration following injury, remains unexplored. Here we character

Development of Recombinase Polymerase Amplification and CRISPR-Cas12a-Enhanced Isothermal Amplification Assays for Strongyloides stercoralis DNA Detection: A Pilot Study.

Soil-transmitted helminth (STH) infections are prevalent worldwide, but the true burden of strongyloidiasis is unclear due to lack of sensitive and field-friendly diagnostic tools. Diagnosis is often based on serological assays that are typically not point-of-care (POC). Although polymerase chain reaction (PCR) tests are sensitive and specific, the need for expensive equipment and highly skilled personnel limits their use in resource limited areas. Isothermal amplification assays are largely instrument-free, making them simpler to implement without loss of either sensitivity or specificity. We

First morphological and molecular characterization of Eustrongylides excisus larvae (L4) (Nematoda: Dioctophymatidae), with description of induced oxidative-inflammatory changes and tissue injury in European perch (Perca fluviatilis) from the Volga River (Astrakhan, Russia).

Eustrongylides spp. are helminths with a multiple-host life cycle that can infect aquatic oligochaetes, various freshwater fish, fish-eating birds, and accidental mammalian hosts, including humans. This work aimed to morphologically and molecularly characterize the larval stages of Eustrongylides spp. isolated from European perch, and to outline the oxidative/nitrosative stress and histoarchitecture changes induced by active larval migration or tissue-encapsulation. Eighty Perca fluviatilis were collected from the Volga River (Astrakhan, Russia). Thirty-seven were tentatively grouped as infect

A New Species of the Acanthocephalan Genus Filisoma (Cavisomidae) from Perciform Fishes in Rio de Janeiro, Brasil.

BACKGROUND: Twelve species of Filisoma Van Cleave, 1928 are recognized parasitizing tropical and subtropical fish. Four of these species were described from kyphosid fish and it has been suggested that a co-speciation may have occurred among species of Kyphosus Lacepède, 1801 and Filisoma, which could provide valuable information about the evolution history of this host-parasite system. PURPOSE: During a survey of the helminth fauna of Kyphosus sectatrix (Linnaeus, 1758) and Kyphosus incisor (Cuvier, 1831) (Kyphosidae Jordan, 1887) off Rio de Janeiro coast, a new species of Filisoma was found
