Deciphering the interactions of phytochemicals with alcohol dehydrogenase 1 (ADH1) in Saccharomyces cerevisiae: a molecular simulations study.

The present study has investigated the interactions of the phytochemicals present in sugarcane bagasse (SCB) with alcohol dehydrogenase (ADH1) enzyme in Saccharomyces cerevisiae using molecular simulations. ADH1 catalyzes the reduction of acetaldehyde to ethanol. The metabolic hindrance induced by phytochemicals can adversely affect kinetics and yield of ethanol fermentation. The phytochemicals in SCB biomass were detected using LC-ESI-MS/QTOF analysis after dilute acid/alkali pretreatment and extraction using methanol and acetonitrile. Molecular docking simulation revealed high binding affini…

Antioxidant and anti-aging properties of a Zn-complex in Saccharomyces cerevisiae: A non-canonical protective mechanism.

Zinc is essential for cellular homeostasis, but its role in oxidative stress resistance and aging remains incompletely understood. We evaluated the antioxidant and anti-aging effects of the zinc(II) coordination compound [Zn2(μ-Cl)2(HPClNOL)2](ClO4)2, (HPClNOL: 1-(bis-pyridin-2-ylmethylamino)-3-chloropropan-2-ol), in Saccharomyces cerevisiae. The crystal structure of this Zn-complex revealed a distorted octahedral geometry around both Zn(II) centers. The equatorial plane comprised one tertiary amine, one alcohol, in the protonated form, and two chloro, which bridge the two Zn(II) centers, resu…

The dynamic pool of Rec8-cohesin is crucial for meiotic recombination and transcription regulation in the yeast Saccharomyces cerevisiae.

Cohesin is a ring-shaped protein complex that mediates sister-chromatid cohesion (SCC) to ensure accurate chromosome segregation during mitosis and meiosis. In Saccharomyces cerevisiae, cohesin consists of four core subunits-Smc1, Smc3, Scc1/Mcd1, and Scc3. During meiosis, the mitotic α-kleisin Scc1/Mcd1 is replaced by the meiosis-specific α-kleisin Rec8. Rec8-containing cohesin is essential for multiple meiotic processes, including chromosome morphogenesis, homologous recombination, axis and synaptonemal complex formation, SCC, and transcriptional regulation. While stable association of Rec8-…

Evaluation of Saccharomyces cerevisiae fermented Moutai distiller's grains as an alternative ingredient for growing-finishing pigs.

Three experiments were conducted to evaluate the effects of incorporating Saccharomyces cerevisiae fermented Moutai distiller's grains (FMDG) into diets for growing-finishing pigs across three body weight (BW) phases: 30 to 50 kg (Exp. 1), 50 to 75 kg (Exp. 2), and 90 to 130 kg (Exp. 3). The experimental diets were corn-soybean meal based with 0%, 5% or 10% FMDG. Metabolomic analysis revealed that fermentation enriched FMDG with beneficial compounds, including carbohydrates, quercetin, and tripeptides, compared to unfermented Moutai dried distiller's grains. Results form the animal experiments…

Unraveling Time-Resolved Transcriptional and Metabolic Shifts in the Mixed Fermentation of Saccharomyces cerevisiae and Hanseniaspora uvarum.

Wine fermentation and flavor formation are shaped by complex biochemical reactions driven by a variety of microorganisms. Non-Saccharomyces yeasts, such as Hanseniaspora uvarum (HU), are often used in mixed fermentation with Saccharomyces cerevisiae (SC) to enhance wine aroma. However, the lack of systematic knowledge regarding transcriptional responses and metabolic behaviors during fermentation has hindered the rational control of the mixed fermentation processes. To address this, we investigated transcriptional dynamics and metabolic behavior throughout the entire fermentation process, with…

Identification and engineering of a cellobiose transporter KmStl1p to enhance cellobiose utilization in Kluyveromyces marxianus and Saccharomyces cerevisiae.

The yeast Kluyveromyces marxianus, a yeast known for its ability to ferment ethanol at high temperatures, can utilize various sugars includi … aused growth defects. To confirm its function, KmSTL1 was co-expressed with a β-glucosidase gene in Saccharomyces cerevisiae EBY.VW1000, which only uptake maltose. This engineered strain grew in cellobiose medium, indicating that KmSTL1 encodes a cellobiose transporter. Expression of GFP-fused KmStl1p in K. marxianus revealed that KmStl1p localized on cell memb …

Identification of a crucial INO2 allele for enhancing ethanol resistance in an industrial fermentation strain of Saccharomyces cerevisiae.

Ethanol toxicity is a major challenge for Saccharomyces cerevisiae during fermentation, affecting its growth and influencing the process. This study investigated the molecular mechanisms of ethanol tolerance using transcriptomics analysis of three S. cerevisiae strains chosen due to their differing levels of resistance to ethanol described in a previous work, which linked them to differences in their membrane compositions. Transcriptomic analysis revealed distinct responses in membrane lipid synthesis genes, particularly those involved in ergosterol biosynthesis, in ethanol-tolerant strains ca…

Agent-based modelling of the early stages of actin polymerisation required to drive endocytosis in Saccharomyces cerevisiae.

Endocytosis is critical. Its complexity means that many aspects remain poorly understood. We have developed an agent-based model covering key components of actin filament generation in endocytosis in Saccharomyces cerevisiae. The model incorporates realistic values for rates, affinities, concentrations, and mobilities, and reproduces essential features of endocytosis, from the arrival of WASp/Las17 and its inhibitor Sla1 at the membrane up to the burst of actin polymerisation. The model yields relative rates and affinities for interactions that cannot be measured experimentally, and places lim…

Feeding Saccharomyces cerevisiae fermentation products lessens the severity of a viral-bacterial coinfection in preweaned calves.

We have previously reported that supplementation with Saccharomyces cerevisiae fermentation products (SCFP) ameliorates clinical signs and lung pathology following experimental bovine respiratory syncytial virus (BRSV) infection in preweaned dairy calves. The objectives of this study were to determine the effect of SCFP supplementation on the metabolic and endocrine responses, and disease outcome of a viral-bacterial coinfection in preweaned calves. Twenty-seven, 1- to 2-d-old Holstein-Angus cross calves were enrolled in the study; one SCFP calf was removed from the trial during the pre-challe…

Vaginal Isolates of Candida glabrata Are Uniquely Susceptible to Ionophoric Killer Toxins Produced by Saccharomyces cerevisiae.

Compared to other species of Candida yeasts, the growth of Candida glabrata is inhibited by many different strains of Saccharomyces killer yeasts. The ionophoric K1 and K2 killer toxins are broadly inhibitory to all clinical isolates of C. glabrata from patients with recurrent vulvovaginal candidiasis, despite high levels of resistance to clinically relevant antifungal therapeutics.

Capric acid secreted by Saccharomyces boulardii influences the susceptibility of Candida albicans to fluconazole and amphotericin B.

The effect of capric acid, secreted by the probiotic yeasts Saccharomyces boulardii, was evaluated on the activities of fluconazole (FLC) and amphotericin B (AMB) against pathogenic Candida albicans fungus. The findings indicated that capric acid may be a promising additive for use in combination with FLC. A FLC-capric acid combination led to reduced efflux activity of multidrug resistance (MDR) transporter Cdr1p by causing it to relocalize from the plasma membrane (PM) to the interior of the cell. The above effect occurred due to inhibitory effect of FLC-capric acid combination of ergosterol …

Use of non-Saccharomyces yeasts in cider fermentation: Importance of the nutrients addition to obtain an efficient fermentation.

The isolation of autochthonous yeast species presents a good strategy to select new microorganisms for developing an adequate inoculum to carry out fermentations and generate representative products of the cider production zone. However, non-Saccharomyces yeasts have been considered to have low capacity to carry out a complete fermentation as Saccharomyces cerevisiae. In this work, five autochthonous yeasts from a cider fermentation process were isolated and identified as Saccharomyces cerevisiae, Kluyveromyces marxianus, Pichia membranaefaciens, P. kluyveri and Zygosaccharomyces rouxii. A ser…

Impact of serine and serine synthesis genes on H2S release in Saccharomyces cerevisiae during wine fermentation.

Excessive hydrogen sulfide (H2S) during fermentation causes undesirable sensory properties in wine. In yeast, serine functions as a precursor in the biosynthesis of S-containing compounds, which facilitates H2S consumption. To investigate the effect of serine on H2S release and the underlying mechanism, extracellular and intracellular serine levels were separately increased under fermentation conditions. The results show that, although the abundance of extracellular serine was ineffective in decreasing H2S levels, increased levels of intracellular serine levels from SER1 and SER2 overexpressio

Aspirin Causes Lipid Accumulation and Damage to Cell Membrane by Regulating DCI1/OLE1 in Saccharomyces cerevisiae.

Aspirin is one of the most commonly used nonsteroidal anti-inflammatory drugs. Various potential pharmacological effects of aspirin, such as anticancer, antibacterial activity, and prolonging life expectancy have been discovered. However, the mechanism of aspirin is not fully elucidated. Herein, the effects of aspirin on fatty acid metabolism in yeast cell model Saccharomyces cerevisiae were studied. The results showed that aspirin can induce lipid accumulation and reduce the unsaturated fat index in cells. The assessment of cell membrane integrity demonstrated that aspirin caused damage to th…
