Fluorescence Lifetime Imaging of pH along the Secretory Pathway.

Many cellular processes are dependent on correct pH levels, and this is especially important for the secretory pathway. Defects in pH homeostasis in distinct organelles cause a wide range of diseases, including disorders of glycosylation and lysosomal storage diseases. Ratiometric imaging of the pH-sensitive mutant of green fluorescent protein, pHLuorin, has allowed for targeted pH measurements in various organelles, but the required sequential image acquisition is intrinsically slow and therefore the temporal resolution is unsuitable to follow the rapid transit of cargo between organelles. Molecular Imaging in Pancreatic Cancer: Current Applications and Future Perspectives.

Pancreatic cancer ranks among the most lethal malignancies, characterized by a five-year survival rate of approximately 10%. This dismal prognosis is largely attributable to diagnoses occurring at advanced stages and the inherent limitations of conventional imaging modalities in detecting early lesions, identifying metastases, and assessing tumor heterogeneity. Consequently, there is a critical need for non-invasive imaging techniques capable of visualizing pancreatic cancer lesions to enable accurate diagnosis, risk assessment, and the development of personalized treatment strategies. Association of Excessive Daytime Sleepiness With Long-Term Incident Dementia: The Role of β-Amyloid and White Matter Injury.

INTRODUCTION: Excessive daytime sleepiness (EDS) is common in community-dwelling older adults, but its association with long-term incident dementia remains unclear. The roles of Alzheimer's disease (AD) pathology and vascular-related white matter injury in this association were merely studied. METHODS: We included 617 dementia-free participants from the Shanghai Aging Study with up to 10 years of follow-up. At baseline, EDS was assessed with the Epworth Sleepiness Scale (ESS) and plasma phosphorylated tau (p-tau)217 was measured using a single-molecule immune-array assay. Cognitive decline wa

Multidimensional roles of acetylcholine receptors in neurobiology and their therapeutic significance.

Dysfunction of cholinergic signaling has emerged as a convergent mechanism across neurological and neuromuscular disorders, linking disturbances in synaptic, immune, and metabolic regulation. This review summarizes recent structural, functional, and imaging advances, demonstrating that nicotinic and muscarinic acetylcholine receptors play central roles in neural plasticity, neurotransmission, and neuroimmune homeostasis. Studies in structural biology and molecular pharmacology have clarified receptor architectures and conformational transitions, which enhance our understanding of subtype-speci

Imaging intracellular Ca²⁺ signals in striatal astrocytes from adult mice using genetically-encoded calcium indicators.

Astrocytes display spontaneous intracellular Ca(2+) concentration fluctuations ([Ca(2+)]i) and in several settings respond to neuronal excitation with enhanced [Ca(2+)]i signals. It has been proposed that astrocytes in turn regulate neurons and blood vessels through calcium-dependent mechanisms, such as the release of signaling molecules. However, [Ca(2+)]i imaging in entire astrocytes has only recently become feasible with genetically encoded calcium indicators (GECIs) such as the GCaMP series. The use of GECIs in astrocytes now provides opportunities to study astrocyte [Ca(2+)]i signals in d

In Situ Self-Assembled Nanoparticles for Cascade-Instructed Photodynamic Therapy and Real-Time Treatment Feedback Imaging.

Although the cancer treatment efficacy has significantly improved, delayed and isolated evaluation methods are difficult to prevent excessive or insufficient treatment. Therefore, the single molecule for simultaneous therapy and feedback has become a promising tool. Herein, we develop an in situ self-assembled probe (HPO-CYFF-TPP) for cascade-instructed therapy and feedback imaging. Under the dephosphorylation of alkaline phosphatase, HPO-CYFF-TPP converts to HO-CYFF-TPP and self-assembles into nanoparticles (NPs) with quenched fluorescent (FL) and opened photoacoustic (PA) signal. After laser

Absorption dipole effects on MINFLUX single molecule localization.

Single-molecule fluorescence localization with minimum photon flux imaging (MINFLUX) can achieve localization precisions in the small nanometer range or better under suitable conditions. Potentially adverse conditions, such as a fixed fluorescence dipole or optical aberrations, that could cause systematic localization errors, have received little attention up to now. Here, we study these effects in simulation. We find that biases occur for fluorophores with a fixed absorption dipole tilted out of the imaging plane. These become larger (up to about 25% of the diameter of the circle spanned by t

Three-color single-molecule localization microscopy in chromatin.

Super-resolution microscopy has revolutionized our ability to visualize structures below the diffraction limit of conventional optical microscopy and is particularly useful for investigating complex biological targets like chromatin. Chromatin exhibits a hierarchical organization with structural compartments and domains at different length scales, from nanometers to micrometers. Single molecule localization microscopy (SMLM) methods, such as STORM, are essential for studying chromatin at the supra-nucleosome level due to their ability to target epigenetic marks that determine chromatin organiz

Direct observation of cargo transfer from HDL particles to the plasma membrane.

BACKGROUND AND AIMS: Exchange of cholesterol between high-density lipoprotein (HDL) particles and cells is a key process for maintaining cellular cholesterol homeostasis. Recently, we have shown that amphiphilic cargo derived from HDL can be transferred directly to lipid bilayers. Here we pursued this work using a fluorescence-based method to directly follow cargo transfer from HDL particles to the cell membrane. METHODS: HDL was either immobilized on surfaces or added directly to cells, while transfer of fluorescent cargo was visualized via fluorescence imaging. RESULTS: In Chinese hamster

Application of dual-view oblique plane microscopy to small-molecule compound screening across 3D glioblastoma stem cell spheroids with single-cell resolution.

Recent advances and increasing adoption of 3-dimensional (3D) model systems, such as tumour spheroids and organoids, attempt to more faithfully recapitulate the pathophysiology and heterogeneity observed in patients' tumours, with the goal of reducing high attrition rates observed in late stage drug development. While established high content imaging systems provide the spatial resolution and throughput necessary to place 3D models at the earliest stages of drug discovery, they yield limited information on disease heterogeneity or drug response at the single cell level in 3D. Improvements in s

Common T-Cell-Receptor Motifs and Features in Patients with Cytomegalovirus (CMV)-Seronegative End-Stage Renal Disease Receiving a Peptide Vaccination against CMV.

After solid-organ transplantation, reactivation of the cytomegalovirus (CMV) is often observed in seronegative patients and associated with a high risk of disease and mortality. CMV-specific T cells can prevent CMV reactivation. In a phase 1 trial, CMV-seronegative patients with end-stage renal disease listed for kidney transplantation were subjected to CMV phosphoprotein 65 (CMVpp65) peptide vaccination and further investigated for T-cell responses. To this end, CMV-specific CD8+ T cells were characterized by bulk T-cell-receptor (TCR) repertoire sequencing and combined single-cell RNA and TC

Rational Design of Sequentially Activated Dual-Locked Near-Infrared Fluorescent Probe for Simultaneous Imaging of Cerebral Amyloid-β Plaques and ClO- in Alzheimer's Disease.

Alzheimer's disease (AD) is a complex neurodegenerative disorder mediated by multiple pathological factors, including amyloid-β (Aβ) plaque deposition and oxidative stress. Herein, we report a new class of sequentially activated dual-lock-responsive fluorescent probes, D-BDY and T-BDY, which were designed by integrating pyrrole-based electron-donating units and molecular rotors into the BODIPY scaffold. This strategic design enabled photoinduced electron transfer (PET) and twisted intermolecular charge transfer (TICT) modulated fluorescence responses to both Aβ1-42 aggregates and hypochlorite

The ganglioside GM1a functions as a coreceptor/attachment factor for dengue virus during infection.

Dengue virus (DENV) is a flavivirus causing an estimated 390 million infections per year around the world. Despite the immense global health and economic impact of this virus, its true receptor(s) for internalization into live cells has not yet been identified, and no successful antivirals or treatments have been isolated to this date. This study aims to improve our understanding of virus entry routes by exploring the sialic acid-based cell surface molecule GM1a and its role in DENV infection. We studied the interaction of the virus with GM1a using fluorescence correlation spectroscopy, fluore

Research Progress in Diffusion Spectrum Imaging.

Studies have demonstrated that many regions in the human brain include multidirectional fiber tracts, in which the diffusion of water molecules within image voxels does not follow a Gaussian distribution. Therefore, the conventional diffusion tensor imaging (DTI) that hypothesizes a single fiber orientation within a voxel is intrinsically incapable of revealing the complex microstructures of brain tissues. Diffusion spectrum imaging (DSI) employs a pulse sequence with different b-values along multiple gradient directions to sample the diffusion information of water molecules in the entire q-sp

Deciphering the complex three-way interaction between the non-integrin laminin receptor, galectin-3 and Neisseria meningitidis.

The non-integrin laminin receptor (LAMR1/RPSA) and galectin-3 (Gal-3) are multi-functional host molecules with roles in diverse pathological processes, particularly of infectious or oncogenic origins. Using bimolecular fluorescence complementation and confocal imaging, we demonstrate that the two proteins homo- and heterodimerize, and that each isotype forms a distinct cell surface population. We present evidence that the 37 kDa form of LAMR1 (37LRP) is the precursor of the previously described 67 kDa laminin receptor (67LR), whereas the heterodimer represents an entity that is distinct from t

Sensitivity evaluation of an optical microfiber featuring interfaces with various gold nanoparticle morphologies: Application to the GFAP detection.

Glial fibrillary acidic protein (GFAP) is a specific blood biomarker for various neurological diseases, including traumatic brain injury (TBI). In this study, we present a cost-effective, portable, and label-free biosensing method for the sensitive and rapid detection of GFAP in body fluids. As the sensitivity of current optical fiber sensors is insufficient to detect the ultralow concentration of GFAP in early body fluids, interfaces of gold nanoparticles with various morphologies were employed to improve the sensitivity of sensor. The optical microfiber sensor with gold nanostar interface de

Characterization of a DNA Aptamer for Ovarian Cancer Clinical Tissue Recognition and in Vivo Imaging.

BACKGROUNDS/AIMS: Ovarian cancer is the most lethal gynaecologic malignancy and is difficult to detect early. The inefficient early diagnosis of ovarian cancer is the main contributor to its high mortality rate. Aptamers, as chemical antibodies, are single-stranded DNA or RNA oligonucleotides that target cells or molecules with high affinity. METHODS: Binding ability of R13 was measured by flow cytometry analysis. Stability of R13 was tested in blood serum of an ovarian cancer patient. Internalization of R13 was verified by confocal microscope imaging. 80 cases ovarian cancer tissues, 10 case

High-resolution imaging for the detection and characterisation of circulating tumour cells from patients with oesophageal, hepatocellular, thyroid and ovarian cancers.

Interest has increased in the potential role of circulating tumour cells in cancer management. Most cell-based studies have been designed to determine the number of circulating tumour cells in a given volume of blood. Ability to understand the biology of the cancer cells would increase the clinical potential. The purpose of this study was to develop and validate a novel, widely applicable method for detection and characterisation of circulating tumour cells. Cells were imaged with an ImageStream(X) imaging flow cytometer which allows detection of expression of multiple biomarkers on each cell

Organization of Myosin H in the Apical Complex of Toxoplasma Gondii Revealed by 3D Single-Molecule Super-Resolution Microscopy.

Toxoplasma gondii is a single-celled eukaryotic parasite with prolific invasion capability. The parasite uses an apical complex comprised of proteinaceous structures and secretory organelles to efficiently enter host cells. As a result, the apical complex remains a vital structure of interest, with many studies dedicated to understanding its protein organization. One such protein is the motor Myosin H (MyoH), which is indispensable for parasite motility and host cell invasion. Given the small size of the complex, roughly a diffraction-limited volume in the visible, high-resolution techniques a
