A gain-of-function mutation in the sodium channel gene Scn2a results in seizures and behavioral abnormalities.

The GAL879-881QQQ mutation in the cytoplasmic S4-S5 linker of domain 2 of the rat brain IIA sodium channel (Na(v)1.2) results in slowed inactivation and increased persistent current when expressed in Xenopus oocytes. The neuron-specific enolase promoter was used to direct in vivo expression of the mutated channel in transgenic mice. Three transgenic lines exhibited seizures, and line Q54 was characterized in detail. The seizures in these mice began at two months of age and were accompanied by behavioral arrest and stereotyped repetitive behaviors. Continuous electroencephalogram monitoring det

Mineralocorticoid hormone signaling regulates the 'epithelial sodium channel' in fibroblasts from human cornea.

We investigated the regulation of sodium absorption by steroid hormones in embryologically diverse cells from the human eye. A cell extract from human corneal fibroblasts was positive for both the epithelial sodium channel (ENaC) and the mineralocorticoid receptor (MCR) as 82- to 85-kD and 102-kD bands, respectively, by the Western blot technique. In fluorescent, confocal and electron microscopy, the MCR was revealed as a nucleocytoplasmic protein, whereas the ENaC was almost exclusively membrane bound; both appeared aligned along actin filaments of corneal keratocytes, and both were widely co

The sgk, an aldosterone-induced gene in mineralocorticoid target cells, regulates the epithelial sodium channel.

The sgk, an aldosterone-induced gene in mineralocorticoid target cells, regulates the epithelial sodium channel. Aldosterone increases sodium reabsorption in tight epithelia. The early phase of this stimulatory effect is thought to involve activation of apical sodium channels. To identify immediate-early genes that initiate this effect, we used a combination of polymerase chain reaction-based subtractive hybridization and differential display techniques. This review summarizes our recent findings. Aldosterone rapidly increases mRNA levels of a putative Ser/Thr kinase, sgk (or serum- and glucoc…

Differential distribution of the tetrodotoxin-sensitive rPN4/NaCh6/Scn8a sodium channel in the nervous system.

Voltage-gated sodium channels underlie the generation of action potentials in excitable cells. Various sodium channel isoforms have been cloned, functionally expressed and distinguished on the basis of their biophysical properties or differential sensitivity to tetrodotoxin (TTX). In the present study, we have investigated the immunolocalization of the TTX-sensitive sodium channel, rPN4/NaCh6/Scn8a, in discrete areas of the rat nervous system. Thus, in naïve animals, PN4 was abundantly expressed in brain, spinal cord, dorsal root ganglia (DRG) and peripheral nerve. The presence of PN4 at the n…

Modification of synaptic transmission and sodium channel inactivation by the insect-selective scorpion toxin LqhalphaIT.

The peptide LqhalphaIT is an alpha-scorpion toxin that shows significant selectivity for insect sodium channels over mammalian channels. We examined the symptoms of LqhalphaIT-induced paralysis and its neurophysiological correlates in the house fly (Musca domestica). Injection of LqhalphaIT into fly larvae produced hyperactivity characterized by continuous, irregular muscle twitching throughout the body. These symptoms were correlated with elevated excitability in motor units caused by two physiological effects of the toxin: 1) increased transmitter release and 2) repetitive action potentials …

Insulin increases sodium (Na+) channel density in A6 epithelia: implications for expression of hypertension.

Essential or primary hypertension is a multifactorial disease that is expressed as a result of complex interactions between genes and environmental influences. Several mutations in many different proteins are associated with expression of hypertension, including abnormalities in the epithelial sodium channel (ENaC) found in absorptive organs (i.e., distal colon, distal tubule of the nephron). Some of these mutations result in structural and/or functional alterations in ENaC-mediated Na+ entry in epithelia responsible for fluid and electrolyte balance and are associated with expression of hyper

Differential actions of pacific ciguatoxin-1 on sodium channel subtypes in mammalian sensory neurons.

Pacific ciguatoxin-1 (P-CTX-1), is a highly lipophilic cyclic polyether molecule originating from the marine dinoflagellate Gambierdiscus to … in peak current amplitude occurred in both channel types. The main actions of 5 nM P-CTX-1 on TTX-S sodium channels were a 13-mV hyperpolarizing shift in the voltage dependence of sodium channel activation and a 22-mV hyperpolarizing shift in steady-state inactivation (hinfinity). In addition, P-CTX-1 caused a rapid rise in the membrane leakage current in cells e …

Inactivation defects caused by myotonia-associated mutations in the sodium channel III-IV linker.

Missense mutations in the skeletal muscle Na+ channel alpha subunit occur in several heritable forms of myotonia and periodic paralysis. Distinct phenotypes arise from mutations at two sites within the III-IV cytoplasmic loop: myotonia without weakness due to substitutions at glycine 1306, and myotonia plus weakness caused by a mutation at threonine 1313. Heterologous expression in HEK cells showed that substitutions at either site disrupted inactivation, as reflected by slower inactivation rates, shifts in steady-state inactivation, and larger persistent Na+ currents. For T1313M, however, the

Distinct regulation of sodium channel types I, II and III following nerve transection.

We investigated mRNA expression of type I, II and III voltage-sensitive sodium channels in motoneurons following facial nerve transection by means of in situ hybridization histochemistry. Type I mRNA expression decreased markedly after nerve transection, while that of type III increased. Type II expression underwent no detectable change following nerve transection. These results suggest that type III sodium channels may be involved in regeneration and plasticity.

The mode of action of ethanol on batrachotoxinin-A benzoate binding to sodium channels in mouse brain cortex.

Since ethanol has local anesthetic activity its effect was examined in vitro on the scorpion toxin-enhanced specific binding of [3H]batrachotoxinin A 20-alpha-benzoate ([3H]BTX-B) to the gating complex in sodium channel preparations from mouse brain cortex by equilibrium and kinetic experiments. Ethanol inhibited the specific binding of [3H]BTX-B in a vesicular preparation with an IC50 value of 310 mM and a Hill number of 1.0. Ethanol increased the equilibrium dissociation constant of batrachotoxin in a concentration dependent manner without changing the maximal binding capacity and decreased

[Effect of toxins from the scorpion Buthus eupeus on the sodium channels of the membranes of nodes of Ranvier].

Effect of three toxins M3, M7 and M8 from Buthus eupeus poison on sodium channels of Ranvier node membrane of the frog was studied under potential fixation. Toxin M3 was found to produce no effect on sodium currents of Ranvier node membrane of the frog. Inactivation kinetics difference between normal and modified sodium channels was used in estimating a fraction of toxin-modified channels. The toxins were found to interact with the channels in the ratio 1 : 1, the dissociation constants for M7 and M8 equalling 1.7 . 10(-7) M and 0.9 . 10(-9) M respectively. Dependence of the effect of scorpion…

Framework for combined functional and computational assessment of variant pathogenicity in the sodium leak channel NALCN.

PURPOSE: The sodium leak channel nonselective (NALCN) is the pore-forming channel component of the NALCN channelosome. This protein complex generates a depolarizing sodium leak in various tissues and contributes to respiratory and circadian rhythms, locomotion, and sleep. De novo and inherited variants of NALCN can lead to severe developmental syndromes referred to as CLIFAHDD and IHPRF1, respectively. Although variants of uncertain significance (VUS) have been studied in heterologous expression systems before, there is no generally accepted framework for how to assess or predict variant patho

Differential Assembly of Native Epithelial Sodium Channel Complexes Across Mouse Epithelial Tissues.

KEY POINTS: The new mouse line, epithelial sodium channel γ - mVenus FLAG (ENaC γ -VF), expresses a γ subunit tagged with mVenus and 3xFLAG that retains wild-type physiological function at baseline. The tags allow fluorescence-based detection and affinity purification of native γ -containing epithelial sodium channel complexes under mild, nondenaturing conditions. γ -containing complexes are heterogeneous, including αβγ channels as well as complexes associated with channel biogenesis, trafficking, and regulation. BACKGROUND: The epithelial sodium channel (ENaC) governs sodium and fluid absorp

FGF12-Related Early-Onset Epileptic Encephalopathies: Therapeutic Response to Sodium Channel Blockers.

Developmental and epileptic encephalopathies (DEEs) comprise a clinically and genetically heterogeneous group of severe neurodevelopmental disorders, frequently caused by pathogenic variants in genes encoding neuronal ion channels or synaptic proteins. The fibroblast growth-factor 12 (FGF12) encodes a binding protein for voltage-gated sodium channels. Variants in FGF12 have recently been associated with autosomal dominant DEEs characterized by early-onset epilepsy and neurodevelopmental impairment. We report three patients with a duplication involving exons 1-4 of FGF12 on chromosome 3q28-q29 …

Decreasing Microtubule Detyrosination Improves Cardiac Mechanics and Sodium Channel Function in Arrhythmogenic Cardiomyopathy.

BACKGROUND: Alterations in microtubule dynamics have been shown to affect cardiomyocyte membrane stiffness and modulate ion channels, including the cardiac sodium channel. While conditions, such as heart failure and Duchenne muscular dystrophy, are associated with increased detyrosination of microtubules and reduced sodium current, a potential role for microtubule detyrosination in arrhythmogenic cardiomyopathy has not been explored. We here investigated the impact of microtubule detyrosination on membrane stiffness, cardiac sodium channel distribution, and function in mouse and human models o

Sodium channel blockers are associated with reduced dementia risk in late-onset unexplained epilepsy.

Late-onset unexplained epilepsy is a potential harbinger of dementia, likely driven by network hyperexcitability that facilitates amyloid-β release and tau propagation. Dampening this activity with antiseizure medications offers a potential disease modifying strategy, yet whether specific agents differentially alter this neurodegenerative trajectory remains unknown. Here, we emulated a target trial using global real-world federated data on patients with late-onset unexplained epilepsy to compare dementia risk across antiseizure monotherapies. Using data from over 75 million adults aged 55 year

Ventricular conduction is a marker for arrhythmic risk in SCN5A-E1784K overlap sodium channel disease.

AIMS: SCN5A-E1784K (c.5350G>A) is the most common variant associated with the long QT (LQTS) and Brugada syndromes (BrS). It can manifest variably as LQTS, BrS, and/or conduction disorders. This presents a challenge for risk stratification. We aimed to describe clinical and ECG characteristics and identify risk markers that associate with arrhythmic events. METHODS AND RESULTS: We undertook a retrospective observational multicentre study of a large cohort of 231 subjects with SCN5A-E1784K from Europe, USA, and Japan. Comprehensive demographic and clinical data, including initial presentation

Calmodulin acts as a selective switch in FGF12A-mediated regulation of the cardiac sodium channel Nav1.5.

The voltage-gated Na+ channel (Nav1.5) is responsible for the rapid depolarization of the cardiac action potential and is associated with the initiation of neuronal action potentials in the central nervous system. Nav1.5 channel gating is modulated by its C-terminal domain (CTD) and the multiple auxiliary proteins that are bound to it, including calmodulin (CaM) and intracellular fibroblast growth factors (iFGFs). A non-canonical CaM-binding site was recently discovered on the N-terminus of iFGF A-splice variants. To test whether FGF12A, CaM-bound or not, regulates Nav1.5 gating, we performed
