Diosgenone synthesis, anti-malarial activity and QSAR of analogues of this natural product.

Solanum nudum Dunal steroids have been reported as being antimalarial compounds; however, their concentration in plants is low, meaning that the species could be threatened by over-harvesting for this purpose. Swern oxidation was used for hemisynthesis of diosgenone (one of the most active steroidal sapogenin diosgenin compounds). Eighteen structural analogues were prepared; three of them were found to be more active than diosgenone (IC50 27.9 μM vs. 10.1 μM, 2.9 μM and 11.3 μM). The presence of a 4-en-3-one grouping in the A-ring of the compounds seems to be indispensable for antiplasmodial a…

Rwandan female genital modification: elongation of the Labia minora and the use of local botanical species.

The elongation of the labia minora is classified as a Type IV female genital mutilation by the World Health Organization. However, the term … cted with thirteen informants. Two botanicals applied during stretching sessions were identified as Solanum aculeastrum Dunal and Bidens pilosa L. Both have wide medicinal use and contain demonstrated beneficial bioactive compounds. We suggest that it is therefore more appropriate to describe Rwandan vaginal practices as female genital modification rather …

Hydroponic potato production on nutrients derived from anaerobically-processed potato plant residues.

Bioregenerative methods are being developed for recycling plant minerals from harvested inedible biomass as part of NASA's Advanced Life Support (ALS) research. Anaerobic processing produces secondary metabolites, a food source for yeast production, while providing a source of water soluble nutrients for plant growth. Since NH4-N is the nitrogen product, processing the effluent through a nitrification reactor was used to convert this to NO3-N, a more acceptable form for plants. Potato (Solanum tuberosum L.) cv. Norland plants were used to test the effects of anaerobically-produced effluent aft…

SlGH3.15, a member of the GH3 gene family, regulates lateral root development and gravitropism response by modulating auxin homeostasis in tomato.

Multiple Gretchen Hagen 3 (GH3) genes have been implicated in a range of processes in plant growth and development through their roles in maintaining hormonal homeostasis. However, there has only been limited study on the functions of GH3 genes in tomato (Solanum lycopersicum). In this work, we investigated the important function of SlGH3.15, a member of the GH3 gene family in tomato. Overexpression of SlGH3.15 led to severe dwarfism in both the above- and below-ground sections of the plant, accompanied by a substantial decrease in free IAA content and reduction in the expression of SlGH3.9, a…

Comparative assessment for hyperaccumulatory and phytoremediation capability of three wild weeds.

The composition and the organization of soil are changing rapidly by the diverged mankind activities, leading to the contamination of enviro … ckel, etc., by the wild weeds and that has been discussed here in detail in case of Cannabissativa, Solanum nigrum and Rorippa globosa. In general, weeds that have the intrinsic capacity to accumulate metals into their shoots and roots, have the ability to form phytochelates and formation of stable compound with ions. This behavior of accumulation along w …

A β-Mannanase Gene, MAN2, Coordinates Tomato Fruit and Seed Development and Salt Stress Tolerance.

Tomato (Solanum lycopersicum) β-mannanase is known as playing an important role in mannan degradation; however, its biological functions and transcriptional regulation remain poorly understood. Here, we characterized the roles of mannan-1,4-β-mannanase 2 (SlMAN2) in tomato development and salt stress tolerance. Results showed that SlMAN2 was preferentially expressed in seeds, stems, stamens, and stigmas. SlMAN2 overexpression significantly increased fruit and seed size, accelerated seed germination, and enhanced salt tolerance under 100 mM NaCl, whereas knockout mutants exhibited the opposite …

WRKY transcription factors in potato research: functional genetic manipulation for multi-stress resilience and crop improvement.

Potato (Solanum tuberosum L.) is a globally important staple crop whose productivity is increasingly threatened by concurrent abiotic and biotic stresses under climate change. Transcription factors of the WRKY family have emerged as central regulatory hubs coordinating stress-responsive multiple networks and specialized metabolism. This article highlights current advances in WRKY research in potato, with a particular focus on functional genetic manipulation approaches aimed at enhancing multi-stress resilience. By integrating targeted literature analysis and bibliometric mapping, we highlight …

Low Cadmium Accumulating Potato Varieties for Safe Food Production: Mechanisms, Genetic Targets, and Integrated Agronomic Strategies.

Cadmium (Cd) contamination of agricultural soils threatens food safety by promoting Cd accumulation in potato (Solanum tuberosum L.) tubers. Low-Cd-accumulating cultivars offer a promising mitigation strategy, although their effectiveness is strongly influenced by genotype × environmental interactions. This review synthesizes current evidence indicating that stable low-Cd phenotypes arise from coordinated regulation of soil Cd bioavailability, rhizosphere processes, root uptake, transporter-mediated translocation, and intracellular sequestration rather than cultivar genetics alone. Particular …

Arbuscular mycorrhizal colonization does not improve root hydraulic supply in tomato and pea.

Arbuscular mycorrhizal (AM) fungi are known to enhance plant drought tolerance, but the physiological mechanism behind this benefit remains unclear. One explanation is that AM colonization improves root hydraulic conductance (Kr), thereby facilitating more efficient water uptake under soil drying, though this mechanism remains highly debated. Here, we measured Kr in tomato (Solanum lycopersicum L.) and pea (Pisum sativum L.) with and without AM using a noninvasive rehydration technique under soil drying, and this was complemented with the evaporative flux method under hydrated conditions. AM c…

Respiratory burst oxidase homolog 1 plays a critical role in spermidine-induced thermotolerance by enhancing CO2 assimilation and cellular redox homeostasis in tomato.

Heat stress increasingly threatens global agriculture by disrupting photosynthesis and cellular redox homeostasis in plants. Spermidine (Spd), a stress-responsive polyamine, enhances thermotolerance by bolstering antioxidant capacity and stabilizing membranes; however, the molecular mechanisms underlying these effects remain incompletely characterized. Here, we demonstrate that Respiratory burst oxidase homolog 1 (RBOH1) is essential for Spd-induced thermotolerance in tomato (Solanum lycopersicum L.). Deficiency of RBOH1 compromised the protective effects of Spd, resulting in exacerbated membr…

Overexpression of S-nitrosoglutathione reductase alleviated iron-deficiency stress by regulating iron distribution and redox homeostasis.

Iron (Fe) is an essential micronutrient element for plant growth. The S-nitrosoglutathione reductase (GSNOR) gene's functions under Fe-deficiency conditions are not well understood. Here, GSNOR expression was induced by Fe deficiency in tomato (Solanum lycopersicum L.) leaves and roots, while its overexpression alleviated chlorosis under Fe-deficiency conditions. GSNOR overexpression positively regulated the Fe distribution from root to shoot, which might result from the transcriptional regulation of genes involved in Fe metabolism. Additionally, the overexpression of GSNOR maintained redox ho…

Botrytis cinerea manipulates the antagonistic effects between immune pathways to promote disease development in tomato.

Plants have evolved sophisticated mechanisms to sense and respond to pathogen attacks. Resistance against necrotrophic pathogens generally r … antagonizes the JA signaling pathway, thereby allowing the fungus to develop its disease in tomato (Solanum lycopersicum). SA-promoted disease development occurs through Nonexpressed Pathogen Related1. We also show that the JA signaling pathway required for tomato resistance against B. cinerea is mediated by the systemin elicitor. These data highlight a n …

Effects of plant-induced hypervitaminosis D on cutaneous structure, cell differentiation and cell proliferation in cattle.

Solanum glaucophyllum (Sg) (synonym S. malacoxylon) is a plant toxic to cattle due to its high levels of 1,25-dihydroxyvitamin D3 as glycoside derivatives. Sg causes a disease characterized by wasting and calcification of soft tissues. The effects of vitamin D are not only important in calcium homeostasis, but also in immune regulation, cell growth and cell differentiation. Skin samples in Sg-intoxicated and control heifers were studied histologically. Cellular differentiation and proliferation were analysed by immunohistochemical expression of cytokeratins, involucrin and proliferating cell n…

Regionally Divergent Relationships Between Microbial Communities and Environmental Gradients in Potato Production Systems.

Using soil microbiome data as indicators of soil health remains a challenging task in agriculture research. Microbiome data are complex and exhibit strong spatiotemporal variation and diverse responses to environmental factors. As a result, measurements of microbiota often are difficult to interpret and do not provide credible inferences about soil health status. In this study, we evaluated the distributions of soil microbial communities in three distinct potato growing regions of the Upper Midwest of the USA and compared their relationships with soil chemistry and management factors across re

Kojic acid attenuates tomato soft rot caused by Pectobacterium carotovorum subsp. carotovorum through quorum quenching.

Pectobacterium carotovorum subsp. carotovorum (Pcc) is the causal agent of bacterial soft rot disease, which triggers rapid decay of infected tomato fruit. Kojic acid, primarily a fermentation metabolite of Aspergillus oryzae, exhibits biocontrol activity against postharvest soft rot of tomato. In this work, we investigated the interference of kojic acid with quorum sensing (QS) in Pcc and elucidated the relevant mechanisms to provide novel theoretical insights into the management of tomato postharvest soft rot. The results demonstrated that kojic acid at a sub-inhibitory concentration (1/4 MI

A beneficial endornavirus enhances the fitness of the phytopathogenic fungus Rhizotonia solani.

Although viruses are primarily characterized as pathogenic agents, certain viruses confer advantages to their hosts. The extent to which a virus can improve host biological performance, however, remains a fascinating topic in virology. An endornavirus, Rhizoctonia solani endornavirus IM (RsEV-IM), was identified as prevalent in Rhizoctonia solani isolates obtained from potato plants. Comparative analysis with a virus-free isogenic strain demonstrated that RsEV-IM infection enhances mycelial growth, sclerotium formation, stress tolerance, and fungal virulence across multiple plant species. Impact of contact parameters on lesion dimensions during circular Pulsed-Field ablation in ex vivo and in vivo models.

Pulsed field ablation (PFA) is a novel nonthermal therapy for cardiac arrhythmias. The influence of contact force (CF), contact status, and application count on lesion characteristics in bipolar biphasic circular PFA remains uncertain. This study investigated lesion dimensions and acute histopathological changes using potato and porcine myocardial models. PFA was delivered in potato models under non-contact conditions or stratified contact forces (0–25 g, 25–50 g, > 50 g; 5 applications each). Porcine myocardium lesions were created with 1, 3, or 5 applications under contact or non-contact. Co
