Agents that are known to be scavengers of hydroxyl radicals inhibit lymphocyte mitogenesis induced by phorbol myristate acetate (PMA) to a greater extent than they inhibit mitogenesis induced by concanavalin A or phytohemagglutinin. These agents include dimethyl sulfoxide, benzoate, thiourea, dimethylurea, tetramethylurea, L-tryptophan, mannitol, and several other alcohols. Their inhibitory effect is not associated with cytotoxicity. A convenient preparation method of N-acylbenzoxazines has been developed, in which phenols react with nitriles and dimethyl sulfoxide (DMSO) in the presence of a catalytic amount of (COCl)2 in CH3CN or chloroform to afford the corresponding N-acylbenzoxazines in moderate-to-good yields. DMSO acts as a source of HCHO, which is generated in situ from the decomposition of a methoxydimethylsulfonium salt. A regeneration cycle of the methoxydimethylsulfonium salt is proposed, which is initiated by a catalytic amount of (COCl)2.

The mechanism proposed for the transformation of cyclopentanone to the dienoic acid 1, as published in this journal, is revealed to be in error. We show that carbon 11 derives not from dimethyl sulfoxide as proposed but from the dichloromethane present in the "quenching" solution. The intermediacy of an α-chloromethyl ketone and its subsequent fragmentation in the presence of a hydroxide ion is supported by experiments described herein and by extensive literature precedent.

Stem cell transplants are established therapy for hematologic and solid tumor malignancies. Known neurological complications of stem cell transplantation include CNS infection, seizures, strokes, metabolic encephalopathy, and hemorrhage. We report two cases of autologous stem cell transplantation complicated by cerebral infarction and myocardial injury. We postulate that the cryopreservative dimethyl sulfoxide may be responsible.

In this study, we synthesized γ-phenyl-β,γ-unsaturated ketones in situ from acetophenones and phenylacetylenes under Trofimov's conditions using KOtBu in a dimethyl sulfoxide (DMSO) solvent. The obtained ketones reacted with α,β-unsaturated carbonyls in a one-pot manner, forming tri- or diarylated benzophenones. The present reaction proceeded efficiently by one-pot manipulation with a suitable carboxylic acid.

An efficient method for the synthesis of β,β-di(hetero)aryl-α,α-difluorinated ketones using readily available organophosphonium salts and difluoroenol silyl ethers has been developed. This mild reaction features a good functional group tolerance, a scaled-up synthesis, and synthetic simplicity. By taking advantage of DMSO as a less-toxic promoter and solvent for the difluoroalkylation and C-P bond functionalization, the use of transition-metal catalysts and sensitive additives could be avoided.

To gain some insight into the role of c-myb and c-fes in myeloid differentiation, the authors have analyzed the ability of HL60 cells to differentiate in response to several different inducers after inhibition of c-myb and c-fes function. The effect of tiacrilast, a mast cell mediator-release inhibitor, was studied in dinitrofluorobenzene-induced allergic and croton oil- or dimethyl sulfoxide-induced irritant murine contact dermatitis. At 1% concentration, the compound significantly reduced the ear swelling in both allergic and irritant dermatitis and preserved the mast cell architecture on histopathology. These findings suggest that mast cells participate in the elicitation of murine contact dermatitis.

The study served to answer the question whether metabolites possibly contribute to the clinical actions of the neuroleptic drug perazine. The primary metabolites demethylperazine and perazine sulfoxide were investigated with regard to influences on behavior in mice, to an antiemetic action in dogs, and to a modification of the pressor effect of noradrenaline in rats. In contrast to perazine, none of the metabolites exhibited effects that can be interpreted to indicate neuroleptic or antidepressive properties of the compounds.
